Combination therapies aim to overcome the limitations of individual drugs by selecting diverse targets of action to enhance effectiveness synergistically. This article reviews the principles of combination therapy and its applications for benign prostatic hyperplasia and overactive bladder. A tannase (E.C. 3.1.1.20) producing fungal strain was isolated from soil and identified as Aspergillus heteromorphus MTCC 8818. Maximum tannase production was achieved on Czapek Dox minimal medium containing 1% tannic acid at a pH of 4.5
and 30°C after 48 h incubation. The crude enzyme was purified by ammonium sulfate precipitation and ion exchange chromatography.
Diethylaminoethyl-cellulose column chromatography led to an overall purification of 39.74-fold with a yield of 19.29%. Optimum
temperature and pH for tannase activity were 50°C and 5.5 respectively. Metal ions such as Ca2+, Fe2+, Cu1+, and Cu2+ increased tannase activity, whereas Hg2+, Na1+, K1+, Zn2+, Ag1+, Mg2+, and Cd2+ acted as enzyme inhibitors. Various organic solvents such as isopropanol, isoamyl alcohol, benzene, methanol, ethanol, toluene,
and glycerol also inhibited enzyme activity. Among the surfactants and chelators studied, Tween 20, Tween 80, Triton X-100,
EDTA, and 1, 10-o-phenanthrolein inhibited tannase activity, whereas sodium lauryl sulfate enhanced tannase activity at 1%

We describe here the development of a reproducible plastid transformation system for potato and regeneration of plants with uniformly transformed plastids. Two distinct tobacco-specific plastid vectors, pZS197 (Prrn/aadA/TpsbA) and pMON30125 (Prrn/GFP/Trps16::PpsbA/aadA/TpsbA), designed for integration into the large single copy and inverted repeat regions of the plastid genome, respectively, were bombarded into leaf explants of potato line FL1607. A total of three transgenic lines were selected out of 46 plates bombarded with pZS197 and three transgenic lines out of 104 plates were obtained with pMON30125. Development of a high frequency leaf-based regenera- tion system, a stringent selection scheme and optimization of biolistic transformation protocol were critical for recovery of plastid transformants. Plastid-expressed green fluorescent protein was used as a visual marker for identification of plastid transformants at the early stage of selection and shoot regeneration. The establishment of a plastid transformation system in potato, which has several advantages over routinely used nuclear transformation, offers new possibilities for genetic improvement of this crop. 相似文献

Lethaia, Vol. 45, pp. 356–370. The siliciclastic sequence of the Upper Devonian of Kurzeme, Western Latvia, is renowned for abundant vertebrate fossils, including the stem tetrapods Obruchevichthys gracilis and Ventastega curonica. During the first detailed taphonomic study of the vertebrate assemblage from the Ogre Formation cropping out at the Langsēde Cliff, Imula River, abundant vertebrate remains have been examined and identified as belonging to one psammosteid, two acanthodian and three sarcopterygian genera; the placoderm Bothriolepis maxima dominates the assemblage. Besides fully disarticulated placoderm and psammosteid plates, separate sarcopterygian scales and teeth, and acanthodian spines, partly articulated specimens including complete distal segments of Bothriolepis pectoral fins, Bothriolepis head shields and sarcopterygian lower jaws have been found. The size distribution of the placoderm bones demonstrates that the individuals within the assemblage are of approximately uniform age. Distinct zones have been traced within the horizontal distribution of the bones. These linear zones are almost perpendicular to the dominant dip azimuth of the cross‐beds and ripple‐laminae and most probably correspond to the depressions between subaqueous dunes. Concavity ratio varies significantly within the excavation area. The degree of fragmentation of the bones and disarticulation of the skeletons suggest that the carcasses were reworked and slightly transported before burial. Sedimentological data suggest deposition in a shallow marine environment under the influence of rapid currents. The fossiliferous bed consists of a basal bone conglomerate covered by a cross‐stratified sandstone with mud drapes, which is in turn overlain by ripple laminated sandstone, indicating the bones were buried by the gradual infilling of a tidal channel. All the Middle–Upper Devonian vertebrate bone‐beds from Latvia are associated with sandy to clayey deposits and have been formed in a sea‐coastal zone during rapid sedimentation episodes, but differ in fossil abundance and degree of preservation. □Agnathans, Devonian, facies analysis, fish, fossil assemblage, palaeoenvironment. 相似文献

Fourteen heat resistant mutant strains were isolated from a wild-type strain (PP201, Nod+ Fix+) of Rhizobium sp. (Cajanus) by giving it a heat shock of 43°C. These mutant strains showed a greater increase in optical density (O.D.) and a higher
viable cell count in both rhizospheric and non-rhizospheric soil at high temperature. Symbiotic studies showed that pigeon
pea plants inoculated with a few mutant strains had ineffective nodules (Nod+ Fix−) under controlled temperature (43°C) conditions, but under natural high temperature (40–45°C) conditions, the host plants
infected with all the mutant strains showed higher total shoot nitrogen than the plants inoculated with the parent strain.
Four mutant strains (HR-3, HR-6, HR-10 and HR-12) were found to be highly efficient for all the symbiotic parameters, and

The effect of plant growth regulator concentrations and ageing of callus on the extent and nature of variation among callus culture regenerants of strawberry (Fragaria × ananassa) cv. Redcoat was examined. Plants regenerated from callus culture had reduced plant vigour, shorter petiole length and smaller leaf size, but more leaves and runners under greenhouse conditions. These responses appeared to be due to a physiological influence of plant growth regulators. No distinct phenotypic variants were observed at plant growth regulator concentrations in the range of 1–10 M each of BA and 2,4-d combination, but the highest concentration (20 M each) of this combination produced a high frequency (10%) of dwarf type variants. The dwarf nature of these variants was maintained in the runner plants produced by the primary regenerants. The plants regenerated from 8-week-old calli did not show any distinct morphological variants. However, a significant proportion of deformed leaf shape (6–13%) and yellow leaf (21–29%) variants was obtained among plants regenerated from 16 and 24-week-old calli. The primary regenerants of the leaf shape variants were established as chimeras. The chimeric plants produced runner progeny with normal plants and plants with completely distorted leaf morphology. Both leaf shape and yellow leaf variants remained stable through runner propagation. Isozyme analysis failed to distinguish any of the variants from the standard runner plants. Flow cytometric analysis indicated the aneuploid nature of leaf shape variants but it could not distinguish dwarf and yellow leaf variants from standard runner plants. 相似文献
