A highly sensitive neutron-induced prompt γ-ray analysis (PGA) system, usable at both cold and thermal neutron beam guides of JRR-3M, has been constructed. The system was designed to achieve the lowest γ-ray background by using lithium fluoride tiles as neutron shielding, by placing the samples in a He atmosphere and by using a Ge-bismuth germanate detector system for Compton suppression. The γ-ray spectrometer can acquire three modes of spectra simultaneously: single, Compton suppression, and pair modes. Because of the low-energy guided neutron beams and the low-background system, analytical sensitivities and detection limits better than those in usual PGA systems have been achieved. Boron and multielemental determination by a comparative standardization have been investigated, and accuracy, precision, and detection limits for the elements in various materials were evaluated. The system has been applied to the determination of B and multielements in samples of various fields such as medical, environmental, and geological sciences.  相似文献

Mitochondria isolated from 5-day-old pea cotyledons had lowrespiration activity and did not respond to exogenous ADP, whilethose from 1-day-old cotyledons respired actively and respondedto ADP. The former mitochondria, but not the latter, were verysusceptible to destruction during extraction and purification.The mitochondrial inner membrane isolated from 5-day-old cotyledonswas less dense than that from 1-day-old cotyledons. The specificactivity of SDH in the former membrane was lower than that inthe latter, while both membranes were similar to each otherwith respect to the specific activity of Cyt ox. Disc electrophoresisof solubilized membrane on polyacrylamide gel containing SDSshowed that the mitochondrial inner membrane from 5-day-oldcotyledons contained lesser amounts of several polypeptidescompared with that from 1-day-old cotyledons. Abstract The hypothesis that the microbially catalyzed pathway proceeds with a step that would yield the highest energy was examined for the reductive dechlorination of chloroanilines (CAs) under anaerobic conditions. The Gibbs free energy of formation was estimated with Benson's method, then the redox potentials were determined using an H+/H2 couple as the reference system. The observed pathways were compared using the redox potential of the reaction, and the results showed that the redox potential correctly predicts the pathway that yields the highest energy for the dechlorination step. Clonal B16 mouse melanoma conv cells are tumorigenic spindle-shaped cells (S-type cells) exhibiting tyrosinase activity and melanosomes under usual culture conditions. When the cells passaged on glass substratum were plated for colony formation on plastic substratum in Eagle's minimum essential medium (MEM) with 10% calf bovine serum, most of them converted to fibroblastlike cells (F-type cells) with the coupled loss of differentiated and tumorigenic phenotypes. However, they continued to be S-type cells provided that they were plated on glass substratum. The conversion from S- to F-type cell was not induced with high frequency even on plastic substratum when the concentration of calf serum in the medium was low (1-2%). These results indicate that both plastic substratum and serum factor are requisites for converting the phenotypic expression of the conv cells. Partial characterization of the serum factor indicates that it is adsorbable to plastic substratum, inactivated at 70 degrees C for 10 min, salted out at 40% of saturated ammonium sulfate; in addition the factor seems to act on cells within 1 day after plating.  相似文献

The decapeptide corresponding to the amino acid sequence of porcine luteinizing hormone-releasing hormone (LH-RH) which involves 1 mol of tryptophan was synthesized via solid phase synthesis with two different deblocking procedures which used hydrogen chloride in formic acid and hydrogen chloride in acetic acid containing 1% 2-mercaptoethanol. After some fundamental studies on the former reagent with respect to deblocking efficiency toward the Boc group, 0.5 M hydrogen chloride (a 10-fold molar excess with respect to the N-terminal Boc group) in formic acid was used in the present synthesis. The two synthetic products exhibited the same chemical and biological properties as an authentic LH-RH. Hydrogen chloride in formic acid has proved effective without a scavenger although loss of peptide from the resin occurred to a somewhat greater extent than that with hydrogen chloride in acetic acid. A derivative of the synthetic LH-RH formylated at the indole nitrogen had a greatly diminished biological activity, indicating that the intact indole side chain is essential for the activity.  相似文献

The antibiotic gramicidin S (GS) has the structure of cyclo (-L-Val1-L-Orn2-L-Leu3-D-Phe4-L-Pro5-L-Val1'-L-Orn2'-L-Leu3'-D-Phe4'-L-Pro5'-) and is basic in character. Five GS analogs including [Gly1,1']-GS and the neutral [L-Hnv2,2']-GS (Hnv represents delta-hydroxynorvaline) were synthesized by the solid-phase method to evaluate the role of L-Val1,1' and L-Orn2,2' residues in GS. The hybrid analogs [( Gly1]-GS and [L-Hnv2]-GS) and [D-Tyr4,4']-GS showed high antibacterial activities, whereas [Gly1,1']-GS and [L-Hnv2,2']-GS possessed no activity. Inhibitory effects by these analogs for the adsorption of 14C-labeled GS on cells of bacteria sensitive to GS were determined. The structure-activity relationship of GS is discussed on the basis of the results on these GS analogs.  相似文献

Adenosine 3',5'-cyclic phosphate (cAMP) is efficiently hydrolyzed at pH 7, 50 degrees C by use of [Co-(trien) (H2O)2]3+ and [Co(tme)2-(H2O)2]3+ complexes as catalysts: trien (diethylenetriamine) and tme (1,1,2,2-tetramethylethylenediamine). The acceleration is remarkable (10(8) to 10(9) fold), decreasing half-life of cAMP from 660,000 years to 7-15 hours.  相似文献
