Myricetin (MYR) is a bioactive secondary metabolite found in plants that is recognized for its nutraceutical value and is an essential constituent of various foods and beverages. It is reported to exhibit a plethora of activities, including antioxidant, antimicrobial, antidiabetic, anticancer, and anti-inflammatory. Alpha-2-macroglobulin (α2M) is a major plasma anti-proteinase that can inhibit proteinases of both human and non-human origin, regardless of their specificity and catalytic mechanism. Here, we explored the interaction of MYR-α2M using various biochemical and biophysical techniques. It was found that the interaction of MYR brings subtle change in its anti-proteolytic potential and thereby alters its structure and function, as can be seen from absorbance and fluorescence spectroscopy. UV spectroscopy of α2M in presence of MYR indicated the occurrence of hyperchromism, suggesting complex formation. Fluorescence spectroscopy reveals that MYR reduces the fluorescence intensity of native α2M with a shift in the wavelength maxima. At 318.15 K, MYR binds to α2M with a binding constant of 2.4 × 103 M−1, which indicates significant binding. The ΔG value was found to be − 7.56 kcal mol−1 at 298.15 K, suggesting the interaction to be spontaneous and thermodynamically favorable. The secondary structure of α2M does not involve any major change as was confirmed by CD analysis. The molecular docking indicates that Asp-146, Ser-172, Glu-174, and Tyr-180 were the key residues involved in α2M-MYR complex formation. This study contributes to our understanding of the function and mechanism of protein and flavonoid binding by providing a molecular basis of the interaction between MYR and α2M. 相似文献

Drought is the major detrimental environmental factor for wheat (Triticum aestivum L.) production. The exploration of genetic patterns underlying drought tolerance is of great significance. Here we report the gene actions controlling the phenological traits using the line × tester model studying 27 crosses and 12 parents under normal irrigation and drought conditions. The results interpreted via multiple analysis (mean performance, correlations, principal component, genetic analysis, heterotic and heterobeltiotic potential) disclosed highly significant differences among germplasm. The phenological waxiness traits (glume, boom, and sheath) were strongly interlinked. Flag leaf area exhibits a positive association with peduncle and spike length under drought. The growing degree days (heat-units) greatly influence spikelets and grains per spike, however, the grain yield/plant was significantly reduced (17.44 g to 13.25 g) under drought. The principal components based on eigenvalue indicated significant PCs (first-seven) accounted for 79.9% and 73.9% of total variability under normal irrigation and drought, respectively. The investigated yield traits showed complex genetic behaviour. The genetic advance confronted a moderate to high heritability for spikelets/spike and grain yield/plant. The traits conditioned by dominant genetic effects in normal irrigation were inversely controlled by additive genetic effects under drought and vice versa. The magnitude of dominance effects for phenological and yield traits, i.e., leaf twist, auricle hairiness, grain yield/plant, spikelets, and grains/spike suggests that selection by the pedigree method is appropriate for improving these traits under normal irrigation conditions and could serve as an indirect selection index for improving yield-oriented traits in wheat populations for drought tolerance. However, the phenotypic selection could be more than effective for traits conditioned by additive genetic effects under drought. We suggest five significant cross combinations based on heterotic and heterobeltiotic potential of wheat genotypes for improved yield and enhanced biological production of wheat in advanced generations under drought. 相似文献

Calcium–calmodulin-dependent protein kinase IV (CAMK4) plays significant role in the regulation of calcium-dependent gene expression, and thus, it is involved in varieties of cellular functions such as cell signaling and neuronal survival. On the other hand, curcumin, a naturally occurring yellow bioactive component of turmeric possesses wide spectrum of biological actions, and it is widely used to treat atherosclerosis, diabetes, cancer, and inflammation. It also acts as an antioxidant. Here, we studied the interaction of curcumin with human CAMK4 at pH 7.4 using molecular docking, molecular dynamics (MD) simulations, fluorescence binding, and surface plasmon resonance (SPR) methods. We performed MD simulations for both neutral and anionic forms of CAMK4-curcumin complexes for a reasonably long time (150 ns) to see the overall stability of the protein–ligand complex. Molecular docking studies revealed that the curcumin binds in the large hydrophobic cavity of kinase domain of CAMK4 through several hydrophobic and hydrogen-bonded interactions. Additionally, MD simulations studies contributed in understanding the stability of protein–ligand complex system in aqueous solution and conformational changes in the CAMK4 upon binding of curcumin. A significant increase in the fluorescence intensity at 495 nm was observed (λexc = 425 nm), suggesting a strong interaction of curcumin to the CAMK4. A high binding affinity (KD = 3.7 × 10?8 ± .03 M) of curcumin for the CAMK4 was measured by SPR further indicating curcumin as a potential ligand for the CAMK4. This study will provide insights into designing a new inspired curcumin derivatives as therapeutic agents against many life-threatening diseases. 相似文献

