北半球高山和极地虎耳草属物种丰富度的地理格局:温度和生境异质性的作用 现代气候、生境异质性和长期气候变化对森林生态系统中分布的木本植物的物种丰富度格局的影响在以往研究中受到广泛关注,但对高寒-极地生态系统中的草本植物物种丰富度格局及其影响因素的研究仍较少。本研究旨在检验以往研究中基于物种丰富度和环境因子关系提出的假说是否能够解释高寒-极地地区典型草本植物-虎耳草属(Saxifraga)的物种丰富度格局。本研究利用全球437种虎耳草属物种分布数据,探讨了全部物种、广域和狭域物种丰富度格局的影响因素。采用广义线性模型和空间自回归模型,评估了现代气候、生境异质性和历史气候对虎耳草属物种丰富度格局的影响。采用偏回归分析了不同变量对物种丰富度的独立解释率和共同解释率,并检验了4种广泛使用的物种丰富度与环境关系模型对物种丰富度格局的解释能力。研究结果表明,温度与虎耳草属所有物种和广域物种的物种丰富度格局呈显著负相关关系,是影响物种丰富度格局最重要的环境因子,这可能反映了虎耳草属对其祖先温带生态位的保守性。生境异质性和末次冰期以来的气候变化是虎耳草属狭域物种丰富度空间变异的最佳预测因子。总体而言,包含5个预测变量的组合模型可以解释大约40%–50%的虎耳草属物种丰富度的空间变异。此外,进化和生物地理过程在虎耳草属物种丰富度格局形成方面可能发挥了重要作用,这有待进一步研究。  相似文献

A new series of 1,3,4-thiadiazole-2-thione derivatives have been prepared and assayed for the inhibition of three physiologically relevant carbonic anhydrase (CA, EC 4.2.1.1) isozymes, the cytosolic human isozymes I and II, and the transmembrane, tumor-associated hCA IX. Against hCA I the investigated thiones, showed inhibition constants in the range of 2.55-222 microM, against hCA II in the range of 2.0-433 microM, and against hCA IX in the range of 1.25-148 microM. Compound 5c, 4-(4,5-dihydro-5-thioxo-1,3,4-thiadiazol-2-yl)-1-(5-nitro-2-oxoindolin-3-ylidene)semicarbazide showed interesting inhibition of the tumor-associated hCA IX with K(I) value of 1.25 microM, being the first non-sulfonamide type inhibitor of such activity. This result is rather important taking into consideration the known antitumor activity of thiones. In addition, docking of the tested compounds into CA II active site was performed in order to predict the affinity and orientation of these compounds at the isozyme active site. The results showed similar orientation of the target compounds at CA II active site compared with reported sulfonamide type CAIs with the thione group acting as a zinc-binding moiety.  相似文献

Weak intracellular penetration of antibiotics makes some infections difficult to treat. The Trojan horse strategy for targeted drug delivery is among the interesting routes being explored to overcome this therapeutic difficulty. Chlamydia trachomatis, as an obligate intracellular human pathogen, is responsible for both trachoma and sexually transmitted diseases. Chlamydia develops in a vacuole and is therefore protected by four membranes (plasma membrane, bacterial inclusion membrane, and bacterial membranes). In this work, the iron-transport protein, human serum-transferrin, was used as a Trojan horse for antibiotic delivery into the bacterial vacuole. Amoxicillin was grafted onto transferrin. The transferrin-amoxicillin construct was characterized by mass spectrometry and absorption spectroscopy. Its affinity for transferrin receptor 1, determined by fluorescence emission titration [KaffTf-amox = (1.3 ± 1.0) x 108], is very close to that of transferrin [4.3 x 108]. Transmission electron and confocal microscopies showed a co-localization of transferrin with the bacteria in the vacuole and were also used to evaluate the antibiotic capability of the construct. It is significantly more effective than amoxicillin alone. These promising results demonstrate targeted delivery of amoxicillin to suppress Chlamydia and are of interest for Chlamydiaceae and maybe other intracellular bacteria therapies.  相似文献

