? Premise of the study: In a large reciprocal transplant experiment, Eriophorum vaginatum tussocks transplanted along a latitudinal gradient in Alaska's interior exhibited genetic differentiation and phenotypic plasticity for vegetative traits. Using the same tussocks 30 yr later, we used estimates of growing season temperature at each site to ask whether there was a climatic cline for stomatal density, size, and conductance. ? Methods: We created impressions of the abaxial leaf surfaces of the transplanted individuals for viewing under a microscope and measured stomatal density (SD) and length (SL) for 224 individuals. We used SD and SL to estimate stomatal conductance (C). Separate one-way analyses of variance were performed to quantify the effect of population genetic differences and latitudinal environmental variation on stomatal characteristics. ? Key results: Our data suggest that stomatal size was influenced by both genetics and environment and that plasticity for stomatal density produced highest densities at the coolest sites. Stomatal conductance increased with decreasing temperature of site from which the populations originated. ? Conclusions: Our results demonstrate a cline in stomatal conductance in E. vaginatum, with some ability of populations to plastically produce an appropriate phenotypic response in a new environment. Because the species is a dominant species in many arctic plant communities, its ability to produce an appropriate stomatal phenotype and to optimize water use efficiency by decreasing stomatal conductance in warmer environments could affect both community composition and total primary productivity in future, warmer climates.  相似文献

The intravenous (IV) administration of synthetic ovine corticotropin-releasing factor (CRF) (10 and 125 μg/kg) to chair restrained rhesus monkeys stimulated the pituitary-adrenal axis. At these doses, increases in plasma concentrations of adrenocorticotropic hormone (ACTH) and cortisol were associated with blood pressure decreases and behavioral effects. These data demonstrate that synthetic ovine CRF (10 and 125 μg/kg) administered IV to the rhesus monkey results in associated endocrine, physiological, and behavioral changes.  相似文献

I analysed geographical variation in size, colour and pattern quantitatively in the Rufous-crowned Tody-Tyrant Poecilotriccus ruficeps and Lulu Von Hagen's Tody-Tyrant P. luluae , two component species of a superspecies inhabiting the northern Andes of South America. The conspicuously bridled P. r. melanomystax , here reported for the first time from the Andes of Venezuela and the Eastern Andes of Colombia, leapfrogs southward over populations of the plain-headed P. r. ruficeps in the Eastern Andes of Colombia, re-occurs in the Central Andes of Colombia, leapfrogs over populations of P. r. ruficeps in south-western Colombia and Ecuador, then 're-appears' as the similarly bridled P. r. peruvianus in southern Ecuador and northern Peru. In a second example of leapfrogging, the recently described P. luluae from the Eastern Andes of northern Peru is separated from superficially similar populations of P. r. ruficeps by the distribution of P. r. peruvianus. Because of their striking plumage differences and lack of interbreeding, the two bridled forms , P. r. melanomystax and P. r. peruvianus , may together constitute a species separable from the plain-headed forms, P. r. ruficeps and P. r. rufigenis . Vocally, however, all forms of P. ruficeps are similar. I recognize Poecilotriccus r. rufigenis provisionally pending the accumulation of new material. Geographical variation in size among populations of P. ruficeps is weakly clinal or chaotic. Crown and back colour are geographically uniform, however, and are identical among all populations of both P. ruficeps and P. luluae . In colour of belly and face, P. luluae is exceptionally stable and unique. This contrasts with striking geographical variation in facial colour and pattern in P. ruficeps . Although differences in song syllables are apparent, samples of tape recordings are too small to justify conclusions about the potential role of voice in reproductive isolation.  相似文献

Although many proteins are known to localize in bacterial cells, for the most part our understanding of how such localization takes place is limited. Recent evidence that the phospholipid cardiolipin localizes to the poles of rod-shaped bacteria suggests that targeting of some proteins may rely on the heterogeneous distribution of membrane lipids. Membrane curvature has been proposed as a factor in the polar localization of high-intrinsic-curvature lipids, but the small size of lipids compared to the dimensions of the cell means that single molecules cannot stably localize. At the other extreme, phase separation of the membrane energetically favors a single domain of such lipids at one pole. We have proposed a physical mechanism in which osmotic pinning of the membrane to the cell wall naturally produces microphase separation, i.e., lipid domains of finite size, whose aggregate sensitivity to cell curvature can support spontaneous and stable localization to both poles. Here, we demonstrate that variations in the strength of pinning of the membrane to the cell wall can also act as a strong localization mechanism, in agreement with observations of cardiolipin relocalization from the poles to the septum during sporulation in the bacterium Bacillus subtilis. In addition, we rigorously determine the relationship between localization and the domain-size distribution including the effects of entropy, and quantify the strength of domain-domain interactions. Our model predicts a critical concentration of cardiolipin below which domains will not form and hence polar localization will not take place. This observation is consistent with recent experiments showing that in Escherichia coli cells with reduced cardiolipin concentrations, cardiolipin and the osmoregulatory protein ProP fail to localize to the poles.  相似文献

