Studies of the impact of inorganic phosphorus (Pi), an important nutrient, on the growth and physiological parameters of single-celled algae are important for investigations of the dynamics of phytoplankton abundance and productivity in natural ecosystems as well as in industrial systems for the cultivation of microalgae. Difficulties in carrying out such studies are associated with the complex kinetics of Pi uptake by cells and the ability of microalgae to store phosphorus in their cells. This situation necessitates efficient methods for express monitoring of microalgal cultures, such as the methods based on the registration of optical properties of cells, in particular absorption and scattering of light and fluorescence of chlorophyll contained in the cells. Here, the results of monitoring the cultures of the chlorophyte Chlorella vulgaris IPPAS C-1 starving for phosphorus are described. It was found that both optical (light absorption in the bands of the key pigments—chlorophylls and carotenoids) and luminescent (variable fluorescence of chlorophyll) parameters closely reflect the culture condition. Registration of optical properties required correction for the contribution of light scattering to the overall extinction of light by microalgal cell suspensions. At the same time, the light scattering signal is an accurate measure of the total number of suspended particles in the suspension. However, it is difficult to monitor cultures containing a significant amount of light-scattering particles lacking photosynthetic pigments (such as heterotrophic bacteria). For such cultures, the use of variable fluorescence- based parameter Fv/Fm reflecting the maximum photochemical efficiency of the photosystem II is advisable.  相似文献

Hammerhead ribozymes with long antisense flanks (>50 bases) have been used successfully to inhibit replication of human immunodeficiency virus type 1 (HIV-1) in living cells. To explain their increased efficacy versus antisense controls or catalytically inactive derivatives, one can consider dissociation of the ribozyme-product complex to allow a complete catalytic cycle. In this work we investigated the dissociation of a double-stranded RNA with 56 bp in vitro. Dissociation was observed in the presence of single-stranded RNA with sequence complementarity to one of the duplex strands. A displacement reaction between RNA single strands and the duplex, but not simple dissociation, was strongly suggested by the concentration dependence of this process, the influence of additional non-complementary sequences on the single strand and by the unusually low Arrhenius activation energy. The strand displacement reaction was slow in vitro at 37 degrees C and physiological ionic strength, but was increased to k approximately 10(3)-10(4)/M/s (approximately 10(4)-fold) at higher temperatures by cetyltrimethylammonium bromide. This compound is thought to enhance non-sequence-specific association of nucleic acids in a mechanistically similar way to that in which cellular hnRNP proteins are thought to act, indicating that strand displacement can be fast and, more importantly, could be tightly regulated in vivo.  相似文献

Oxygen evolution from aScenedesmus obliquus dominated outdoor culture was followed in a small volume chamber, irradiated either by continuous white light or under light/dark frequencies between 0.05 to 5000 Hz, using arrays of high intensity red light emitting diodes (LED's). By placing neutral density filters in the path of the white light, light saturation curves of the oxygen evolution (P/I curves) were measured using diluted aliquots of algal cultures. The results clearly showed that photosynthetic rates increased exponentially with increasing light/dark frequencies, that a longer dark period in relation to the light period does not necessarily lead to higher photosynthetic rates (efficiencies), and that algae do not acclimate to a specific light/dark frequency. One of the most important factors that influenced photosynthetic rates, either under continuous illumination or intermittent, was whether the algae were dark or light acclimated. Low light/dark frequencies were perceived by the algae as low light conditions, whilst the opposite was true for high frequencies. The light utilisation efficiency in a fluctuating light/dark environment depended on the acclimated state of the algae, the specific frequency of the fluctuations and the duration of the exposure. Since the frequencies determined the perceived quantities of light, dark reactions played an important role in determining the average photosynthetic efficiencies. These results have important implications for algal biotechnology.  相似文献

