Helichrysum (family Asteraceae) is a large, heterogeneous and polyphyletic genus. Most of the Helichrysum species have ornamental and medicinal values. Helicrysum leucocephalum Boiss., has wide geographical distribution in Iran and forms several local populations. The present investigation was performed to study inter populations, genetic, cytogenetic and morphological diversity of this medicinal species. The AMOVA test showed significant genetic difference among the studied populations but the Mantel test did not show correlation between the genetic distance and geographical distance of these populations. STRUCTURE and reticulation analyses showed some degree of genetic admixture and gene exchange among the studied populations. These populations had different chromosome numbers and also differed significantly in the size of their chromosomes.  相似文献

Azo dyes are the largest class of synthetic dyes, which are widely used in the textile industry. The amount of dyestuff does not bind to the fibers and is lost in wastewater during textile processing. The discharge of colored effluents into the environment is not only aesthetically unpleasing. Moreover, dyes and their break-down products cause toxic effects and they affect photosynthetic activity of aquatic systems by reducing light penetration. A number of microorganisms belonging to different taxonomic groups of bacteria, algae, fungi and yeast have been reported for their ability to decolorize azo dyes. In the literature the ability to decolorize azo dyes by yeasts, compared to bacterial and fungal species, has been studied in a few reports. Within this review, an attempt is made to elucidate some basic biological aspects associated with the azo dye degradation by yeasts and enzymes involved that are responsible for degradation process.  相似文献

Dendritic cell (DC)-based vaccination is a promising approach to enhance anti-tumor immunity that could be considered for acute myeloid leukemia (AML) patients with high-risk of relapse. Our purpose was to study the efficiency and to optimize the immunogenicity of a DC-based vaccine in a preclinical AML murine model. In this report, C57BL6 mice were vaccinated with DC pulsed with peptides eluted (EP) from the syngeneic C1498 myelomonocytic leukemic cell line in a prophylactic setting. In this model, a natural antileukemic immunity mediated by NK cells was observed in the control unloaded DC-vaccinated group. On the other hand, we showed that the cytotoxic antileukemic immune response induced by vaccination with eluted peptides pulsed-DC (DC/EP), in vitro and in vivo, was mainly mediated by CD4+ T cells. Treatment with anti-CD25 antibody to deplete CD4+ CD25+ regulatory T cells before DC-vaccination dramatically improved the antileukemic immune response induced by immunization, and allowed the development of long-lasting immune responses that were tumor protective after a re-challenge with leukemic cells. Our results suggest that this approach could be successful against weakly immunogenic tumors such as AML, and could be translated in human.  相似文献

Gradient distribution of bio‐adhesive proteins can regulate multiple cellular processes, including adhesion, growth, and migration. The ability to control the cell function by changing the surface density of immobilized ligands has become increasingly important in design of implantable medical devices and tissue regenerating scaffolds. Recent techniques in fabrication of substrates with controlled surface properties allow the examination of cell sensitivity to a wide range of adhesion gradients. Understanding the mechanisms by which cells sense and respond to these directional cues warrants a quantitative assessment of macroscopic cellular response to the surface gradients, supported by predictive theoretical models. This article presents a theoretical basis to examine the effect of ligand gradients on cellular adhesion, using an equilibrium thermodynamic model. The model facilitates a systematic investigation of the complex interplay of cell–substrate specific adhesions, non‐specific repulsions, and membrane elasticity. This purely mechanistic model predicts a biphasic dependence between the extent of cell spreading and its position across the gradient substrate. Biotechnol. Energy drink consumption has been anecdotally linked to the development of adverse cardiovascular effects in consumers, although clinical trials to support this link are lacking. The effects of Red Bull® energy drink on cardiovascular and neurologic functions were examined in college-aged students enrolled at Winona State University. In a double-blind experiment where normal calorie and low calorie Red Bull® were compared to normal and low calorie placebos, no changes in overall cardiovascular function nor blood glucose (mg/dL) were recorded in any participant (n = 68) throughout a 2-h test period. However, in the second experiment, nine male and twelve female participants subjected to a cold pressor test (CPT) before and after Red Bull® consumption showed a significant increase in blood sugar levels pre- and post Red Bull® consumption. There was a significant increase in diastolic blood pressure of the male volunteers immediately after submersion of the hand in the 5°C water for the CPT. Under the influence of Red Bull®, the increase in diastolic pressure for the male participants during the CPT was negated. There were no significant changes in the blood pressure of the female participants for the CPT with or without Red Bull®. Finally, the CPT was used to evaluate pain threshold and pain tolerance before and after Red Bull® consumption. Red Bull® consumption was associated with a significant increase in pain tolerance in all participants. These findings suggest that Red Bull® consumption ameliorates changes in blood pressure during stressful experiences and increases the participants’ pain tolerance.  相似文献

