Surface charge of synaptosomes was studied with cationized ferritin (CF) binding at pH between 4 and 7. The characteristic of binding resembles that of the electrophoretic mobility and suggests an electrostatic interaction between CF and membranes. When sialic acid was partially removed from synaptosomes, a reduction in binding was detected. This suggests that sialic acid is one determinant of surface charge of synaptosomes. The present method differs from other CF studies in that the amount of bound CF was measured chemically instead of by electron microscopy. This method could be useful in other cell systems.  相似文献

Gonococci (strain BS3) passaged three times and harvested directly from plastic chambers implanted subcutaneously in guinea pigs were compared with the parent strain (BS) grown in vitro. The strain grown in vivo produced smaller colonies than that grown in vitro and when examined directly in chamber fluid was sometimes not pilated. It was more resistant to the bactericidal action of human serum and more infective for guinea-pig chambers. In gel diffusion, extracts of the organisms adapted in vivo and cultured once on agar appeared to contain one or two antigens that were different from those in extracts of the in vitro grown organisms; and on polyacrylamide gels, electrophoresis of similar extracts showed one or more protein components for strain BS3 which were not seen for strain BS. Gonococci grown in guinea-pig subcutaneous chambers appear to be suitable for studies on the determinants of gonoccal pathogenicity.  相似文献

Summary Earlier studies using Golgi silver impregnations from the labellar sensilla of adult Drosophila melanogaster revealed seven types of sensory axons projecting into the suboesophageal ganglion of the brain. These sensory terminals were designated as coiled fibres (type-I), shrubby fibres (type-II), ipsilateral ventral fibres (type-III), ipsilateral dorsal fibres (type-IV), contralateral ventral fibres (type-V), contralateral dorsal fibres (type-VI), and central fibres (type-VII). The present study identifies the projections of sensory neurons present in a single labellar taste-sensillum, using the neuronal marker horseradish peroxidase (HRP). Although the taste sensillum in question has five neurons, in a given experiment only one or at the most two neurons are labelled. The type of neuron labelled was usually specific to the stimulant solute (sucrose, sodium chloride or potassium chloride) present in the HRP solution. Although type-II fibres get labelled most of the time, irrespective of the stimulant present in HRP solution, type-IV fibres are labelled when attractants (0.1 M sucrose or 0.1 M sodium chloride) are used as stimulants in HRP solution. Type-VI fibres are labelled when the stimulant is 0.1 M potassium chloride, a repellent. HRP dissolved in distilled water revealed type-I coiled fibres. Besides revealing projections of sensillar neurons to the brain the present technique also inferred their possible function. Incubation of whole-brain tissue with 0.04% 3,3-diaminobenzidine tetrahydrochloride in presence of 0.06% hydrogen peroxide suggested that the glomerular organization is also present in the taste-sensory region as it is in olfactory neuropile.  相似文献

Protoplasts were isolated from cotyledon, hypocotyl and mesophyll cells of Corchorus capsularis L., a major fibre crop, by one step enzyme digestion method. They were further cultured successfully on modified KM-8p (Kao and Michayluk, 1975) medium to form microcalli. The required cultural conditions could be used to achieve 34% to 78% plating efficiency, depending upon the source of protoplasts. Hypocotyl protoplasts gave the highest plating efficiency. On transfer to regeneration medium, somatic embryos developed at high frequency. The endogenous substrate proteins of rat cardiac protein kinase C type I, II, and III isozymic forms were studied in rat cardiac sarcolemma. The 19-, 21-, 29-, 35-, and 95-kDa proteins were phosphorylated by both types II and III, but not type I. The extent of phosphorylation by individual protein kinase C isozymic forms was additive and equal to the extent of phosphorylation observed when a mixture of isozymic forms was employed. The extent of phosphorylation of the 21-kDa protein by type III was much higher than that by type II. These results suggest that the protein kinase C isozymes have preferences for specific endogenous substrate proteins. The phosphorylation of these endogenous substrate proteins by protein kinase C isozymes probably plays a role in cardiac cell functions.  相似文献

Besides isolation and characterization of isorhamnetin-3-O-rutinoside, isorhamnetin-3-O-glucopyranoside and taraxasteryl acetate, a new flavonol trisaccharide was isolated from the aerial parts of Calotropis gigantea, and its structure was established as isorhamnetin-3-O-[2-O-beta-D-galactopyranosyl-6-O-alpha-L-rhamnopy ranosyl]- beta-D-glucopyranoside by a combination of fast atom bombardment mass spectroscopy, 1H and 13C NMR spectra and some chemical degradations.  相似文献

The sera of 36 normal controls, 45 patients with various diseases and 11 pregnant women were screened for circulating immune complexes using three relatively simple and inexpensive techniques. These included inhibition of agglutination of IgG coated latex particles with a serum having rheumatoid factor activity, polyethylene glycol precipitation and anti-complementary activity test. The circulating immune complexes were detected in a significantly higher proportion of patients as compared to normal controls. In the patients, the presence of circulating immune complexes did not always correlate with clinically detectable immunoinflammatory tissue damage indicating that pathogenic as well as nonpathogenic immune complexes were being detected by the above mentioned techniques. The alpha-1-antitrypsin/C3 ratio, however, correlated well with clinically apparent immuno-inflammation.  相似文献