The effects of fluctuations in the irradiance onScenedesmus quadricauda, Chlorella vulgaris andSynechococcus elongatus were studied in dilute cultures using arrays of red light emitting diodes. The growth rate and the rate of photoinhibition were compared using intermittent and equivalent continuous light regimes in small-size (30 ml) bioreactors. The CO2 dependent photosynthetic oxygen evolution rates in the intermittent and continuous light regimes were compared for different light/dark ratios and different mean irradiances. The kinetics of the electron transfer reactions were investigated using a double-modulation fluorometer. The rates of photosynthetic oxygen evolution normalized to equal mean irradiance were lower or equal in the intermittent light compared to the maximum rate found in the equivalent optimal continuous light regime. In contrast, the growth rates in the intermittent light can be higher than the growth rate in the equivalent continuous light. Photoinhibition is presented as an example of a physiological process affecting the growth rate that occurs at different rates in the intermittent and equivalent continuous lights. The difference in the dynamics of the redox state of the plastoquinone pool is proposed to be responsible for the low photoinhibition rates observed in the intermittent light.  相似文献

In outdoor thin-layer sloping reactors algae are batch cultured and harvested at biomass concentrations of about 15 g (dw) I-1 whereafter a portion is used as inoculum for the next cycle. Light saturation curves of the oxygen evolution (PII curves) of the algae were measured using diluted aliquots of suspension taken from the reactors. The maximum specific photosynthetic rates (P B max) and the light intensity at the onset of saturated photosynthesis (I k ) decreased whilst the maximum specific photosynthetic efficiency ( B ) increased with an increase in the biomass concentration, during the production cycle. These differences reflect transition from light- to dark-acclimated state of the algae that occurs as a result of an increase of the suspension concentration during the production cycle. During these experiments the thin-layered smooth sloping cultures (TLSS, culture depth 5–7 mm) had higher photosynthetic rates per volume than the thin-layered baffled sloping cultures (TLBS, culture depth 5–15 mm). This was ascribed to the higherP B max values of the algae grown in the TLSS cultures, allowing them to utilise high incident irradiancies more effectively. However, the areal productivity of the TLBS was higher than the TLSS indicating a higher photosynthetic efficiency of the TLBS reactors. The specific productivity decreased rapidly with an increase in the biomass concentration, but the yield remained linear during the batch production cycle, even at high areal densities.  相似文献

A 900- to 1100-bp fragment encompassing intron 1 of the nuclear transthyretin (prealbumin) gene was examined in 12 taxa of Old World hystricognath rodents of the families Bathyergidae, Petromuridae, Thryonomyidae, and Hystricidae. Within the Bathyergidae, Heterocephalus glaber (naked mole-rat) was basal, and the other East African species, Heliophobius argenteocinereus (silvery mole-rat), was sister to a southern African clade containing Bathyergus, Cryptomys, and Georychus (dune, common, and cape mole-rats). These results are congruent with studies using mitochondrial 12S rRNA gene sequences. A combined analysis of transthyretin and 12S rRNA data resulted in a well-supported topology with better resolution than either gene analyzed separately. These data support the findings by M. W. Allard and R. L. Honeycutt (1992, Mol. Biol. Evol. 9: 27-40) and R. L. Honeycutt (1992, Am. Sci. 80: 43-53) that complex social systems evolved independently at least twice, in the common and naked mole-rats.  相似文献