Biochar application to agricultural soils is rapidly emerging as a new management strategy for its potential role in carbon sequestration, soil quality improvements, and plant growth promotion. The aim of our study was to investigate the effects of biochars derived from white clover residues and poultry manure on soil quality characteristics, growth and N accumulation in maize (Zea mays L.) and wheat (Triticum aestivum L.) grown in a loam soil under greenhouse conditions. Treatments comprised of: untreated control; mineral N fertilizer (urea N, UN) at the rate of 200, and 100 mg N kg-1, white clover residues biochar (WCRB), poultry manure biochar (PMB) at 30 Mg ha–1, and the possible combinations of WCRB+PMB (50:50), UN+WCRB (50:50), UN+PMB (50:50), and UN+WCRB+PMB (50:25:25). The treatments were arranged in a completely randomized design with three replications. Results indicated a significant increase in the growth and biomass production of maize and wheat supplemented with biochars alone or mixed with N fertilizer. Biochars treatments showed varying impact on plant growth depended upon the type of the biochar, and in general plant growth under PMB was significantly higher than that recorded under WCRB. The growth characteristics in the combined treatments (half biochar+half N) were either higher or equivalent to that recorded under full fertilizer N treatment (N200). The biochar treatments WCRB, PMB, and WCRB+PMB (50:50) increased maize shoot N by 18, 26 and 21%, respectively compared to the control while wheat shoot N did not show positive response. The N-uptake by maize treated with WCRB, PMB, and WCRB+PMB (50:50) was 54, 116, and 90 mg g-1 compared to the 33 mg g-1 in the control while the N-uptake by wheat was 41, 60, and 53 mg g-1 compared to 24 mg g-1 in the control. The mixed treatments (half biochar+half N) increased N-uptake by 2.3folds in maize and 1.7 to 2.5folds in wheat compared to the N100 showing increasing effect of biochar on N use efficiency of applied N. Post-harvest soil analysis indicated a significant increase in pH, organic matter, organic C, total N, C:N, and porosity (% pore space) by the added biochars while bulk density (BD) was significantly decreased. The organic matter content in the soil amended with biochars ranged between 19.5 and 23.2 g kg-1 compared to 11.7 and 10.2 g kg-1 in the control and N fertilizer treatments while the BD of biochars amended soils (WCRB, PMB, and WCRB+PMB) was 1.07, 1.17, and 1.11 g cm-3 compared to 1.28 g cm-1 in the control. In summary, the results of present study highlight the agronomic benefits of biochars in improving the quality of the soil, and promoting growth, yield and N accumulation of both maize and wheat with a consequent benefit to agriculture. 相似文献

Design and implementation of robust network modules is essential for construction of complex biological systems through hierarchical assembly of ‘parts’ and ‘devices’. The robustness of gene regulatory networks (GRNs) is ascribed chiefly to the underlying topology. The automatic designing capability of GRN topology that can exhibit robust behavior can dramatically change the current practice in synthetic biology. A recent study shows that Darwinian evolution can gradually develop higher topological robustness. Subsequently, this work presents an evolutionary algorithm that simulates natural evolution in silico, for identifying network topologies that are robust to perturbations. We present a Monte Carlo based method for quantifying topological robustness and designed a fitness approximation approach for efficient calculation of topological robustness which is computationally very intensive. The proposed framework was verified using two classic GRN behaviors: oscillation and bistability, although the framework is generalized for evolving other types of responses. The algorithm identified robust GRN architectures which were verified using different analysis and comparison. Analysis of the results also shed light on the relationship among robustness, cooperativity and complexity. This study also shows that nature has already evolved very robust architectures for its crucial systems; hence simulation of this natural process can be very valuable for designing robust biological systems. 相似文献

