The aminoacyl-tRNA synthetases covalently link transfer RNAs to their cognate amino acids. Some of the tRNA synthetases have evolved editing mechanisms to ensure fidelity in this first step of protein synthesis. The amino acid editing site for leucyl- (LeuRS) and isoleucyl- (IleRS) tRNA synthetases reside within homologous CP1 domains. In each case, a threonine-rich peptide and a second conserved GTG region that are separated by about 100 amino acids comprise parts of the hydrolytic editing site. While a number of sites are conserved between these two enzymes and likely confer a commonality to the mechanisms, some positions are idiosyncratic to LeuRS or IleRS. Herein, we provide evidence that a conserved arginine and threonine at respective sites in LeuRS and IleRS diverged to confer amino acid substrate recognition. This site complements other sites in the amino acid binding pocket of the editing active site of Escherichia coli LeuRS, including Thr252 and Val338, which collectively fine-tune amino acid specificity to confer fidelity.  相似文献

American bollworm (ABW), Helicoverpa armigera (Hubner), is considered as a major pest of cotton, Gossypium hirsutum, all over the globe. Due to its destructive feeding nature and continuous consumption of the same chemicals, it devolved resistant against many insecticides. Therefore, a combined application of bio- and synthetic-pesticide need to evaluate against this pest. The entomopathogenic viruses like nuclear polyhedrosis virus (NPV), a member of baculoviruses, can be the potential candidates for better control against ABW. The present study was conducted to assess the comparative efficacy of NPV and Spinosad 240SC (with the concentration of 250 mL · ha ̄?) against ABW in the controlled environment. The ABW was treated with different concentrations of NPV and Spinosad separately and in a combination of NPV with 0.1% Spinosad. The results revealed that highest concentrations showed highest mortality (95%) followed by 95%, 92%, 84%, 82% and 78% mortality at 1 × 109, 1 × 108, 1 × 107, 1 × 106 and 1 × 105 POBs, respectively. Spinosad when mixed in diet give 100% mortality at 0.8% followed by 50.87%, 42.10%, 29.82%, 26.31% and 22.80% mortality at 0.4%, 0.2%, 0.1%, 0.5% and 0.025% respectively. The results of this study revealed that microbial control of ABW through NPV is an effective tool. The repeated use of synthetic pesticides caused the resurgence of many insect pests, and this study results would provide useful insight to build a framework for future investigations for the management of many major insect pests.  相似文献

Verticillium dahliae toxin (Vd toxin) was employed as pathogen free model system to induce osmotic stress on upland cotton and its amelioration is investigated using epibrassinolide (EBR). In this study, we observed the physiological and biochemical differences among Vd toxin alone and EBR + Vd toxin treated plants using different levels of root (5, 10, 15 nM) and shoot (50, 100, 200 nM) applied EBR. Results revealed that in absence of EBR, Vd toxin caused 83 % plant wilting and the levels of glycine betaine and proline were 33 and 61 % higher than non treated control, respectively. However, the accumulation of these osmolytes was decreased in EBR treated plants with minimum values at 5 and 200 nM. Furthermore, the results depicted a remarkable decline in soluble sugars, photosynthesis, transpiration, chlorophyll content and chlorophyll florescence (Fv/Fm) in Vd toxin alone treated plants than EBR + Vd toxin. Besides, the activities of sucrose synthase, sucrose phosphate synthase and acid invertase were high in Vd + EBR treated plants with increased root and shoot biomass than Vd toxin alone treated plants. Moreover, EBR remarkably regulated the levels of plasma membrane ATPases and contents of total ATPase and Na+ K+-ATPase were elevated while contents of Ca2+ Mg2+-ATPase and H+ K+-ATPase were decreased as compared to Vd toxin alone treated plants. This study broadens our understanding of Verticillium wilt and demonstrates the potential role of EBR in mediating tolerance against Vd toxin induced stress of V. dahliae in cotton.  相似文献

A newly isolated fungus Penicillium verruculosum SG was evaluated for the production and characterization of bioactive colored secondary metabolites using solid-state fermentation along with their cytotoxic activities against normal and cancer cell lines. Logical fragmentation pattern following column chromatography, thin layer chromatography and liquid chromatography and mass spectrometry of crude culture filtrate of fungus revealed the presence of different polyketide pigments and other bioactive compounds. Cytotoxicity of the selected colored fractions of fungal filtrate containing different compounds revealed IC50 (μg/ml) values ranging from 5 to 100. It was significantly higher in case of orevactaene (5 + 0.44) and monascorubrine followed by pyripyropene (8 + 0.63) against cancer cell line KA3IT. Overall, these compounds considerably showed less toxicity toward normal cell lines NIH3T3, HSCT6, HEK293 and MDCK. XRD of a yellow crystalline compound (224.21 m/z) confirmed its 3-dimensional structure as phenazine 1 carboxylic acid (C13H8N2O2) (broad spectrum antibiotic), and it is first time reported in fungi.  相似文献

