At saturating concentrations, tropomyosin inhibited the rate of spontaneous polymerization of ATP-actin and also inhibited by 40% the rates of association and dissociation of actin monomers to and from filaments. However, tropomyosin had no effect on the critical concentrations of ATP-actin or ADP-actin. The tropomyosin-troponin complex, with or without Ca2+, had a similar effect as tropomyosin alone on the rate of polymerization of ATP-actin. Although tropomyosin binds to F-actin and not to G-actin, the absence of an effect on the actin critical concentration is probably explicable in terms of the highly cooperative nature of the binding of tropomyosin to F-actin and its very low affinity for a single F-actin subunit relative to the affinity of one actin subunit for another in F-actin.  相似文献

Soil erosion and the effects of soil erosion on crop productivity have become emotional issues and have attracted the attention of agriculturists, environmentalists, and the public in general. In spite of heavy investments in research and development, the global rates of accelerated erosion are now presumbly higher than ever before. However, the data from available records obtained by diverse methods are uncomparable, unreliable, confusing, and often vary by several orders of magnitude. Reports of erosion‐caused alterations in crop productivity and soil properties are also contradictory and subjective. In addition to the lack of standardized methodology in evaluating soil erosion and its effects on crops, controversial interpretations are attributed to differences in soil profile characteristics, nutrient status, crops grown, and prevailing climatic conditions. Although erosion is generally associated wtih yield reductions, there are examples of where soil erosion has had no effect or has had a positive effect on crop production. Accelerated erosion affects productivity both directly and indirectly. Directly, the erosion‐induced reduction in crop yields is attributed to loss of rooting depth, degradation of soil structure, decrease in plant‐available water reserves, reduction in organic matter, and nutrient imbalance. Depending on soil properties and the degree of degradation, adverse effects of erosion on crop yields can be mostly compensated for by additional inputs of macronu‐trients (N, P, K) and macronutrients plus organic matter, by supplemental applications of some micronu‐trients, and by irrigation. For some soils, e.g., tropical soils, crop yields from severely eroded soils are significantly lower than those from uneroded lands and are often uneconomic in spite of additional inputs. Specific examples of yield alterations are given in relation to the loss of plant nutrients, soil water reserves, and alterations in soil properties. Criteria for soil‐loss tolerance are discussed, and productivity restoration of eroded soils is reviewed in relation to soil organic matter content and nutrient requirments. Research and development priorities are presented.  相似文献

A procedure has been developed for the clonal propagation of Picrorhiza kurroa Royle ex Benth. through shoot tip culture. Murashige and Skoog's medium (1962) supplemented with kinetin (3.0 to 5.0 mg/l) supported rapid proliferation of multiple shoots from the explants. Addition of indole-3-acetic acid (1.0 mg/l) to the kinetin containing medium showed marked improvement in the growth of regenerated shoots. However, presence of IAA in the medium did not alter the frequency of shoot multiplication. Rooting was readily achieved upon transferring shoots onto MS medium containing -naphthaleneacetic acid (1.0 mg/l). Shoot-tip explants of Rheum emodi Wall. (Polygonaceae) gave rise to multiple shoots when cultured on a Murashige and Skoog (1962) medium (MS) with 2.0 mg/l 6-benzylaminopurine (BAP) and 1.0 mg/l indole-3-butyric acid (IBA). Also, shoot buds developed from leaf explants using MS medium with 2.0 mg/l BAP and 0.25 to 1.0 mg/l indole-3-acetic acid (IAA) or IBA. Roots were induced when the resulting shoots were placed on MS medium with 1.0 mg/l IBA. Both regeneration procedures gave rise to healthy plantlets that were established in soil under glasshouse conditions at 80% frequency after hardening phase of two weeks. Regenerated plants showed a constant chromosome number of 2n=2x=22, same as the parent plant. The use of liquid shake cultures minimized the time and culture medium requirements for propagation. The biotransformation of estradiol (E2) and estrone (E1) in the uterus of rabbits treated with norgestrel (NG), norethindrone (NET), norethindrone acetate (NETA), progesterone (P4), and E2 either by subcutaneous injection in oil or by intrauterine steroid-releasing silastic implants was carried out under an in vitro short-term incubation system. The studies have shown that E2 stimulates 17 beta-hydroxysteroid dehydrogenase (17 beta-OHSD) much more than P4 as compared to untreated controls. The kinetic studies on E2 metabolism in the presence of added coenzyme NAD showed an initial rapid estrone formation and a gradual reconversion of E1 to E2. The addition of NADPH, ATP, and glucose-6-phosphate facilitates the reconversion of E1 to E2. The interconversion of E2 and estrone in the presence of coenzymes was five- to ten-fold higher in the endometrium than in the myometrium per milligram protein. Both E2 and progestins stimulate the uterine 17 beta-OHSD activity in rabbit uterus. This study further suggested that the hormone-induced metabolism of estradiol and estrone in the rabbit uterus is essentially modulated by the availability of coenzymes.  相似文献

