Purification of plasmid DNA by chromatographic methods

Chromatographic methods have been used to purify the DNA of plasmid RP1. DNA was purified in two stages. DNA was precipitated by ethanol and separated from RNA and proteins in Sepharose 4B column after lysis of plasmid containing cells by alkaline solution of sodium dodecylsulphate. Separation of the total DNA preparation and isolation of plasmid DNA was achieved at the second stage by chromatography on the hydroxyapatite column. The resulting purified plasmid DNA was free of RNA, protein and linear fragments of chromosomal DNA. The plasmid DNA kept intact native structure and possessed the transforming activity. The DNA of RP1 yield after purification by the described technique presented 70-80 micrograms per g of wet biomass.     

A genetically isolated population of Saccharomyces cerevisiae in Malaysia

A divergent population of Saccharomyces cerevisiae has been identified in Malaysia by molecular and genetic analysis. It has also demonstrated that the yeast S. bayanus may be found in South America. The origin of S. cerevisiae is discussed.     

Sensitivity to mouse toxin and level of macrophage 5-nucleotidase activity]

It is shown that immunomodulator of bacterial origin salmozan causes alternations of sensitivity to mouse toxin in mice CBA. A correlation exists between the sensitivity to mouse toxin and the level of 5-nucleotidase of peritoneal exudate macrophages.     

Lipid peroxidation in the adrenal cortex during exhausting stress

Under prolonged stress which is connected with exhaustion of functional resources of adrenal cortex the activation of lipid peroxidation processes in this gland was found. It is possible that the reason for such lipid peroxidation activation is the decrease in the content of adrenal cortex ascorbic acid and alpha-tocopherol.
