Eutrema edwardsii R.Br. (Brassicaceae) is an arctic-alpine mustard with a circumpolar distribution. Its closest relative, Eutrema penlandii Rollins, is a federally listed, threatened species that is endemic to the Mosquito Range in the Southern Rocky Mountains of Colorado, USA. As part of a larger project addressing the systematics of this species complex in North America, we conducted chromosome counts, flow cytometry, and allozyme analysis to test the hypothesis that these taxa comprise an autopolyploid complex. Within that context, it should be noted that a chromosome count has not been reported previously for E. penlandii. Results obtained from mitotic counts obtained for two populations of E. penlandii reveal this taxon to be diploid. Diploidy was confirmed using flow cytometry for an additional 15 individuals representing four populations. Previously published chromosome counts for E. edwardsii reveal a polyploid complex of tetraploid, hexaploid, and octaploid populations for which an autopolyploid origin has been presumed. However, allozyme analysis revealed an allopolyploid origin for E. edwardsii, as evidenced from fixed heterozygosity at six loci. Although our data suggest that E. penlandii is a close relative of one of the progenitors of E. edwardsii, the taxonomic identity of the other progenitor(s) cannot be elucidated from these data. The data reported herein support the recognition of E. penlandii as taxonomically distinct, which has implications for conservation, and reveal cryptic variation within E. edwardsii.  相似文献

Riparian ecosystems are hotspots for ecological restoration globally because of the disproportionately high value and diversity of the ecological functions and services which they support and their high level of vulnerability to anthropogenic pressures, including climate change. Degraded riparian ecosystems are associated with many serious anthropogenic problems including increased river bank erosion, water quality decline, increased flood risk and biodiversity loss. Conventional approaches to riparian restoration, however, are frequently too narrow in focus – spatially, temporally, ecologically and socially – to adequately or equitably address the goals to which they aspire. Climate change, along with the intensification of other human pressures, means that static, historically oriented restoration objectives focused solely on prior ecological composition and structure are unlikely to be defensible, achievable or appropriate in the Anthropocene. Conversely, open‐ended restoration strategies lacking clear objectives and targets entail substantial risks such as significant biodiversity losses, especially of native species. A functional approach to planning and prioritising riparian restoration interventions offers an intermediate alternative that is still framed by measurable targets but allows for greater consideration of broader temporal, spatial and cultural influences. Here, we provide an overview of major riparian functions across multiple scales and identify key drivers of, and threats to, these. We also discuss practical approaches to restoring and promoting riparian functions and highlight some key concerns for the development of policy and management of robust riparian restoration in the Anthropocene.  相似文献

This paper reports a handheld multiphoton fluorescence microscope designed for clinical imaging that incorporates axial motion compensation and lateral image stabilization. Spectral domain optical coherence tomography is employed to track the axial position of the skin surface, and lateral motion compensation is realised by imaging the speckle pattern arising from the optical coherence tomography beam illuminating the sample. Our system is able to correct lateral sample velocities of up to approximately 65 μm s?1. Combined with the use of negative curvature microstructured optical fibre to deliver tunable ultrafast radiation to the handheld multiphoton scanner without the need of a dispersion compensation unit, this instrument has potential for a range of clinical applications. The system is used to compensate for both lateral and axial motion of the sample when imaging human skin in vivo.   相似文献