Splenic lymphocyte proteins from New Zealand Black (NZB) mice, which spontaneously develop autoimmune disease, and several control strains were analyzed by two-dimensional polyacrylamide gel electrophoresis. A number of strain- and age-related differences were observed, among which was the persistently elevated synthesis of two peptides, 12.5 KD and 10.5 KD, by spleen cells from older NZB mice. Although synthesis of these peptides was moderately high in young NZB and control mice, it diminished with age in control mice. These proteins were found in the cytoplasm and were not expressed on the plasma membrane nor secreted into the medium. Production of these proteins was restricted to B and null cells; T cells did not synthesize these peptides. These proteins appear to be indicators of disease activity, because their increased synthesis was associated with lymphocyte subset alterations associated with the onset of overt autoimmune disease in NZB mice.  相似文献

The present study describes the comparative analysis of five genetic markers viz., random amplified polymorphic DNA (RAPD), enterobacterial repetitive intergenic consensus (ERIC), BOX-elements, mating type (MAT) locus and microsatellites for genetic analysis of virulent isolates of Fusarium oxysporum f. sp. ciceri (FOC) representing seven races from chickpea. Phylogenetic analysis of translation elongation factor 1-α and internal transcribed spacer region separated all the FOC isolates into two major clades. Majority of the isolates (FOC 63, FOC 33, FOC 40, FOC 100, FOC 6, FOC 22, FOC 31, FOC 79 and NDFOC 98) representing race 1, 2, 5 and 6 grouped in clade I, while isolates (FOC 90, FOC 108 and FOC 88) belonging to race 3, 4 and 7 were clustered in clade II. Isolates (FOC 33, FOC 40, FOC 17 and FOC 100) representing race 2 had MAT-2 loci, while race 1 isolates (FOC 63, FOC 72 and FOC 76) contained MAT-1 loci only. The principal component analysis (PCA) of RAPD, ERIC, BOX and microsatellite marker data explained 39.94, 39.98, 42.04 and 62.59% of the total variation among test isolates, respectively. Furthermore, there was no correlation existed between genetic diversity, virulence, race compositions or geographic origin of the isolates. Overall, these findings will assist in better understanding of the genetic variability and ideally, will improve disease management practices.  相似文献

Congress grass, Parthenium hysterophorus L. (Asteraceae, tribe Heliantheae), is an erect and much branched annual or ephemeral herb, known as a pest for environmental, medical and agricultural reasons. Survey of fungal pathogens of P. hysterophorus was conducted in Kurukshetra and its adjoining areas, Haryana, India. A leaf spot pathogen Alternaria sp. PMK1 was isolated. Koch's postulates were performed and proved that this isolate was pathogenic to this weed. This is the first report of this pathogen causing leaf spot on parthenium weed.  相似文献

No‐till (NT) practices are among promising options toward adaptation and mitigation of climate change. However, the mitigation effectiveness of NT depends not only on its carbon sequestration potential but also on soil‐derived CH4 and N2O emissions. A meta‐analysis was conducted, using a dataset involving 136 comparisons from 39 studies in China, to identify site‐specific factors which influence CH4 emission, CH4 uptake, and N2O emission under NT. Comparative treatments involved NT without residue retention (NT0), NT with residue retention (NTR), compared to plow tillage (PT) with residue removed (PT0). Overall, NT0 significantly decreased CH4 emission by ~30% (< 0.05) compared to PT0 with an average emission 218.8 kg ha−1 for rice paddies. However, the increase in N2O emission could partly offset the benefits of the decrease in CH4 emission under NT compared to PT0. NTR significantly enhanced N2O emission by 82.1%, 25.5%, and 20.8% (< 0.05) compared to PT0 for rice paddies, acid soils, and the first 5 years of the experiments, respectively. The results from categorical meta‐analysis indicated that the higher N2O emission could be mitigated by adopting NT within alkaline soils, for long‐term duration, and with less N fertilization input when compared to PT0. In addition, the natural log (lnR) of response ratio of CH4 and N2O emissions under NT correlated positively (enhancing emission) with climate factors (temperature and precipitation) and negatively (reducing emission) with experimental duration, suggesting that avoiding excess soil wetness and using NT for a long term could enhance the benefits of NT. Therefore, a thorough understanding of the conditions favoring greenhouse gas(es) reductions is essential to achieving climate change mitigation and advancing food security in China.  相似文献
