Thrips tabaci is a major pest of high-value vegetable crops and understanding its population genetics will advance our knowledge about its ecology and management. Mitochondrial cytochrome oxidase subunit I (COI) gene sequence was used as a molecular marker to analyze T. tabaci populations from onion and cabbage fields in New York. Eight COI haplotypes were identified in 565 T. tabaci individuals collected from these fields. All T. tabaci were thelytokous and genetically similar to those originating from hosts representing seven plant families spanning five continents. The most dominant haplotype was NY-HT1, accounting for 92 and 88% of the total individuals collected from onion fields in mid-summer in 2005 and 2007, respectively, and 100 and 96% of the total in early fall in 2005 and 2007, respectively. In contrast, T. tabaci collected from cabbage fields showed a dynamic change in population structure from mid-summer to early fall. In mid-summer, haplotype NY-HT2 was highly abundant, accounting for 58 and 52% of the total in 2005 and 2007, respectively, but in early fall it decreased drastically to 15 and 7% of the total in 2005 and 2007, respectively. Haplotype NY-HT1 accounted for 12 and 46% of the total in cabbage fields in mid-summer of 2005 and 2007, respectively, but became the dominant haplotype in early fall accounting for 81 and 66% of the total in 2005 and 2007, respectively. Despite the relative proximity of onion and cabbage fields in the western New York landscape, T. tabaci populations differed seasonally within each cropping system. Differences may have been attributed to better establishment of certain genotypes on specific hosts or differing colonization patterns within these cropping systems. Future studies investigating temporal changes in T. tabaci populations on their major hosts in these ecosystems are needed to better understand host-plant utilization and implications for population management.  相似文献

Repellents used to reduce by-kill of birds during pest control must not compromise acceptance by target species. Two repellents combined, anthraquinone (AQ; 0.4 g kg?1) and d-pulegone (DP; 1.0) did not reduce the palatability of blue-coloured carrot baits to laboratory rats (Rattus norvegicus); nor did DP (2.0). Green-coloured carrot baits coated with AQ, DP or AQ + DP were taken from bait stations by wild possums (Trichosurus vulpecula) and rats. Toxic (1080) bait coated with AQ (0.4) and peanut oil (0.1) had reduced palatability but was accepted by laboratory rats. However, laboratory rats did not consume enough baits coated with AQ and bacon, peanut butter, cinnamon or DP to be killed. Anthraquinone (0.4 or 0.8) plus cinnamon and DP (0.5) did not affect palatability or lethality to captive ship rats (R. rattus) or possums. Anthraquinone and DP as surface coatings on baits are therefore acceptable to possums and possibly rats, at concentrations that deter some bird species.  相似文献

The response of potato, Solanum tuberosum L., tuber yield to stem injury by European corn borer, Ostrinia nubilalis (Hübner), larvae was investigated in the Mid-Atlantic region of the United States for 3 yr. This response was described for 'Superior', 'Atlantic', and 'Snowden' potato, which are early-season, midseason, and late-season maturing cultivars, respectively. To model the yield/injury relationship, a range of corn borer injury levels was established in the field by augmenting the natural infestation with varying densities of laboratory-reared larvae. Linear and nonlinear regression analyses (plateau and second-order polynomial models) were used to describe the relationship between yield of U.S. No. 1 grade tubers and the percentage of stems injured by corn borer larvae. The maturity of the cultivar did not affect the response of potato yield to stem injury. In nine of 14 experiments, potato tolerated high levels of corn borer injury (55-90% of stems injured) without yield loss, suggesting that control of corn borer may not be necessary. Yet, in one of five Superior tests, in two of four Atlantic tests and in two of five Snowden tests, corn borer injury significantly reduced yield. Of the five data sets in which corn borer injury reduced yield, the plateau model fit two data sets and the quadratic model fit one data set. In two of the three cases, these models accounted for nearly identical amounts of total variation in yield as that accounted for by the linear model. The linear model fit four of the five data sets, but the R2 values were low for three of the four tests (0.10, 0.18, and 0.31). The parameter or parameters that interact with corn borer injury to cause tuber yield reduction should be identified before economic injury levels and thresholds are developed.  相似文献

Although the corn leafhopper Dalbulus maidis (DeLong and Wolcott) is the most important vector of maize pathogens in Latin America, little is known about how and where it overwinters (passes the dry season), particularly in Mexico. The objectives of this study were (1) to monitor the abundance of D. maidis adults throughout the dry season in maize and maize-free habitats and (2) to determine where and how D. maidis adults, exposed or nonexposed to the maize pathogen Spiroplasma kunkelii Whitcomb, overwinter in a maize-free habitat. Work for the first objective was done during the two consecutive dry seasons of 1999-2000 and 2000-2001; the second objective was done during the dry seasons of 2003-2004 and 2005-2006. During the dry winter seasons, D. maidis was prevalent as long as maize was present in irrigated areas. The leafhopper was found in 52 of the 58 irrigated maize fields sampled in Mexico at the end of the dry seasons of 1999-2000 and 2000-2001. However, leafhopper adults were not found in nonirrigated maize-free habitats at high elevation during the dry winter season (February, March, and April), although leafhopper adults were prevalent on perennial wild grasses in January after maize harvest. Additional experiments revealed, however, that corn leafhopper adults, although few in number, survived the entire dry season in these nonirrigated maize-free fields. Also, no detectable difference in survival existed between leafhoppers exposed and those not exposed to S. kunkelli during the two dry seasons in the maize-free habitat.  相似文献

