Über die Blutkapillaren und perivaskulären Strukturen der Area postrema des Kaninchens und über ihr Verhalten im Pentamethylentetrazol-(„Cardiazol“-)Krampf

Zusammenfassung Die sinusförmigen Kapillaren der Area postrema werden von zwei Basalmembranen umgeben, die einen Raum einschließen, in dem Makrophagen liegen. Verästelte Ausläufer der äußeren Basalmembran dringen tief zwischen die Gliafortsätze ein und bilden ein Membranlabyrinth. Das Kapillarendothel ist fenestriert. Nach Cardiazolkrampf entsteht ein perivaskuläres Oedem, das das Membranlabyrinth öffnet und den perivaskulären Raum vergrößert. Das Endothel zeigt hierbei starke Cytopempsis. Die funktionelle Bedeutung der Strukturen wird diskutiert.

Summary The sinusoid capillaries of the area postrema are surrounded by two basement membranes enclosing a perivascular space, in which macrophages are found. Ramified extensions of the outer basement membrane penetrate deeply between the glial processes forming a labyrinth of basement membranes. The capillary endothelium is fenestrated. After a Cardiazolinduced convulsion a perivascular edema opens the basement membrane labyrinth and the perivascular space is greatly enlarged. The endothelium shows pinocytotic activity. The functional significance of these structures is discussed.

Zur Frage einer intraventrikulären Neurosekretion

Zusammenfassung In der vorderen Wand des IV. Ventrikels und des Canalis centralis gegenüber der Area postrema liegen beim Kaninchen Nervenzellen, deren Fortsätze intraventrikulär mit 6 großen Protoplasmakugeln endigen, die mit Mitochondrien angefüllt sind. Das Perikaryon ist reich an Ergastoplasma, alle Teile der Zelle enthalten leere, 600 bis 1500 Å messende, und ein dichtes Granulum enthaltende, 650–1000 Å große Bläschen. Vermutlich werden Stoffe in den Liquor cerebrospinalis abgegeben. Dem Plasmalemm der Nervenzellen liegen Synapsen an.

Summary In the anterior wall of the IVth ventricle, opposite to the area postrema, nerve cells are found. Their processes end with spheroid swellings extending into the ventricle. These bulbs contain a large number of mitochondria, while ergastoplasm is seen in the perikaryon. The cytoplasm of the whole cell shows vesicles (600–1500 Å) and dense-core vesicles (650–1000 Å) which might be extruded into the liquor cerebrospinalis. The plasmalemma of the perikaryon is in contact with synapses.

A new gene coding for an antigen recognized by autologous cytolytic T lymphocytes on a human renal carcinoma

Previous reports have described antigens that are recognized on human melanoma cells by autologous cytolytic T lymphocytes (CTL). The genes coding for a number of these antigens have been identified. Here we report the cloning of a gene that codes for an antigen recognized by autologous CTL on a human renal carcinoma cell line. This antigen is presented byHLA-B7 and is encoded by a new gene that we have namedRAGE1. No expression ofRAGE1 was found in normal tissues other than retina. RAGE1 expression was found in only one of 57 renal cell carcinoma samples, and also in some sarcomas, infiltrating bladder carcinomas, and melanomas. This represents the first identification of an antigen recognized by autologous CTL on a renal tumor.     

Yeast proteins can activate expression through regulatory sequences of the amdS gene of Aspergillus nidulans

The upstream regulatory region of the amdS gene of Aspergillus nidulans contains a CCAAT sequence known to be important in setting both basal and derepressed levels of expression. We have investigated whether the CCAAT-binding HAP2/3/4 complex of the yeast Saccharomyces cerevisiae can recognise this sequence in an amdS context. Sequences from the 5′ region of amdS were cloned in front of the CYCI-lacZ fusion gene bearing a minimal promoter and transformed into wild-type and hap2 strains of yeast. This study has indicated that amdS sequences are capable of promoting regulated expression of the fusion gene in response to carbon limitation. The yeast HAP2/3/4 complex can recognise the amdS CCAAT sequence and activate expression from this sequence. In addition, the results indicate that other yeast proteins can also regulate expression from the A. nidulans amdS 5′ sequences under carbon-limiting conditions.     

GAPs for rho-related GTPases

Ras-related GTP-binding proteins (GTPases) of the rbo subfamily play important roles in regulating the organization of the actin cytoskeleton. A large number of multifunctional proteins that can stimulate their intrinsic GTPase activity have been identified. Here, we discuss the nature of such GTPase-activating proteins (GAPs) and their potential importance for cell signalling.     

Characterization of pea chloroplastic carbonic anhydrase. Expression inEscherichia coli and site-directed mutagenesis

A cDNA encoding the mature, chloroplast-localized carbonic anhydrase in pea has been expressed inE. coli. The enzyme is fully active and yields of up to 20% of the total soluble protein can be obtained from the bacteria. This expression system was used to monitor the effects of site-directed mutagenesis of seven residues found within conserved regions in the pea carbonic anhydrase amino acid sequence. The effects of these modifications are discussed with respect to the potential of various amino acids to act as sites for zinc coordination or intramolecular proton shuttles.