The black cutworm, Agrotis ipsilon, is an insect pest with great agricultural importance worldwide. In Egypt, it has acquired resistance against most of the conventional pesticides, particularly organophosphate insecticides, applied against it. Therefore, the present study aimed to investigate the effect of two benzoylureas (chitin synthesis inhibitors) namely chlorfluazuron and flufenoxuron on some developmental and reproductive parameters of Agrotis ipsilon that survived treatment of newly molted fourth instars with the LC50 of these compounds (1.00 and 4.68 mg/L, respectively) using leaf–dip method. These two compounds significantly enhanced the mean larval and pupal durations, whereas they significantly declined the mean percentage pupation, adult emergence, pupal weight, adult longevity, fecundity, and fertility compared to untreated insects. The mean pre-oviposition period was not significantly affected, whereas the mean oviposition period was significantly decreased. Both chlorfluazuron and flufenoxuron significantly decreased the mean total protein, carbohydrate, and lipid content in the ovaries compared to untreated insects, except for insignificant decrease in the total protein in flufenoxuron-treated ones. Additionally, the total ovariolar length was significantly atrophied. In conclusion, chlorfluazuron and flufenoxuron interfere with vitellogenesis of A. ipsilon. Moreover, they could suppress the population size of this insect species by affecting its development and reproductive potential.  相似文献

The alkaline and acid phosphatases were demonstrated histochemically along the whole length of the silk gland in normal and paralysed larvae of the rice mothCorcyra cephalonica. The activity of both enzymes was found to be much reduced as a result of the toxin injected in the host's body by the parasiteBracon hebetor. In both the normal and paralysed larvae the two enzymes were found to be more intense in the nuclei than in the cytoplasm. Both enzymes could not be detected in the cells of the productive tube region of the glands in paralyzed larvae.  相似文献

Brain cooling has pronounced effects on seizures and epileptic activity. The aim of the present study is to evaluate the anticonvulsant effect of brain cooling on the oxidative stress and changes in Na+, K+-ATPase and acetylcholinesterase (AchE) activities during status epilepticus induced by pilocarpine in the hippocampus of adult male rat in comparison with α-lipoic acid. Rats were divided into four groups: control, rats treated with pilocarpine for induction of status epilepticus, rats treated for 3 consecutive days with α-lipoic acid before pilocarpine and rats subjected to whole body cooling for 30 min before pilocarpine. The present findings indicated that pilocarine-induced status epilepticus was accompanied by a state of oxidative stress as clear from the significant increase in lipid peroxidation (MDA) and superoxide dismutase (SOD) and significant decrease in reduced glutathione and nitric oxide (NO) levels and the activities of catalase, AchE and Na+, K+-ATPase. Pretreatment with α-lipoic acid ameliorated the state of oxidative stress and restored AchE to nearly control activity. However, Na+, K+-ATPase activity showed a significant decrease. Rats exposed to cooling for 30 min before the induction of status epilepticus revealed significant increases in MDA and NO levels and SOD activity. AchE returned to control value while the significant decrease in Na+, K+-ATPase persisted. The present data suggest that cooling may have an anticonvulsant effect which may be mediated by the elevated NO level. However, brain cooling may have drastic unwanted insults such as oxidative stress and the decrease in Na+, K+-ATPase activity.  相似文献

It is not unusual to find common molecules among parasites of different species, genera, or phyla. When those molecules are antigenic, they may be used for developing drugs or vaccines that simultaneously target different species or genera of parasite. In the present study, we used a proteomic-based approach to identify proteins that are common to adult Fasciola hepatica and Schistosoma mansoni. Whole-worm extracts from each parasite were separated by 2-dimensional electrophoresis (2-DE), and digital images of both proteomes were superimposed using imaging software to identify proteins with identical isoelectric points and molecular weights. Protein identities were determined by mass spectrometry. Imaging and immunoblot analyses identified 28 immunoreactive proteins that are common to both parasites. Among these molecules are antioxidant proteins (thioredoxin and glutathione-S-transferase), glycolytic enzymes (glyceraldehyde 6-phosphate dehydrogenase and enolase), proteolytic enzymes (cathepsin-L and -D), inhibitors (Kunitz-type, Stefin-1), proteins with chaperone activity (heat shock protein 70 and fatty acid-binding protein), and structural proteins (calcium-binding protein, actin, and myosin). Some of the identified proteins could be used to develop drugs and vaccines against fascioliasis and schistosomiasis.  相似文献