Hypopharyngeal cancer (HC) is the most common subset of head and neck cancers. These tumors often have an aggressive clinical outcome characterized by local invasion and regional nodal metastasis. Upregulated miRNAs might be useful as biomarkers for prognosis and molecular targets for these tumors. We determined tumor expression of candidate miRNAs using microarray in 8 HC patients and validated in 372 HC patients. We also used paired tumorous and mucosal tissue to verify the miRNA expression. Log-rank test and Cox model were used to evaluate the survival; and Harrell's C-index was used to compare concordance of Cox models. Our results indicated 7 miRNAs aberrantly expressed in HC. Three of these candidate miRNAs (miRNA-4415, miRNA-200a, and miRNA-30b) were selected for further qRT-PCR validation and all of them were frequently found upregulated in HC tumors; with miR-4451 being the most differentially expressed. Moreover, high expression of miR-4451 was positively correlated with advanced tumor stage and increased mortality risk (HR: 1.6, 95% CI: 1.2–2.3; adjusted HR: 1.5, adjusted 95% CI: 1.1–2.1). Finally, significantly higher expression of miR-4451 in tumors compared to in fresh adjacent normal tissues indicates an oncogenic role of miR-4451 in this tumor. Upregulated miR-4451 in HC samples were frequently found and is significantly associated with advanced stage and poor survival of HC, which may indicate an association of this miRNA with the carcinogenesis process in this tumor site; and they could serve as a prognostic biomarker as well as help develop potential new targets for therapy.  相似文献

RC–LH1–PufX complexes from a genetically modified strain of Rhodobacter sphaeroides that accumulates carotenoids with very long conjugation were studied by ultrafast transient absorption spectroscopy. The complexes predominantly bind the carotenoid diketospirilloxanthin, constituting about 75% of the total carotenoids, which has 13 conjugated C=C bonds, and the conjugation is further extended to two terminal keto groups. Excitation of diketospirilloxanthin in the RC–LH1–PufX complex demonstrates fully functional energy transfer from diketospirilloxanthin to BChl a in the LH1 antenna. As for other purple bacterial LH complexes having carotenoids with long conjugation, the main energy transfer route is via the S2–Qx pathway. However, in contrast to LH2 complexes binding diketospirilloxanthin, in RC–LH1–PufX we observe an additional, minor energy transfer pathway associated with the S1 state of diketospirilloxanthin. By comparing the spectral properties of the S1 state of diketospirilloxanthin in solution, in LH2, and in RC–LH1–PufX, we propose that the carotenoid-binding site in RC–LH1–PufX activates the ICT state of diketospirilloxanthin, resulting in the opening of a minor S1/ICT-mediated energy transfer channel.  相似文献

Spatially averaged models of root–soil interactions are often used to calculate plant water uptake. Using a combination of X‐ray computed tomography (CT) and image‐based modelling, we tested the accuracy of this spatial averaging by directly calculating plant water uptake for young wheat plants in two soil types. The root system was imaged using X‐ray CT at 2, 4, 6, 8 and 12 d after transplanting. The roots were segmented using semi‐automated root tracking for speed and reproducibility. The segmented geometries were converted to a mesh suitable for the numerical solution of Richards' equation. Richards' equation was parameterized using existing pore scale studies of soil hydraulic properties in the rhizosphere of wheat plants. Image‐based modelling allows the spatial distribution of water around the root to be visualized and the fluxes into the root to be calculated. By comparing the results obtained through image‐based modelling to spatially averaged models, the impact of root architecture and geometry in water uptake was quantified. We observed that the spatially averaged models performed well in comparison to the image‐based models with <2% difference in uptake. However, the spatial averaging loses important information regarding the spatial distribution of water near the root system.  相似文献

The GLIS family zinc finger 3 isoform (GLIS3) is a risk gene for Type 1 and Type 2 diabetes, glaucoma and Alzheimer's disease endophenotype. We identified GLIS3 binding sites in insulin secreting cells (INS1) (FDR q < 0.05; enrichment range 1.40–9.11 fold) sharing the motif wrGTTCCCArTAGs, which were enriched in genes involved in neuronal function and autophagy and in risk genes for metabolic and neuro-behavioural diseases. We confirmed experimentally Glis3-mediated regulation of the expression of genes involved in autophagy and neuron function in INS1 and neuronal PC12 cells. Naturally-occurring coding polymorphisms in Glis3 in the Goto-Kakizaki rat model of type 2 diabetes were associated with increased insulin production in vitro and in vivo, suggestive alteration of autophagy in PC12 and INS1 and abnormal neurogenesis in hippocampus neurons. Our results support biological pleiotropy of GLIS3 in pathologies affecting β-cells and neurons and underline the existence of trans?nosology pathways in diabetes and its co-morbidities.  相似文献
