目的:探讨钒配合物LMc对拓扑异构酶Ⅰ、Ⅱ(Topo-Ⅰ、Topo-Ⅱ)的影响及其抗肿瘤活性。方法:采用DNA松弛实验观察LMC对Topo-Ⅰ、活性的影响并探讨其相关分子作用机制;采用MTT法、流式细胞术在细胞水平观察了IMC的抗肿瘤作用。结果:LMC可明显抑制Topo-Ⅰ活性,对Topo-Ⅱ无明显抑制作用,对多种肿瘤细胞株A549、Hela、BEL-7402具有明显抑制生长的作用,且可将细胞阻断在G2/M期,而对正常细胞株L-02生长无明显影响。结论:钒配合物LMC具有抑制Topo-Ⅰ活性而发挥抗肿瘤的作用。 相似文献

The hydraulic conductivity of plant roots (Lpr) is determined in large part by the activity of aquaporins. Mechanisms occurring at the post‐translational level, in particular phosphorylation of aquaporins of the plasma membrane intrinsic protein 2 (PIP2) subfamily, are thought to be of critical importance for regulating root water transport. However, knowledge of protein kinases and phosphatases acting on aquaporin function is still scarce. In the present work, we investigated the Lpr of knockout Arabidopsis plants for four Ca2+‐dependent protein kinases. cpk7 plants showed a 30% increase in Lpr because of a higher aquaporin activity. A quantitative proteomic analysis of wild‐type and cpk7 plants revealed that PIP gene expression and PIP protein quantity were not correlated and that CPK7 has no effect on PIP2 phosphorylation. In contrast, CPK7 exerts a negative control on the cellular abundance of PIP1s, which likely accounts for the higher Lpr of cpk7. In addition, this study revealed that the cellular amount of a few additional proteins including membrane transporters is controlled by CPK7. The overall work provides evidence for CPK7‐dependent stability of specific membrane proteins. 相似文献

The voltage-gated proton channel exists as a dimer, although each protomer has a separate conduction pathway, and when forced to exist as a monomer, most major functions are retained. However, the proton channel protomers appear to interact during gating. Proton channel dimerization is thought to result mainly from coiled-coil interaction of the intracellular C-termini. Several types of evidence are discussed that suggest that the dimer conformation may not be static, but is dynamic and can sample different orientations. Zn2+ appears to link the protomers in an orientation from which the channel(s) cannot open. A tandem WT-WT dimer exhibits signs of cooperative gating, indicating that despite the abnormal linkage, the correct orientation for opening can occur. We propose that C-terminal interaction functions mainly to tether the protomers together. Ethanol is a known neuromodulatory agent with reported actions at a range of neurotransmitter receptors. Here, we measured the effect of alcohol on metabolism of [3‐13C]pyruvate in the adult Guinea pig brain cortical tissue slice and compared the outcomes to those from a library of ligands active in the GABAergic system as well as studying the metabolic fate of [1,2‐13C]ethanol. Analyses of metabolic profile clusters suggest that the significant reductions in metabolism induced by ethanol (10, 30 and 60 mM) are via action at neurotransmitter receptors, particularly α4β3δ receptors, whereas very low concentrations of ethanol may produce metabolic responses owing to release of GABA via GABA transporter 1 (GAT1) and the subsequent interaction of this GABA with local α5‐ or α1‐containing GABA(A)R. There was no measureable metabolism of [1,2‐13C]ethanol with no significant incorporation of 13C from [1,2‐13C]ethanol into any measured metabolite above natural abundance, although there were measurable effects on total metabolite sizes similar to those seen with unlabelled ethanol.

Molecular phylogenetic evidence that the phylum Haplosporidia has an alveolate ancestry 总被引:1,自引:0,他引:1
The phylogenetic position of the phylum Haplosporidia among other protists
was investigated with the complete 16S-like rRNA gene sequences from two
species in the phylum: Haplosporidium nelsoni, a parasite of oysters, and
Minchinia teredinis, a parasite of shipworms. Because the lack of obvious
morphological homologies with other protists hampered decisions regarding
taxonomic composition for sequence alignment and phylogenetic analysis, the
complete sequences for these two haplosporidians were directed as search
queries to the blast/ncbi.nlm.nih.gov electronic mail server. The results
of this heuristic similarity search provided a basis for constructing a
preliminary higher-taxonomic-level analysis comparing the haplosporidians
with species from the slime molds, fungi, algae, amoebae, ciliates,
dinoflagellates, and apicomplexans. Maximum parsimony yielded equivocal
results, whereas transversionally weighted parsimony suggested an affinity
with the alveolates (i.e., the ciliates, dinoflagellates, and
apicomplexans). Multiple alignment of the two haplosporidian sequences
against 17 taxa in a secondary analysis focusing on the alveolates and
subsequent parsimony analysis placed the phylum Haplosporidia as a
monophyletic group within the Alveolata and as a taxon of equal rank with
the other three alveolate phyla. The precise placement within the Alveolata
was sensitive to weighting.

A genetic analysis was performed to study the frequency of recombination for intervals across the Brassica S locus region. No recombination was observed between the S locus glycoprotein gene and the S receptor kinase gene in the segregating populations that we analyzed. However, a number of recombination breakpoints in regions flanking these genes were identified, allowing the construction of an integrated genetic and physical map of the genomic region encompassing one S haplotype. We identified, based on the pollination phenotype of plants homozygous for recombinant S haplotypes, a 50-kb region that encompasses all specificity functions in the S haplotype that we analyzed. Mechanisms that might operate to preserve the tight linkage of self-incompatibility specificity genes within the S locus complex are discussed in light of the relatively uniform recombination frequencies that we observed across the S locus region and of the structural heteromorphisms that characterize different S haplotypes. 相似文献

A cDNA library of rice (Oryza sativa L.) has been constructed from anthers at an early stage of pollen development. By differential screening of the library, we have isolated cDNAs of two genes, designated as Osc4 and Osc6, that are abundantly expressed in anthers containing tetrads and uninucleate microspores, but are not expressed in leaves or roots. Expression of Osc4 is absent in mature anthers, while Osc6 is present although the expression decays during pollen maturation. A comparison of the nucleotide and deduced amino acid sequences with those in data banks has not shown significant homology to known molecules. 相似文献