The two-line hybrid system in rice is becoming more important and employs environment-conditioned genic male sterile (EGMS) lines sensitive to photoperiod (photoperiod-sensitive genic male sterile), temperature [temperature genic male sterile (TGMS)], or a combination of the two (photoperiod temperature genic male sterile). At least 18 EGMS genes have been mapped, and two cloned, but controversies exist. For example, three different genes were reported to underlie the TGMS trait in three independently identified progenitors, Annong S-1, Zhu 1S, and Guangzhan 63S, while another study demonstrated that the TGMS genes in Annong S-1 and Zhu 1S are allelic. In the present study, we confirmed the allelism of the three TGMS genes, which means there is a common TGMS gene(s) in these lines. Knowing there is an association between the mutant allele (RNZ m ) of a ribonuclease gene (RNZ) with the TGMS trait in Guangzhuan 63S, we then sequenced RNZ for 14 commercial EGMS and 21 non-EGMS lines, and we developed two derived cleaved amplified polymorphic sequence (dCAPS) markers to detect RNZ m alleles in 32 EGMS and 310 non-EGMS lines. The analyses showed that the RNZ m allele existed exclusively in EGMS lines; all non-EGMS lines contained the functional RNZ gc or RNZ tc allele. Furthermore, two segregating populations that included 2,429 individuals were developed by crossing Zhu 1S (RNZ m ) to two non-EGMS lines (both with RNZ tc ); examination of the segregation of male sterile and fertile plants indicated that the TGMS trait was under the control of a single gene; analysis of the markers revealed the RNZ m allele exclusively in TGMS plants and the RNZ tc allele only in non-TGMS plants in both populations. The dCAPS markers could therefore help select TGMS progeny in breeding programs, which will save time and labor, and improve breeding efficiency and accuracy.  相似文献

Accurate allele frequencies are important for measuring subclonal heterogeneity and clonal evolution. Deep-targeted sequencing data can contain PCR duplicates, inflating perceived read depth. Here we adapted the Illumina TruSeq Custom Amplicon kit to include single molecule tagging (SMT) and show that SMT-identified duplicates arise from PCR. We demonstrate that retention of PCR duplicate reads can imply clonal evolution when none exists, while their removal effectively controls the false positive rate. Additionally, PCR duplicates alter estimates of subclonal heterogeneity in tumor samples. Our method simplifies PCR duplicate identification and emphasizes their removal in studies of tumor heterogeneity and clonal evolution.

Introduction

Remission is an important goal of therapy in rheumatoid arthritis (RA), but data on duration of remission are lacking. Our objective was to describe the duration of remission in RA, assessed by different criteria.

Methods

We evaluated patients from the Brigham and Women''s Rheumatoid Arthritis Sequential Study (BRASS) not in remission at baseline with at least 2 years of follow-up. Remission was assessed according to the Disease Activity Score 28-C-reactive protein (DAS28-CRP4), Simplified Disease Activity Index (SDAI), and Clinical Disease Activity Index (CDAI) scores, and the recently proposed American College of Rheumatology (ACR)/European League against Rheumatism (EULAR) criteria for remission. Analyses were performed by using Kaplan-Meier survival curves.

Results

We identified 871 subjects with ≥2 years of follow-up. Of these subjects, 394 were in remission at one or more time-points and not in remission at baseline, according to at least one of the following criteria: DAS28-CRP < 2.6 (n = 309), DAS28-CRP < 2.3 (n = 275), SDAI (n = 168), CDAI (n = 170), and 2010 ACR/EULAR (n = 158). The median age for the 394 subjects at entrance to BRASS was 56 years; median disease duration was 8 years; 81% were female patients; and 72% were seropositive. Survival analysis performed separately for each remission criterion demonstrated that < 50% of subjects remained in remission 1 year later. Median remission survival time was 1 year. Kaplan-Meier curves of the various remission criteria did not significantly differ (P = 0.29 according to the log-rank test).

Conclusions

This study shows that in clinical practice, a minority of RA patients are in sustained remission.  相似文献

Background  

DNA methylation plays an important role in the process of tumorigenesis. Identifying differentially methylated genes or CpG islands (CGIs) associated with genes between two tumor subtypes is thus an important biological question. The methylation status of all CGIs in the whole genome can be assayed with differential methylation hybridization (DMH) microarrays. However, patient samples or cell lines are heterogeneous, so their methylation pattern may be very different. In addition, neighboring probes at each CGI are correlated. How these factors affect the analysis of DMH data is unknown.  相似文献

Pantoea agglomeransstrains are among the most promising biocontrol agents for a variety of bacterial and fungal plant diseases, particularly fire blight of apple and pear. We apply historical biogeography techniques to the macaques, baboons and their relatives (Primata: Papionini) and relate the inferred history of range shifts, and associated evolutionary events, to the latitudinal distribution of extant species, which is strongly tropical. The results of reversible parsimony, weighted ancestral area and dispersal-vicariance analyses all agree that Central Africa was part of the range of the ancestor of the tribe. Tropical regions with high current species richness (Central Africa, South-east Asia, Indonesia) have: (1) had a relatively long history of occupation, (2) experienced both a greater number and a greater average rate of speciation events and (3) given rise to more dispersal events to other regions. However, nested sister-taxon comparisons across the tribe show no overall association between differences in latitude and differences in rates of cladogenesis. Our historical reconstructions are largely consistent with previous hypotheses and fossil data, and suggest that both the passage of time since colonization and rates of cladogenesis have enhanced tropical species richness.
