Over two hundred samples were collected in tropical headwater forested catchments in the lowland Amazon basin near Juruena, Mato Grosso Brazil. These were analyzed for fluorescence characteristics and DOC concentrations, and represented a range of terrestrial hydrologic flowpaths and first-order streams during baseflow and stormflow conditions. The fluorescence index (FI) of McKnight et al. (2001) was found to have a significant relationship with DOC concentrations for stream water at baseflow conditions, but FI values within individual terrestrial flowpaths and stormflow varied little for the range of DOC concentrations observed. FI values were seen to increase for increasing residence time of water within the terrestrial ecosystem, while DOC concentration decreased for increasing hydrologic residence time. The FI of terrestrial flow paths indicated that DOC became increasingly characterized by microbially derived carbon for flow paths with longer residence times, on the order through fall and overland flow < percolating soil water < groundwater. Base flow samples of stream water had a mean FI value of 1.78, compared with 1.51 and 1.44 for through fall and overland flow, respectively, and 1.65 for percolating soil water. We previously described a dominant suppressor of the splicing defect conferred by an A----C intron branchpoint mutation in S. cerevisiae. Suppression occurs by increasing the frequency with which the mutant branchpoint is utilized. We have now cloned the genomic region encoding the prp16-1 suppressor function and have demonstrated that PRP16 is essential for viability. A 1071 amino acid open reading frame contains sequence motifs characteristic of an NTP binding fold and further similarities to a superfamily of proteins that includes members with demonstrated RNA-dependent ATPase activity. A single nucleotide change necessary to confer the prp16-1 suppressor phenotype results in a Tyr----Asp substitution near the "A site" consensus for NTP binding proteins. We propose that PRP16 is an excellent candidate for mediating one of the many ATP-requiring steps of spliceosome assembly and that accuracy of branchpoint recognition may be coupled to ATP binding and/or hydrolysis. 相似文献
A new parasitic copepod, Tiddergasilus bipartitusn. sp. (Copepoda, Ergasilidae), is described herein based on three adult females found attached to the gills of the yellow-tail lambari Astyanax lacustris (Lütken), sampled in Pardo River, municipality of Botucatu, São Paulo State, Brazil. Tiddergasilus bipartitusn. sp. was recognized as a new member of the monotypic genus Tiddergasilus Marques & Boeger, 2018 by having: antennule 6-segmented; maxillary basis armed with 2 terminal teeth and ornamented with multiple spinules; second and third leg both with endopod 3-segmented. The new copepod can be distinguished from its congener, Tiddergasilus iheringi (Tidd, 1942), by the morphology of the antennal claw, number of somites/segments in abdomen and fourth leg, and by the ornamentation of the first leg. This report expands the geographic distribution and diversity of fish species parasitized with Tiddergasilus spp. in Brazil. Moreover, it also represents the first report of an ergasilid species in the Pardo River. A list of diagnostic features for the Brazilian species of Ergasilus von Nordmann, 1832 is provided herein, in order to aid comparisons with the species of this complex genus.
The methylerythritol 4-phosphate (MEP) pathway synthesizes the precursors for an astonishing diversity of plastid isoprenoids, including the major photosynthetic pigments chlorophylls and carotenoids. Since the identification of the first two enzymes of the pathway, deoxyxylulose 5-phoshate (DXP) synthase (DXS) and DXP reductoisomerase (DXR), they both were proposed as potential control points. Increased DXS activity has been shown to up-regulate the production of plastid isoprenoids in all systems tested, but the relative contribution of DXR to the supply of isoprenoid precursors is less clear. In this work, we have generated transgenic Arabidopsis thaliana plants with altered DXS and DXR enzyme levels, as estimated from their resistance to clomazone and fosmidomycin, respectively. The down-regulation of DXR resulted in variegation, reduced pigmentation and defects in chloroplast development, whereas DXR-overexpressing lines showed an increased accumulation of MEP- derived plastid isoprenoids such as chlorophylls, carotenoids, and taxadiene in transgenic plants engineered to produce this non-native isoprenoid. Changes in DXR levels in transgenic plants did not result in changes in␣DXS gene expression or enzyme accumulation, confirming that the observed effects on plastid isoprenoid levels in DXR-overexpressing lines were not an indirect consequence of altering DXS levels. The results indicate that the biosynthesis of MEP (the first committed intermediate of the pathway) limits the production of downstream isoprenoids in Arabidopsis chloroplasts, supporting a role for DXR in the control of the metabolic flux through the MEP pathway. 相似文献