Neutral endopeptidase (EC 3.4.24.11) is a major constituent of kidney brush border membranes. It is also present in the brain where it has been shown to be involved in the inactivation of opioid peptides, methionine- and leucine-enkephalins. For this reason this enzyme is often called 'enkephalinase'. In order to characterize the primary structure of the enzyme, oligonucleotide probes were designed from partial amino acid sequences and used to isolate clones from kidney cDNA libraries. Sequencing of the cDNA inserts revealed the complete primary structure of the enzyme. Neutral endopeptidase consists of 750 amino acids. It contains a short N-terminal cytoplasmic domain (27 amino acids), a single membrane-spanning segment (23 amino acids) and an extracellular domain that comprises most of the protein mass. The comparison of the primary structure of neutral endopeptidase with that of thermolysin, a bacterial Zn-metallopeptidase, indicates that most of the amino acid residues involved in Zn coordination and catalytic activity in thermolysin are found within highly honmologous sequences in neutral endopeptidase.  相似文献

A quantitative assay was used to measure the rate of collection of a population of embryonic neural retina cells to the surface of cell aggregates. The rate of collection of freshly trysinized cells was limited in the initial stages by the rate of replacement of trypsin-sensitive cell- surface components. When cells were preincubated, or "recovered," and then added to cell aggregates, collection occurred at a linear rate and was independent of protein and glycoprotein synthesis. The adhesion of recovered cells was temperature and energy dependent, and was reversibly inhibited by cytochalasin B. Colchicine had little effect on collection of recovered cells. Antiserum directed against recovered cell membranes was shown to bind to recovered cells by indirect immunofluorescence. The antiserum also was shown to inhibit collection of recovered cells to aggregates, suggesting that at least some of the antigens identified might be involved in the adhesion process. The inhibitory effect of the antiserum was dose dependent . Freshly trypsinized cells absorbed neither the immunofluorescence activity nor the adhesion-inhibiting activity. Recovered cells absorbed away both activities. In specificity studies, dorsal neural retina cells adhered to aggregates of ventral optic tectum in preference to aggregates of dorsal optic tectum. The adhesive specificity of the dorsal retina cells was less sensitive to trypsin than the adhesive specificity of ventral retina cells which adhered preferentially to dorsal tectal aggregates only after a period of recovery.  相似文献

A helper component from maize chlorotic dwarf machlovirus (MCDV)-infected plants was necessary for insect transmission of purified MCDV. Purified MCDV, WS strain, when acquired by membrane feeding, was transmitted byGraminella nigrifrons (Forbes) (Homoptera: Cicadellidae) which fed first on MCDV (M1 strain)-infected maize. Conversely, feeding on MCDV-WS-infected maize allowed transmission of purified MCDV-M1, indicating that the helper component was not strain specific. ViruliferousG. nigrifrons lost the ability to transmit MCDV-M1 after feeding for 24 h on healthy maize, but retained the ability to acquire and transmit purified MCDV-WS for up to 36 h. Wheng. nigrifrons fed initially on MCDV-infected plants and then on a second strain of purified MCDV, the second strain of MCDV could be transmitted alone, suggesting that the helper component from MCDV-infected plants was something other than the virion itself. Lengthening acquisition feeding ofG. nigrifrons on MCDV-M1-infected maize or purified MCDV-WS did not significantly change the transmission frequency of MCDV-WS.Amblysellus grex (Oman), an experimental vector of MCDV, also transmitted purified MCDV-WS after an initial acquisition feeding on MCDV-M1-infected maize. A larval immersion bioassay was developed to identify susceptibility of onion maggot, Delia antiqua (Meigen) (Diptera: Anthomyiidae), to chlorpyrifos and to determine whether this assay could be used to predict control in onion fields. Laboratory colonies were established from larvae collected in New York onion fields during 2003 and 2004, providing us with test insects to use in bioassays. The larval assay effectively determined susceptibility of D. antiqua to chlorpyrifos, and results were congruent with an adult bioassay. However, use of similar-aged larvae (4 d old) in the assays was critical because larvae became more tolerant to chlorpyrifos as they aged. In a field survey, six of the 13 populations had LC50 values above the recommended field rate of 3,600 ppm (range 4,031-6,869). Over two successive seasons in the same field, susceptibility of D. antiqua to chlorpyrifos decreased in two of three fields (by 45 and 42%) and remained the same in another field, indicating that resistance is not predictable from year to year. Based on the relationship between damage in the field and LC50 values from 11 of the populations mentioned above, all five populations that had LC50 values above the field rate caused unacceptable levels of damage, whereas five of six populations that had LC50 values below the field rate did not cause serious damage.  相似文献
