Ethacrinic acid (0.5 mg/ml), similarly to strophanthin K, increased sodium content and decreased potassium content in the tissue of the frog urinary bladder. Furocemide (0.1 mg/ml) failed to change the ionic content of this tissue. These data indicated a difference in the intracellular action of furosemide and ethacrynic acid.  相似文献

Electron and confocal microscopy, using immunocytochemical methods, was employed to assess osmotic water permeability of the frog (Rana temporaria) urinary bladder during transcellular water transport, induced by antidiuretic hormone (ADH) or by wash-out of autacoids from serosal, ADH-free Ringer solution. The increase of osmotic water permeability of the urinary bladder was accompanied by relevant ultrastructural changes, the most remarkable being: (1) the appearance of aggregates of intramembranous particles in the apical membrane of granular cells, and the extent of the membrane area covered by the aggregates proportional to that of the water flow; (2) redistribution of actin filaments in the cytoplasm of granular cells; judging from the anti-actin label density, the number of actin filaments in the apical region of cytoplasm was reduced by 2.5–4 times compared with normal; (3) a decrease in the total electron density of the cytoplasm due to the increased water content of granular cells.  相似文献

(1) The Na, K, Ca, Mg concentration of blood serum have been studied in nearly 60 fish species from various waterbodies (the Neva, Lake Dal'nee, the Volga, Lake Baikal, the White Sea, the Black Sea, the Barents Sea etc.). In fishes (Cyprinidae, Percidae, Siluridae etc.) from fresh water (Lake Dal'nee, the Baikal, the Neva) with low 0.18–0.24 mEq/1 Na concentration Na content in blood serum was lower than in closely related species from fresh water (the Volga, Lyutoga River) with larger Na concentration–nearly 0.5–1 mEq 1. Blood serum of marine teleosts Na concentration (156–210 mEq/1) was higher than in migrating and freshwater species (90–174) mEq/1) and it did not depend on seawater salinity: Barents Sea–465, White Sea–255, Black Sea–227 mEq/1.
(2) Rather a considerable specific variation of ion concentration was found in blood serum of fishes from different families living in the same water body (Lake Baikal: Cyprinidae 90–95 mEq/1, Percidae 114 mEq/1, Salmonidae 126–129 mEq/1 etc.; the Volga: Siluridae 139 mEq/1, Percidae 142–150 mEq/1, Clupeidae 174 mEq/1).
(3) No relation has been detected between K, Ca and Mg concentrations in blood serum of fishes and the concentrations of these ions in the environment.
(4) Na concentration in the blood serum of sockeye salmon juveniles (101 mEq/1) migrating seaward was less than in adult fishes (132 mEq/1) returning to the lake to spawn. Na concentration in the blood of non-migrating salmonids ( Oncorhynchus nerka adult residual 104 mEq/1, Salvelinus alpinus juvenile 140 mEq/1, adult residual 141 mEq/1) changed insig- nificantly during life history. Notable hypopotassemia was observed in spawning time in Salmonidae.
(5) The comparison of blood serum ion concentration in males and females of fishes from different taxonomic groups (Acipenseridae, Salmonidae, Cyprinidae) showed no significant variation.  相似文献

The aim of this study was to compare the effects of exenatide, a glucagon-like peptide-1 (GLP-1) mimetic, on glucose and water–salt homeostasis in animals with different levels of renal proximal tubular reabsorption, rats (Rattus norvegicus) and frogs (Rana temporaria). Following the glucose tolerance test, in rats exenatide promoted fast recovery of normoglycemia, whereas in frogs it delayed this process. In water-loaded rats, exenatide augmented solute-free water clearance and enhanced natriuresis in furosemide-treated animals. In frogs, exenatide did not alter the urinary flow rate, urinary sodium excretion and solute-free water clearance under water diuresis and furosemide treatment. It is suggested that the involvement of GLP-1 in regulation of water–salt homeostasis in mammals was preceded by a key evolutionary transformation, the increase in the glomerular filtration rate and proximal tubular reabsorption.  相似文献

Changes in osmolality and the concentration of cations (Na, K, Ca, Mg) were studied in blood serum of pregnant women from two weeks after conception, throughout the whole pregnancy, and within the first week after delivery. Altogether 239 women from 18 to 40 years of age were studied. Blood serum osmolality decreased from 287±0.8 to 278±1.6 mOsm/kg H2O from the fifth week of pregnancy and remained virtually at this level until the end of pregnancy. Hyponatremia was found during the three trimesters of pregnancy, in trimesters II and III hypokalemia was not observed, whereas hypocalcemia and hypomagnemia were found. On the first day after delivery, the blood serum osmolality and concentrations of magnesium ions returned to their levels in nonpregnant women, whereas concentrations of sodium and calcium ions remained decreased. No correlation was found between hypoosmia and changes in blood serum concentrations of ions under study during the three trimesters of pregnancy. Thus, in normal pregnancy, hypoosmia develops from the fifth week after conception and persists until delivery. The concentrations of sodium, potassium, calcium, and magnesium ions are regulated by independent mechanisms to provide retention of these parameters within certain periods of pregnancy at the level of nonpregnant women on the background of hypoosmia.  相似文献

Functional expression of bovine visual rhodopsin in the cell-free translation system with cotranslational insertion of the protein into phosphatidylcholine liposomes is described. The recombinant rhodopsin has spectral and functional properties similar to those of natural rhodopsin from bovine retina. Two mutant rhodopsins with amino acid substitutions in the hydrophilic C-terminal domain were obtained using oligonucleotide-directed mutagenesis. It was found that substitution Cys-316----Ser does not affect rhodopsin's ability to activate the visual amplification cascade, whereas double mutation Asp-330----Asn, Asp-331----Asn dramatically lowers the rhodopsin functional activity.  相似文献
