The protein acetyltransferase (MTAase) function of glutamine synthetase of Mycobacterium smegmatis was established earlier. In this paper, studies were undertaken to examine MTAase function of recombinant glutamine synthetase (rGlnA1) of Mycobacterium tuberculosis, which showed >80% similarity with M. smegmatis GlnA. The specificity of MTAase to several acyl derivative of coumarins was examined. The results clearly indicated that MTAase exhibited differential specificities to several acyloxycoumarins. Further, MTAase was also found capable of transferring propionyl and butyryl groups from propoxy and butoxy derivatives of 4-methylcoumarin. These observations characterized MTAase in general as a protein acyltransferase. MTAase catalyzed acetylation of GST by 7,8-diacetoxy-4-methylcoumarin (DAMC), a model acetoxy coumarin was confirmed by MALDI-TOF-MS as well as western blot analysis using acetylated lysine polyclonal antibody. In order to validate the active site of rGlnA1 for TAase activity, effect of DAMC and L-methionine-S-sulfoximine (MSO) on GS and TAase activity of rGlnA1 were studied. The results indicated that the active sites of GS and TAase were found different. Acetyl CoA, a universal biological acetyl group donor, was also found to be a substrate for MTAase. These results appropriately characterize glutamine synthetase of Mtb exhibiting transacylase action as a moonlighting protein.  相似文献

Bone marrow architecture is grossly distorted at the diagnosis of ALL and details of the morphological changes that accompany response to Induction chemotherapy have not been reported before. While marrow aspirates are widely used to assess initial response to ALL therapy and provide some indications, we have enumerated marrow components using morphometric analysis of trephine samples with the aim of achieving a greater understanding of changes in bone marrow niches. Morphometric analyses were carried out in the bone marrow trephine samples of 44 children with ALL, using a NanoZoomer HT digital scanner. Diagnostic samples were compared to those of 32 control patients with solid tumors but without marrow involvement. Samples from patients with ALL had significantly increased fibrosis and the area occupied by bony trabeculae was lower than in controls. Cellularity was higher in ALL samples due to leukemic infiltration while the percentage of normal elements such as megakaryocytes, adipocytes, osteoblasts and osteoclasts were all significantly lower. During the course of Induction therapy, there was a decrease in the cellularity of ALL samples at day 15 of therapy with a further decrease at the end of Induction and an increase in the area occupied by adipocytes and the width of sinusoids. Reticulin fibrosis decreased throughout Induction. Megakaryocytes increased, osteoblasts and osteoclasts remained unchanged. No correlation was found between clinical presentation, early response to treatment and morphological changes. Our results provide a morphological background to further studies of bone marrow stroma in ALL.  相似文献

Calamus tenuis (Roxb.), a versatile, dioecious rattan species predominant in northeast India, has emerged as an economical material for light furniture and cottage industries. For the genetic improvement of the species, it is essential to be able to recognize male and female plants at the seedling stage. Screening of genomic DNA with inter-simple sequence repeat (ISSR) primers was used to discover sex-specific polymerase chain reaction (PCR) amplification products. Thirty ISSR primers were screened on female and male C. tenuis plants from five different provinces of Assam, India. A putative female-specific marker was identified. The applicability of ISSR-PCR analysis for development of sex-linked molecular markers in Calamus is discussed.  相似文献

Ureases isolated from leguminous sources, Canavalia ensiformis and Cajanus cajan were immobilized onto gold nanoparticles (nano-ureases). Optimization of the urease immobilization was carried using response surface methodology based on Central Composite Design. Immobilization efficiency of nano-urease from C. ensiformis and C. cajan were found to be 215.10% and 255.92%, respectively. The methodology adopted has deviation of 2.56% and 3.01% with respect to experimental values in case of C. ensiformis and C. cajan, respectively. Nano-urease from C. cajan has broad physico-chemical parameters with pH optimum from 7.1 to 7.3 and temperature optimum from 50 to 70 °C. Nano-urease from C. ensiformis has sharp pH and temperature optima at 7.3 and 70 °C, respectively. Fourier transform infra-red spectroscopy has revealed involvement of groups viz. amino, glycosyl moiety, etc. in urease immobilization onto gold nano-particles. Transmission and scanning electron micrographs revealed that arrangement of urease onto gold nano-particles from C. ensiformis was uniform while it was localized in case of C. cajan. Nano-urease from C. ensiformis has higher specificity and catalysis toward urea as compared to nano-urease from C. cajan. Nano-ureases from both sources are equally stable for 6 months under dried conditions and can be used for 10 washes.  相似文献

A drill core through the Lower Ordovician Tøyen Shale Formation at Lerhamn, NW Scania, southern Sweden, provides important new information for the precise biostratigraphic resolution of the Floian to lower Darriwilian time interval in southern Scandinavia. The Hunnegraptus copiosus, Tetragraptus phyllograptoides, Cymatograptus protobalticus (new), Baltograptus vacillans (new), Baltograptus sp. cf. Baltograptus deflexus (new), Baltograptus minutus (new), Isograptus victoriae, Undulograptus austrodentatus (Arienigraptus zhejiangensis and Undulograptus sinicus Subzones) and the (?)Corymbograptus retroflexus (new) biozones are differentiated in the core and their correlation in Scania is discussed. The Lerhamn drill core provides the most detailed graptolite record of the Floian Stage in Scandinavia. The interval is dominated by a number of species of the genus Baltograptus, endemic to the Atlantic Faunal realm and highly useful for regional biostratigraphic correlation. The biostratigraphic framework is based on endemic and pandemic faunal elements. The mixture of both elements in the drill core allows a more precise inter‐continental correlation of Lower to Middle Ordovician graptolite faunas and may – in the future – provide information as to the climatic history of regions dominated by Baltograptus faunas in the Floian.