Introduction

As cancer cells are affected by many factors in their microenvironment, a major challenge is to isolate the effect of a specific factor on cancer stem cells (CSCs) while keeping other factors unchanged. We have developed a synthetic inert 3D polyethylene glycol diacrylate (PEGDA) gel culture system as a unique tool to study the effect of microenvironmental factors on CSCs response. We have reported that CSCs formed in the inert PEGDA gel by encapsulation of breast cancer cells maintain their stemness within a certain range of gel stiffness. The objective was to investigate the effect of CD44 binding peptide (CD44BP) conjugated to the gel on the maintenance of breast CSCs.

Methods

4T1 or MCF7 breast cancer cells were encapsulated in PEGDA gel with CD44BP conjugation. Control groups included dissolved CD44BP and the gel with mutant CD44BP conjugation. Tumorsphere size and density, and expression of CSC markers were determined after 9 days. For in vivo, cell encapsulated gels were inoculated in syngeneic Balb/C mice and tumor formation was determined after 4 weeks. Effect of CD44BP conjugation on breast CSC maintenance was compared with integrin binding RGD peptide (IBP) and fibronectin-derived heparin binding peptide (FHBP).

Results

Conjugation of CD44BP to the gel inhibited breast tumorsphere formation in vitro and in vivo. The ability of the encapsulated cells to form tumorspheres in the peptide-conjugated gels correlated with the expression of CSC markers. Tumorsphere formation in vitro was enhanced by FHBP while it was abolished by IBP.

Conclusion

CD44BP and IBP conjugated to the gel abolished tumorsphere formation by encapsulated 4T1 cells while FHBP enhanced tumorsphere formation compared to cells in the gel without peptide. The PEGDA hydrogel culture system provides a novel tool to investigate the individual effect of factors in the microenvironment on CSC maintenance without interference of other factors.  相似文献

Halophile proteins can tolerate high salt concentrations. Understanding halophilicity features is the first step toward engineering halostable crops. To this end, we examined protein features contributing to the halo-toleration of halophilic organisms. We compared more than 850 features for halophilic and non-halophilic proteins with various screening, clustering, decision tree, and generalized rule induction models to search for patterns that code for halo-toleration. Up to 251 protein attributes selected by various attribute weighting algorithms as important features contribute to halo-stability; from them 14 attributes selected by 90% of models and the count of hydrogen gained the highest value (1.0) in 70% of attribute weighting models, showing the importance of this attribute in feature selection modeling. The other attributes mostly were the frequencies of di-peptides. No changes were found in the numbers of groups when K-Means and TwoStep clustering modeling were performed on datasets with or without feature selection filtering. Although the depths of induced trees were not high, the accuracies of trees were higher than 94% and the frequency of hydrophobic residues pointed as the most important feature to build trees. The performance evaluation of decision tree models had the same values and the best correctness percentage recorded with the Exhaustive CHAID and CHAID models. We did not find any significant difference in the percent of correctness, performance evaluation, and mean correctness of various decision tree models with or without feature selection. For the first time, we analyzed the performance of different screening, clustering, and decision tree algorithms for discriminating halophilic and non-halophilic proteins and the results showed that amino acid composition can be used to discriminate between halo-tolerant and halo-sensitive proteins.  相似文献

During recent years, hazel cell suspension culture has been significantly considered as a new important source of paclitaxel. Artificial polyploidy alters different characters in plants which results in amplifying the secondary metabolites in medicinal plants. In this paper, the effects of tetraploidy induction on paclitaxel content and gene expression in hazel cell suspension culture were investigated. Various concentrations of colchicine and duration of exposure in solid and liquid media were examined and the ploidy level of cells was determined using flow cytometric analysis. The tetraploid cells were obtained from 0.2% colchicine in the exposure time of 5 and 6 days in solid medium and 0.3% colchicine in the exposure time of 3 and 4 days in liquid medium. Tetraploid cells were employed to prepare cell suspension. 3 µM of phenylalanine and 0.05 mM of vanadyl sulfate were added to both tetraploid and diploid (control) suspension to elicit paclitaxel induction. High performance liquid chromatography analysis demonstrated that the tetraploid cell suspensions produced paclitaxel of about [9.88 µg g?1 (DW)] 1.7-fold compared with diploid cells [5.74 µg g?1 (DW)]. The application of phenylalanine and vanadyl sulfate increased the concentration of paclitaxel in both diploid and tetraploid cells. Moreover, qRT-PCR analysis showed that the expression of GGPPS gene was significantly increased and PAL gene expression was altered after tetraploidization.  相似文献