BackgroundαA-crystallin plays an important role in eye lens development. Its N-terminal domain is implicated in several important biological functions. Mutations in certain conserved arginine residues in the N-terminal region of αA-crystallin lead to cataract with characteristic cytoplasmic/nuclear aggregation of the mutant protein. In this study, we attempt to gain mechanistic insights into the congenital cataract caused by the R54C mutation in human αA-crystallin.MethodsWe used several spectroscopic techniques to investigate the structure and function of the wild-type and R54CαA-crystallin. Immunoprecipitation, chromatin-enrichment followed by western blotting, immunofluorescence and cell-viability assay were performed to study the interaction partners, chromatin-association, stress-like response and cell-death caused by the mutant.ResultsAlthough R54CαA-crystallin exhibited slight changes in quaternary structure, its chaperone-like activity was comparable to that of wild-type. When expressed in lens epithelial cells, R54CαA-crystallin exhibited a speckled appearance in the nucleus rather than cytoplasmic localization. R54CαA-crystallin triggered a stress-like response, resulting in nuclear translocation of αB-crystallin, disassembly of cytoskeletal elements and activation of caspase 3, leading to apoptosis. Analysis of the “interactome” revealed an increase in interaction of the mutant protein with nucleosomal histones, and its association with chromatin.ConclusionsThe study shows that alteration of “interactome” and nucleosomal association, rather than loss of chaperone-like activity, is the molecular basis of cataract caused by the R54C mutation in αA-crystallin.General significanceThe study provides a novel mechanism of cataract caused by a mutant of αA-crystallin, and sheds light on the possible mechanism of stress and cell death caused by such nuclear inclusions. 相似文献

Glyphosate is one of the most widely used herbicides in cereal-growing regions worldwide. In the present work, the protein expression profile of rice leaves exposed to glyphosate was analyzed in order to investigate the alternative effects of glyphosate on plants. Two-week-old rice leaves were subjected to glyphosate or a reactive oxygen species (ROS) inducing herbicide paraquat, and total soluble proteins were extracted and analyzed by two-dimensional gel electrophoresis (2-DE) coupled with matrix-assisted laser desorption/ionization-time of flight (MALDI-TOF) mass spectrometry (MS) analysis. A total of 25 differentially expressed proteins were identified from the glyphosate treated sample, wherein 18 proteins were up-regulated and 7 proteins were down-regulated. These proteins had shown a parallel expression pattern in response to paraquat. Results from the 2-DE analysis, combined with immunoblotting, clearly revealed that ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) large subunit was significantly decreased by the treatment of both herbicides. An increased accumulation of antioxidant enzymes including ascorbate peroxidase, glutathione S-transferase, thioredoxin h-type, nucleoside diphosphate kinase 1, peroxiredoxin and a superoxide dismutase [Cu–Zn] chloroplast precursor in the glyphosate-treated sample suggests that a glyphosate treatment possibly generates oxidative stress in plants. Moreover, a gene expression analysis of five antioxidant enzymes by Northern blot confirmed their mRNA levels in the rice leaves. A histo-cytochemical investigation with DAB (3,3-diaminobenzidine) to localize H2O2 and increases of the thiobarbituric acid reactive substances (TBARS) concentration revealed that the glyphosate application generates ROS, which resulted in the peroxidation and destruction of lipids in the rice leaves. 相似文献

Alpha macroglobulins are large glycoproteins which are present in the body fluids of both invertebrates and vertebrates. Alpha‐2‐macroglobulin (α2M), a key member of alpha macroglobulin superfamily, is a high‐molecular weight homotetrameric glycoprotein. α2M has many diversified and complex functions, but it is primarily known by its ability to inhibit a broad spectrum of proteases without the direct blockage of the protease active site. α2M is also known to be involved in the regulation, transport, and a host of other functions. For example, apart from inhibiting proteinases, it regulates binding of transferrin to its surface receptor, binds defensin and myelin basic protein, etc., binds several important cytokines, including basic fibroblast growth factor (bFGF), platelet‐derived growth factor (PDGF), nerve growth factor (NGF), interleukin‐1β (IL‐1β), and interleukin‐6 (IL‐6), and modify their biological activity. α2M also binds a number of hormones and regulates their activity. α2M is said to protect the body against various infections, and hence, can be used as a biomarker for the diagnosis and prognosis of a number of diseases. However, this multipurpose antiproteinse is not “fail safe” and could be damaged by reactive species generated endogenously or exogenously, leading to various pathophysiological conditions. J. Cell.
