Background
Lutzomyia intermedia and Lutzomyia whitmani (Diptera: Psychodidae) are important and very closely related vector species of cutaneous leishmaniasis in Brazil, which are distinguishable by a few morphological differences. Autoproteolytic cleavage of the inactive acid ceramidase (AC) precursor into the active heterodimer exposes a free cysteine residue, leading us to study whether AC could be regulated by one or more members of the cystatin family. Co-expression of the full-length AC and cystatin SA (cysSA) cDNAs led to significant reduction of AC activity in the transfected cells. Expression of cysSA also inhibited endogenous AC activity in cells and increased ceramide. Conversely, cysSA siRNA expression led to elevated AC activity and reduction in ceramide. The effects of cysSA siRNA expression could be reversed by the addition of recombinant cysSA into the culture media. These results were consistent with detection of a physical interaction between AC and cysSA, assessed by co-immunoprecipitation and nickel-nitrilotriacetic acid affinity chromatography, and further supported by co-localization of the endogenous proteins using confocal microscopy. In vitro kinetic analysis of purified, recombinant AC and cysSA confirmed the transfection results and suggested a non-competitive type of inhibition with a K(i) in the low micromolar range. Processing of the AC precursor into the active form was not affected by cysSA expression, suggesting that it likely inhibits AC by allosteric interference. Computer modeling and expression studies identified several potential inhibitory domains in cysSA, including a small "AC-like" domain (identical to the AC cleavage site, TICT). Small peptides, synthesized with combinations of this and a "cystatin-like" domain (QXVXG), exhibited significant AC inhibition as well. Such peptide-based AC inhibitors could potentially be used to regulate AC activity in cancer cells that are known to overexpress this enzyme alone and in combination with conventional anti-cancer drugs. 相似文献

Background
Lutzomyia longipalpis, the main vector of visceral leishmaniasis in Latin America, is a complex of sibling species. In Brazil, a number of very closely related sibling species have been revealed by the analyses of copulation songs, sex pheromones and molecular markers. However, the level of divergence and gene flow between the sibling species remains unclear. Brazilian populations of this vector can be divided in two main groups: one producing Burst-type songs and the Cembrene-1 pheromone and a second more diverse group producing various Pulse song subtypes and different pheromones.Methodology/Principal Findings
We analyzed 21 nuclear loci in two pairs of Brazilian populations: two sympatric populations from the Sobral locality (1S and 2S) in northeastern Brazil and two allopatric populations from the Lapinha and Pancas localities in southeastern Brazil. Pancas and Sobral 2S are populations of the Burst/Cembrene-1 species while Lapinha and Sobral 1S are two putative incipient species producing the same pheromone and similar Pulse song subtypes. The multilocus analysis strongly suggests the occurrence of gene flow during the divergence between the sibling species, with different levels of introgression between loci. Moreover, this differential introgression is asymmetrical, with estimated gene flow being higher in the direction of the Burst/Cembrene-1 species.Conclusions/Significance
The results indicate that introgressive hybridization has been a crucial phenomenon in shaping the genome of the L. longipalpis complex. This has possible epidemiological implications and is particularly interesting considering the potential for increased introgression caused by man-made environmental changes and the current trend of leishmaniasis urbanization in Brazil. 相似文献127.

A contribution towards resolving the nomenclature of Citharexylum (Verbenaceae): typification of names linked to South American taxa

During ongoing taxonomic studies in the genus Citharexylum (Verbenaceae), twenty names were found in need of typification or typification clarifications. Fourteen names (C. andinum, C. herrerae, C. ilicifolium, C. kunthianum, C. laetum, C. myrianthum var. acuminatum, C. myrianthum var. rigidum, C. pernambucense, C. poeppigii, C. quercifolium, C. solanaceum, C. solanaceum var. macrocalyx, C. surrectum and C. weberbaueri) are here lectotypified, and epitypes are selected for three names (C. herrerae, C. kunthianum and C. pernambucense). Neotypes are designated for the names C. berteroi, C. broadwayi and C. coriaceum. Furthermore, updates and supporting information for three names already typified by Harold Moldenke (C. macrochlamys, C. punctatum and C. subthyrsoideum) are provided. 相似文献

The Aloysia gratissima complex is a group of 22 accepted, specific and infraspecific taxa, the boundaries of which are controversial. Various different authors have chosen different characters as criteria for delimiting taxa in this complex, which has been reflected in the variable number of accepted species. To resolve the taxonomy of this group, a modified population aggregation analysis, based on the phylogenetic species concept, was employed. As a result, the Aloysia gratissima complex is here circumscribed as compromising only four species, A. decipiens, A. gratissima s.s., A. oblanceolata and A. schulziana. These species are sustained by a different combination of four characters, of the 16 qualitative attributes analysed. A new approach based on cluster analysis is proposed here delimiting varieties. As a consequence two varieties, supported by significant differences in quantitative characters, are recognized here: A. gratissima var. gratissima and A. gratissima var. sellowii. A complete taxonomic revision of the accepted taxa is presented and six new synonyms are proposed.