The evolutionary history of the red panda (Ailurus fulgens) plays a pivotal role in the higher-level phylogeny of the "bear-like" arctoid carnivoran mammals. Characters from morphology and molecules have provided inconsistent evidence for placement of the red panda. Whereas it certainly is an arctoid, there has been major controversy about whether it should be placed with the bears (ursids), ursids plus pinnipeds (seals, sea lions, walrus), raccoons (procyonids), musteloids (raccoons plus weasels, skunks, otters, and badgers [mustelids]), or as a monotypic lineage of uncertain phylogenetic affinities. Nucleotide sequence data from three mitochondrial genes and one nuclear intron were analyzed, with more complete taxonomic sampling of relevant taxa (arctoids) than previously available in analyses of primary molecular data, to clarify the phylogenetic relationships of the red panda to other arctoid carnivorans. This study provides detailed phylogenetic analyses (both parsimony and maximum-likelihood) of primary character data for arctoid carnivorans, including bootstrap and decay indices for all arctoid nodes, and three statistical tests of alternative phylogenetic hypotheses for the placement of the red panda. Combined phylogenetic analyses reject the hypotheses that the red panda is most closely related to the bears (ursids) or to the raccoons (procyonids). Rather, evidence from nucleotide sequences strongly support placement of the red panda within a broad Musteloidea (sensu lato) clade, including three major lineages (the red panda, the skunks [mephitids], and a clearly monophyletic clade of procyonids plus mustelids [sensu stricto, excluding skunks]). Within the Musteloidea, interrelationships of the three major lineages are unclear and probably are best considered an unresolved trichotomy. These data provide compelling evidence for the relationships of the red panda and demonstrate that small taxonomic sample sizes can result in misleading or possibly erroneous (based on prior modeling, as well as conflict between the results of our analyses of less and more complete data sets) conclusions about phylogenetic relationships and taxonomy.  相似文献

The sensitivity of marine algal biotest ISO 10253 to the photosystem 2 (PS2) herbicide diuron (DCMU) was determined. Using the diatom Phaeodactylum tricornutum, we found that the algal growth rate was reduced to 50 % of the control value (EC50) for ca. 200 nM DCMU. This value is too high to allow a practical application of the biotest for concentrations of the PS2 herbicides found in natural waters. The mechanisms causing the low sensitivity of the biotest to the PS2 herbicide were investigated by measuring parameters of photosynthetic apparatus in the diatom prior and during the biotest. The apparent dissociation constant for DCMU in P. tricornutum found by measurements of inhibition of oxygen evolution and of variable fluorescence was in the range 60–90 nM. This should lead to a much higher sensitivity of the biotest than found in our experiments. The low biotest sensitivity is caused by an acclimation to sub-lethal DCMU concentrations. The acclimation is manifested by the chlorophyll content per cell that is increasing with the DCMU concentration. During a prolonged exposure to sub-lethal herbicide concentrations, we observed also a selection of DCMU resistant organisms indicating that also an adaptation may decrease the test sensitivity. The biotest sensitivity may increase when the acclimation and adaptation are limited by shortening of the experiment duration. Plants are exposed to a temporally and spatially heterogeneous environment, and photosynthesis is well adapted to these fluctuations. Understanding of the complex, non-linear dynamics of photosynthesis in fluctuating light requires novel-modeling approaches that involve not only the primary light and dark biochemical reactions, but also networks of regulatory interactions. This requirement exceeds the capacity of the existing molecular models that are typically reduced to describe a partial process, dynamics of a specific complex or its particular dynamic feature. We propose a concept of comprehensive model that would represent an internally consistent, integral framework combining information on the reduced models that led to its construction. This review explores approaches and tools that exist in engineering, mathematics, and in other domains of biology that can be used to develop a comprehensive model of photosynthesis. Equally important, we investigated techniques by which one can rigorously reduce such a comprehensive model to models of low dimensionality, which preserve dynamic features of interest and, thus, contribute to a better understanding of photosynthesis under natural and thus fluctuating conditions. A modification of the double-modulation fluorometer is described that allows measuring very dilute phytoplankton samples. The high sensitivity is achieved by increasing the sample volume and by collecting the fluorescence from the large volume by an integrating sphere. The sensitivity of the instrument increased approximately proportionally to the volume of the sample. A further improvement of the sensitivity was achieved by replacing the PIN photodiode of the earlier versions by a photomultiplier. The instrument was used to measure fluorescence induction, F0 and Fm parameters, and QA -reoxidation kinetics at concentrations at and below 100 pM chlorophyll.
