Wolbachia are symbiotic intracellular bacteria, which are classified as reproductive parasites. Although generally facultative, Wolbachia is necessary for Asobara tabida (Hymenoptera), because aposymbiotic females do not produce any offspring. Interestingly, the ovarian phenotype of aposymbiotic females is variable: some females do not produce any eggs, whereas others do produce some eggs, but these are aborted. Here, we show that the ovarian phenotype of aposymbiotic females is highly polymorphic within populations, although dependence remains complete in both cases. We also identified some lines in which aposymbiotic females were able to produce a very few viable offspring, further extending the range of variation observed. These results suggest that various factors actively maintain polymorphism. We demonstrated that Wolbachia is necessary to trigger oogenetic processes, but that the ovarian phenotype was determined by the host only. Phenotypic variation was also correlated with the differential expression of genes controlling iron homeostasis and oxidative stress, which are potentially involved in the evolution of dependence. This suggests that variation in the ovarian phenotype could reflect selection for different levels of compensatory mechanisms in response to Wolbachia infection, and that polymorphism is maintained through selection on different antagonist traits influenced by oxidative stress.  相似文献

Abstract

The pollen morphology of the Iberian taxa of the genus Xanthium s.l. and of the non-Iberian species X. ambrosioides was studied to help to identify these poisonous, allergenic and invasive taxa and clarify their taxonomic relationships. In addition, the question of the existence and structure of a mesoaperture in the apertural system of Xanthium was also considered. The pollen grains of sections Acanthoxanthium and Xanthium clearly differ by their characters, which supports the separation of both taxa and allows for their distinction based on pollen morphology. It is suggested that the morphological and palynological differences between Xanthium and Acanthoxanthium, together with the lack of hybridisation between the two sections and the monophyly of Acanthoxanthium, could lead to the elevation of the latter to the generic status. The two species of section Xanthium present in the Iberian Peninsula (X. orientale and X. strumarium) and of section Acanthoxanthium are well separated by their palynological features. Likewise, the Iberian subspecies of Xanthium, X. orientale subsp. orientale and X. orientale subsp. italicum are palynologically distinguishable. However, no significant difference could be found between the pollen of X. strumarium subsp. strumarium and of X. strumarium subsp. brasilicum. The pollen data do not support the hypotheses of a hybrid origin for X. strumarium subsp. brasilicum. A mesoaperture intersecting the foot layer and the upper part of the endexine is present in both sections, which is congruent with the results already found for other Asteroideae.  相似文献

Varicella zoster virus (VZV) is a significant cause of morbidity and mortality following umbilical cord blood transplantation (UCBT). For this reason, antiherpetic prophylaxis is administrated systematically to pediatric UCBT recipients to prevent complications associated with VZV infection, but there is no strong, evidence based consensus that defines its optimal duration. Because T cell mediated immunity is responsible for the control of VZV infection, assessing the reconstitution of VZV specific T cell responses following UCBT could provide indications as to whether prophylaxis should be maintained or can be discontinued. To this end, a VZV specific gamma interferon (IFN-γ) enzyme-linked immunospot (ELISpot) assay was developed to characterize IFN-γ production by T lymphocytes in response to in vitro stimulation with irradiated live attenuated VZV vaccine. This assay provides a rapid, reproducible and sensitive measurement of VZV specific cell mediated immunity suitable for monitoring the reconstitution of VZV specific immunity in a clinical setting and assessing immune responsiveness to VZV antigens.    相似文献

Refsum disease is an autosomal recessive disorder of peroxisomal metabolism biochemically characterized by highly elevated concentrations of phytanic acid (Phyt) in a variety of tissues including the cerebellum. Reduction of plasma Phyt levels by dietary restriction intake ameliorates ataxia, a common clinical manifestation of this disorder, suggesting a neurotoxic role for this branched-chain fatty acid. Therefore, considering that the underlying mechanisms of cerebellum damage in Refsum disease are poorly known, in the present study we tested the effects of Phyt on important parameters of bioenergetics, such as the activities of the respiratory chain complexes I to IV, creatine kinase and Na+, K+- ATPase in cerebellum preparations from young rats. The activities of complexes I, II, I–III and II–III and Na+, K+- ATPase were markedly inhibited (65–85 %) in a dose-dependent manner by Phyt. In contrast, creatine kinase and complex IV activities were not altered by this fatty acid. Therefore, it is presumed that impairment of the electron flow through the respiratory chain and inhibition of Na+, K+- ATPase that is crucial for synaptic function may be involved in the pathophysiology of the cerebellar abnormalities manifested as ataxia in Refsum disease and in other peroxisomal disorders in which brain Phyt accumulates.  相似文献