Visual rod arrestin has the ability to self-associate at physiological concentrations. We previously demonstrated that only monomeric arrestin can bind the receptor and that the arrestin tetramer in solution differs from that in the crystal. We employed the Rosetta docking software to generate molecular models of the physiologically relevant solution tetramer based on the monomeric arrestin crystal structure. The resulting models were filtered using the Rosetta energy function, experimental intersubunit distances measured with DEER spectroscopy, and intersubunit contact sites identified by mutagenesis and site-directed spin labeling. This resulted in a unique model for subsequent evaluation. The validity of the model is strongly supported by model-directed crosslinking and targeted mutagenesis that yields arrestin variants deficient in self-association. The structure of the solution tetramer explains its inability to bind rhodopsin and paves the way for experimental studies of the physiological role of rod arrestin self-association.  相似文献

Quorum sensing, a process of bacterial cell-cell communication, relies on production, detection, and response to autoinducer signaling molecules. LuxN, a nine-transmembrane domain protein from Vibrio harveyi, is the founding example of membrane-bound receptors for acyl-homoserine lactone (AHL) autoinducers. We used mutagenesis and suppressor analyses to identify the AHL-binding domain of LuxN and discovered LuxN mutants that confer both decreased and increased AHL sensitivity. Our analysis of dose-response curves of multiple LuxN mutants pins these inverse phenotypes on quantifiable opposing shifts in the free-energy bias of LuxN for occupying its kinase and phosphatase states. To understand receptor activation and to characterize the pathway signaling parameters, we exploited a strong LuxN antagonist, one of fifteen small-molecule antagonists we identified. We find that quorum-sensing-mediated communication can be manipulated positively and negatively to control bacterial behavior and, more broadly, that signaling parameters can be deduced from in vivo data.  相似文献

Studies on the longevity and migration patterns of wild animals rely heavily on the ability to track individual adults. Non-extractive sampling methods are particularly important when monitoring animals that are commercially important to ecotourism, and/or are rare. The use of unique body patterns to recognize and track individual vertebrates is well-established, but not common in ecological studies of invertebrates. Here we provide a method for identifying individual Wunderpus photogenicus using unique body color patterns. This charismatic tropical octopus is commercially important to the underwater photography, dive tourism, and home aquarium trades, but is yet to be monitored in the wild. Among the adults examined closely, the configurations of fixed white markings on the dorsal mantle were found to be unique. In two animals kept in aquaria, these fixed markings were found not to change over time. We believe another individual was photographed twice in the wild, two months apart. When presented with multiple images of W. photogenicus, volunteer observers reliably matched photographs of the same individuals. Given the popularity of W. photogenicus among underwater photographers, and the ease with which volunteers can correctly identify individuals, photo-identification appears to be a practical means to monitor individuals in the wild.  相似文献

Flagellated bacteria, such as Escherichia coli, perform directed motion in gradients of concentration of attractants and repellents in a process called chemotaxis. The E. coli chemotaxis signaling pathway is a model for signal transduction, but it has unique features. We demonstrate that the need for fast signaling necessitates high abundances of the proteins involved in this pathway. We show that further constraints on the abundances of chemotaxis proteins arise from the requirements of self-assembly both of flagellar motors and of chemoreceptor arrays. All these constraints are specific to chemotaxis, and published data confirm that chemotaxis proteins tend to be more highly expressed than their homologs in other pathways. Employing a chemotaxis pathway model, we show that the gain of the pathway at the level of the response regulator CheY increases with overall chemotaxis protein abundances. This may explain why, at least in one E. coli strain, the abundance of all chemotaxis proteins is higher in media with lower nutrient content. We also demonstrate that the E. coli chemotaxis pathway is particularly robust to abundance variations of the motor protein FliM.  相似文献
