The data have been compared on the yeast genomic divergence obtained in genetic molecular and biological studies of the yeast genera Williopsis and Zygowilliopsis taken as an example. The hybridization analysis, the enzyme electrophoresis and DNA reassociation comprise the noncontradictory estimation of the yeasts relation. Different methods demonstrate a convincing species discreteness among the yeast genomes. The reproductive isolation of the yeast taxons is defined more invariably by the genetic methods. A new combination Williopsis sargentensis (Wickerham et Kurtzman) Naumov comb. nov. has been formed.  相似文献

After reevaluation of the taxonomic position of 27 yeast collection strains of different origin by UP-PCR followed by dot-hybridization, only 22 strains were assigned to the biological species Zygowilliopsis californica (Lodder) Kudriavzev. Four strains were identified as Williopsis suaveolens (Kl?cker) Naumov et al. Universal primers L45 and N21 are recommended for identification of the Z. californica yeasts.  相似文献

In the Americas, areas with a high risk of malaria transmission are mainly located in the Amazon Forest, which extends across nine countries. One keystone step to understanding the Plasmodium life cycle in Anopheles species from the Amazon Region is to obtain experimentally infected mosquito vectors. Several attempts to colonise Ano- pheles species have been conducted, but with only short-lived success or no success at all. In this review, we review the literature on malaria transmission from the perspective of its Amazon vectors. Currently, it is possible to develop experimental Plasmodium vivax infection of the colonised and field-captured vectors in laboratories located close to Amazonian endemic areas. We are also reviewing studies related to the immune response to P. vivax infection of Anopheles aquasalis, a coastal mosquito species. Finally, we discuss the importance of the modulation of Plasmodium infection by the vector microbiota and also consider the anopheline genomes. The establishment of experimental mosquito infections with Plasmodium falciparum, Plasmodium yoelii and Plasmodium berghei parasites that could provide interesting models for studying malaria in the Amazonian scenario is important. Understanding the molecular mechanisms involved in the development of the parasites in New World vectors is crucial in order to better determine the interaction process and vectorial competence.  相似文献

Peculiarities of forming emission flows of carbon dioxide and methane in forest-steppe raised Sphagnum bogs have been studied in view of coenotic structure and damage to the vegetation as a result of fire. The reasons of relatively low rate of methane emission in pine-dwarf shrub Sphagnum bogs under the marginal conditions have been analyzed. Regulation of the cardiac rhythm is intricate and occurs at least at two major levels, intrinsic and extrinsic. In turn, each of these levels can be divided into several sublevels. The factors regulating the cardiac activity eventually affect the duration of spontaneous diastolic depolarization of pacemaker myocytes of the sinoatrial node and, to a far lesser extent, the conduction velocity in the conduction system of the heart. Intrinsic regulation of the heart rate (HR) includes the myogenic sublevel and the sublevels of cell-to-cell communication, the cardiac nervous system, and humoral factors produced within the heart. Myogenic regulation is considered to be the first sublevel in control of the cardiac function. The available data suggest myogenic regulation only for the contractility of the myocardium. The cell-to-cell regulation sublevel involves two principal mechanisms. One depends on the heterogeneous structure of the sinoatrial node and within-node shifts of the dominant pacemaker, which is a group of cells that determine the HR and govern all other cells of the sinoatrial node. The other mechanism is based on the effects of peptides produced by cardiomyocytes and endothelial cells on pacemaker cells of the sinoatrial node. Regulatory peptides are also produced by the cardiac nervous system, which includes sensory and effector autonomic fibers, represents the cardiac part of the metasympathetic system, and forms intramural ganglia. In addition to modulating the HR, these peptides affect the contractility, microcirculation, coronary blood flow, preload, and afterload. Currently available data demonstrate that the autonomic nervous system is far more intricate than believed earlier. Fifty-three strains of saturn-spored yeasts were analyzed by means of restriction analysis of the amplified fragment of rDNA which comprised the 5.8S rRNA gene and the internal transcribed spacers ITS1 and ITS2. The use of endonucleases HaeIII and MspI enabled clear differentiation of yeast species Williopsis mucosa, W. salicorniae, Zygowilliopsis californica, Komagataea pratensis, and the Williopsis sensu stricto complex. Minisatellite primer M13 was proposed for the differentiation between twin species of Williopsis sensu stricto, which have identical restriction profiles. PCR with primer M13 enabled reidentification of a number of collection strains, species identification of saturn-spored isolates from the Far East, and detection of three strains affiliated to novel taxa. The latter have unique PCR profiles and differ in the nucleotide sequences of ITS1 and ITS2 fragments of rDNA. Possible variations in the results obtained with different molecular methods are discussed.  相似文献

Amino acid sequence analysis of alpha-galactosidases and other proteins of glycoside hydrolase family 27 (GH27) allowed isolation of three major subfamilies, 27a-27c. Unique isomalto-dextranase of Arthrobacter globiformis clustered separately. Eukaryotic proteins formed five clusters on a phylogenetic tree of the family. Bacterial GH27 proteins, which are relatively few, did not form stable clusters. A monophyletic origin of the GH27 family was demonstrated with the use of related proteins of the GH36 family. The structure of the active center and evolution of alpha-galactosidases are discussed.  相似文献

A new bidentate chelating pyrazolylpyrimidine ligand bearing a strong electron-donating substituent, i.e. 4-(3,5-diphenyl-1H-pyrazol-1-yl)-6-(piperidin-1-yl)pyrimidine (L) (Scheme 1), has been synthesized and used to obtain the copper(II) complexes by reaction with CuCl2. The molar ratio Cu:L = 1:2 leads to isolation of a complex having CuL2Cl2 empirical formula, while the molar ratio Cu:L = 1:1 gives a complex with CuLCl2 empirical formula. The crystal structure of L as well as the structures of both complexes were studied by single crystal X-ray diffraction. The crystal structure of CuL2Cl2 compound is formed by trans-[CuL2Cl2] mononuclear molecules. Surprisingly, in contrast to the previous compound having molecular structure, the crystal structure of CuLCl2 consists of mononuclear [CuL2Cl]+ complex cations and dinuclear [Cu2Cl6]2− anions. Thus, formula of CuLCl2 complex can be represented as [CuL2Cl]2[Cu2Cl6]. In both complexes molecules of L adopt bidentate chelating coordination mode through N2 atom of pyrazole and N3 atom of pyrimidine rings forming five-membered CuN3C metallocycles. Owing to C-H···N interactions and π-π-stacking L molecules form 2D network. In the structure of trans-[CuL2Cl2] there exist double lone pair(N(piperidine))-π(pyrimidine) interactions and C-H···Cl contacts resulting in the formation of 1D chains. Layered 2D structure of [CuL2Cl]2[Cu2Cl6] results from C-H···Cl, C-H···π and double lone pair(Cl([CuL2Cl]+ complex cation)-π(pyrimidine) interactions.  相似文献

A significant heterogeneity of the species Zygowilliopsis californica was revealed using RFLP-analysis of the PCR-amplified rDNA fragment spanning the 5.8S rRNA gene and the internal transcribed spacers ITS1 and ITS2. Phylogenetic analysis of the nucleotide sequences of ITS1 and ITS2 rDNA differentiated three varieties: Z. californica var. californica, Z. californica var. dimennae, and Z. californica var. fukushimae. The most variable was the ITS 2 region, where 7-26 nucleotide substitutions were revealed. The varieties formed semisterile hybrids with meiotic segregation of control markers. The limits of the phylogenetic species concept are discussed.  相似文献
