We have devised a novel method for automated microsatellite analysis using "universal" fluorescent labeling. This system is based on polymerase chain reactions driven by sequence-specific primers and a reporter primer labeled with a fluorescent dye at its 5' end. The forward sequence-specific primer is designed with a tag region bearing no homology to any human genomic sequence. Complementary tag sequences act as templates for the 6-carboxyfluorescein-labeled reporter primer, and those products can be analyzed with an autosequencer. The results we achieved with this assay system were consistent with the results of conventional assays using radioisotope-labeled primers, and diagnosis required less time. Furthermore, the fluorescent-labeled reporter primer is "universal" in that it can be used with different sequence-specific primers designed to carry the appropriate tag sequence at their 5'-ends. Our observations suggest that the "universal" fluorescent labeling method is an efficient tool for analyzing sequence variations in human DNA.  相似文献

A deoxyfluorocellulose derivative, with regioselective substitution mostly at the C-6 carbon atoms, has been studied by an X-ray diffraction method. The results showed that the crystallinity of the fluorinated cellulose is low but can be enhanced by a specific conversion treatment. The hydrothermally treated sample displayed a spectrum similar to that of cellulose IV.  相似文献

In previous studies, we identified a novel gene, factor for adipocyte differentiation 24 (fad24), which plays an important role during the early stages of adipogenesis in mouse 3T3-L1 cells. Moreover, overexpression of fad24 increased the number of smaller adipocytes in white adipose tissue and improved glucose metabolic activity in mice, thus indicating that fad24 functions as a regulator of adipogenesis in vivo. However, the physiological roles of fad24 in vivo are largely unknown. In this study, we attempted to generate fad24-deficient mice by gene targeting. No fad24-null mutants were recovered after embryonic day 9.5 (E9.5). Although fad24-null embryos were detected in an expected Mendelian ratio of genotypes at E3.5, none of the homozygous mutants developed into blastocysts. In vitro culture experiments revealed that fad24-null embryos develop normally to the morula stage but acquire growth defects during subsequent stages. The number of nuclei decreased in fad24-deficient morulae compared with that in wild-type ones. These results strongly suggested that fad24 is essential for pre-implantation in embryonic development, particularly for the progression to the blastocyst stage.  相似文献

Lipid transfer proteins (LTPs) are the key contributor of organelle-specific lipid distribution and cellular lipid homeostasis. Here, we report a novel implication of LTPs in phagocytosis, trogocytosis, pinocytosis, biosynthetic secretion, recycling of pinosomes, and motility of the parasitic protist E. histolytica, the etiological agent of human amoebiasis. We show that two StAR-related lipid transfer (START) domain-containing LTPs (named as EhLTP1 and 3) are involved in these biological pathways in an LTP-specific manner. Our findings provide novel implications of LTPs, which are relevant to the elucidation of pathophysiology of the diseases caused by parasitic protists.  相似文献

Although agricultural systems in tropical monsoon Asia play a central role in the global nitrogen (N) cycle, details of the N cycle in this region on a watershed scale remain unclear. This study quantified the N budget in a tropical watershed of 221 km2 on Java Island, where paddy fields cover 28% of the land, by conducting field surveys. The amount of net biochemical gaseous N loss to the atmosphere (X GB ), which is generally difficult to determine, was calculated as the residual of the N balance. Assuming that NH3 volatilization balances deposition, and hence subtracting NH4–N from the N import with atmospheric deposition, the average total import and export of N per year was found to be 46.5 kg ha−1 year−1 over the watershed. Of this, 71% was imported as fertilizer (M F ) and 29% with atmospheric deposition (M AD ). On the export side, 42% was lost as X GB , 37% with incineration of rice residues and wood fuel (X GI ), 13% with river discharge (X D ) and 9% with rice surplus export (X R ). A large portion of X GB , and consequently, a small portion of X D could be explained by the high rate of denitrification resulting from the high temperature and humid climate, and are thought to be common features of tropical watersheds where paddy fields are found.  相似文献