Bordetella bronchiseptica produces respiratory disease primarily in mammals including humans. Although a considerably amount of research has been generated regarding lipopolysaccharide (LPS) role during infection and stimulating innate and adaptive immune response, mechanisms involved in LPS synthesis are still unknown. In this context we searched in B. bronchiseptica genome for putative glycosyltransferases. We found possible genes codifying for enzymes involved in sugar substitution of the LPS structure. We decided to analyse BB3394 to BB3400 genes, closed to a previously described LPS biosynthetic locus in B. pertussis. Particularly, conservation of BB3394 in sequenced B. bronchiseptica genomes suggests the importance of this gene for bacteria normal physiology. Deletion of BB3394 abolished resistance to naive serum as described for other LPS mutants. When purified LPS was analyzed, differences in the LPS core structure were found. Particularly, a GalNA branched sugar substitution in the core was absent in the LPS obtained from BB3394 deletion mutant. Absence of GalNA in core LPS alters immune response in vivo but is able to induce protective response against B. bronchiseptica infection. 相似文献
Global interest in sugarcane has increased significantly in recent years due to its economic impact on sustainable energy production. Sugarcane breeding and better agronomic practices have contributed to a huge increase in sugarcane yield in the last 30?years. Additional increases in sugarcane yield are expected to result from the use of biotechnology tools in the near future. Genetically modified (GM) sugarcane that incorporates genes to increase resistance to biotic and abiotic stresses could play a major role in achieving this goal. However, to bring GM sugarcane to the market, it is necessary to follow a regulatory process that will evaluate the environmental and health impacts of this crop. The regulatory review process is usually accomplished through a comparison of the biology and composition of the GM cultivar and a non-GM counterpart. This review intends to provide information on non-GM sugarcane biology, genetics, breeding, agronomic management, processing, products and byproducts, as well as the current technologies used to develop GM sugarcane, with the aim of assisting regulators in the decision-making process regarding the commercial release of GM sugarcane cultivars. 相似文献
This study calculated the compositional nutrient diagnosis (CND) norms of cowpea (Vigna unguiculata (L.) Walp.), as well as identified significant nutrient interactions of this crop growing in an irrigated calcareous desert soil. Three genotypes were distributed in rows in a 2-ha field. The soil showed high heterogeneity in its chemical properties. For statistical analysis, 86 foliar composite samples from healthy plants were used. Preliminary CND norms were developed using a cumulative variance ratio function and the 2 distribution function. Means and standard deviations of row-centered log ratios VX of five nutrients (N, P, K, Ca, and Mg) and a filling value R, which included all nutrients not chemically analyzed. Preliminary CND norms are: VN*=0.174±0.095, VP*=–2.172±0.234, VK*=–0.007±0.267, VCa*=–0.022±0.146, VMg*=–1.710±0.132, and VR5*=3.728±0.084. These CND norms are associated with dry bean yields higher than 1.88 t ha–1, and are associated with the following foliar concentrations: 26.2 g N kg–1, 2.5 g P kg–1, 22.9 g K kg–1, 21.6 g Ca kg–1, and 4 g Mg kg–1. Cowpea plants growing in desert calcareous soils took up lower amounts of N, P, and K than those considered as optimum in a previous report. Six interactions were strongly indicated for cowpea through principal component analyses: positive for Ca–Mg, and negative for N–Ca, N–Mg, Ca–P, Mg–P, and K–P. A species of Isospora Schneider, 1881 (Protozoa: Apicomplexa: Eimeriidae) considered as new to science is described and characterised molecularly from the eastern white-throated spadebill Platyrinchus mystaceus Vieillot in the Parque Nacional do Itatiaia, southeastern Brazil. Isospora lopesi n. sp. has oöcysts that are subspheroidal to ovoidal, 18–24 × 18–22 (20.6 × 19.7) µm, with smooth, bilayered wall, c.1.5 μm thick. Micropyle and oöcyst residuum are absent, but one polar granule is present. Sporocysts are ellipsoidal, 12–16 × 8–11 (14.4 × 8.6) µm. The Stieda body is flattened to half-moon-shaped and sub-Stieda body is rounded. Sporocyst residuum is present, consisting of numerous spherules of different sizes. Sporozoites are vermiform with anterior and posterior refractile bodies and nucleus. Molecular analysis was conducted at the mitochondrial cytochrome c oxidase subunit 1 (cox1) gene. This new isolate exhibited similarity greater than 98% with Isospora spp. isolates from spectacled warblers Sylvia conspicillata Temminck, 1820. This is the fourth isosporoid coccidian described from New World tyrannid birds, but is the first to have a complementary molecular characterisation. 相似文献