The thiol protease was purified from adult Paragonimus ohirai by alpha 1-antitrypsin-Sepharose, Sephadex G-75 and CM-cellulose, measuring its activities to hydrolyze hemoglobin and tosyl-L-lysine alpha-naphthyl-ester. The purified protease showed a single band on polyacrylamide disc gel isoelectrophoresis as zymogram with Tos-Lys-NE and also by protein staining, and its pI was found to be 6.4. The molecular weight was calculated to be 29,000 by gel filtration and 27,000 by SDS-polyacrylamide gel electrophoresis as a single polypeptide. The protease hydrolyzed hemoglobin and Tos-Lys-NE optimally at pH 4.0 and 5.0, respectively. The both hydrolyzing activities were inhibited by alpha 1-AT and soybean trypsin inhibitor as well as thiol protease inhibitors such as antipain, E-64 and p-hydroxymercuriphenylsulfonate. These results indicate that this enzyme is a new type thiol protease.  相似文献

A practical and convenient method for discriminating between the presence and the absence of sialic acid in carbohydrate chains of glycoproteins was devised using paramagnetic beads and two lectins, Sambucus sieboldiana lectin (SSA) and Ricinus communis agglutinin (RCA120). The glycoproteins of transferrin or thyroglobulin were firstly captured to paramagnetic beads that were previously coated with corresponding antibody, and then the lectins of RCA120-biotin and SSA-FITC were bound to the glycoproteins on the paramagnetic beads. Finally, the fluorescence intensity of the beads was measured to determine the ratios of lectins RCA120-biotin/Cy5-streptavidin to SSA-FITC. The mixed lectins method showed bigger difference of the ratios between the presence and the absence of sialic acid, indicating higher discrimination efficiency than the ordinary non-mixed lectins method. Furthermore, statistical analysis by two-side t-test indicated that the mixed lectins method was more highly reliable than the ordinary non-mixed lectins method in discriminating between the presence and the absence of sialic acid. The reaction with the two lectins can be performed in a single tube and readily automated taking advantage of the use of paramagnetic beads.  相似文献

RS-8359, (+/-)-4-(4-cyanoanilino)-5,6-dihydro-7-hydroxy-7H-cyclopenta[d]-pyrimidine is a selective and reversible MAO-A inhibitor. The (S)-enantiomer of RS-8359 has been demonstrated to be inverted to the (R)-enantiomer after oral administration to rats. In the current study, we investigated the chiral inversion mechanism and the properties of involved enzymes using rat liver subcellular fractions. The 7-hydroxy function of RS-8359 was oxidized at least by the two different enzymes. The cytosolic enzyme oxidized enantiospecifically the (S)-enantiomer with NADP as a cofactor. On the other hand, the microsomal enzyme catalyzed more preferentially the oxidation of the (S)-enantiomer than the (R)-enantiomer with NAD as a cofactor. With to product enantioselectivity of reduction of the 7-keto derivative, it was found that only the alcohol bearing (R)-configuration was formed by the cytosolic enzyme with NADPH and the microsomal enzyme with NADH at almost equal rate. The reduction rate was much larger than the oxidation rate of 7-hydroxy group. The results suggest that the chiral inversion might occur via an enantioselectivity of consecutive two opposing reactions, oxidation and reduction of keto-alcohol group. In this case, the direction of chiral inversion from the (S)-enantiomer to the (R)-enantiomer is governed by the enantiospecific reduction of intermediate 7-keto group to the alcohol with (R)-configuration. The enzyme responsible for the enantiospecific reduction of the 7-keto group was purified from rat liver cytosolic fractions and identified as 3alpha-hydroxysteroid dehydrogenase (3alpha-HSD) via database search of peptide mass data obtained by nano-LC/MS/MS.  相似文献

We studied rattan communities in four forest types in Borneo and Java to clarify the pattern of species diversity and vegetation structure. Within six plots of 4.82 ha in total, we found 42 rattan species in six genera with 4,736 stems (≥20 cm long). There was a significant positive correlation between rattans and trees in species diversity, decreasing in the order mixed dipterocarp (for rattans: Shannon–Wiener diversity index, H′ = 2.87–3.34) > alluvial (1.96) > lower montane (1.43) > peat swamp forests (1.34). This pattern coincided with the diversity of the regional flora. The density of rattan stems (ha–1) decreased in the order lower montane (5,997) > mixed dipterocarp (598–992) > alluvial (592) > peat swamp forests (162). The maximum height of rattans would be determined by the canopy height, rather than by the maximum tree height. The rattan stem volume ranged from 0.25 to 1.88 m3 ha−1 and was not correlated with the stem density. The cool montane climate seemed to decrease the maximum height of rattans, and a few small rattans made denser colonies. Swampy environments apparently suppressed the survival of non-climbing rattans strongly, but that of tall climbing rattans less so.  相似文献
