Top predator loss is a major global problem, with a current trend in biodiversity loss towards high trophic levels that modifies most ecosystems worldwide. Most research in this area is focused on large-bodied predators, despite the high extinction risk of small-bodied freshwater fish that often act as apex consumers. Consequently, it remains unknown if intermittent streams are affected by the consequences of top-predators’ extirpations. The aim of our research was to determine how this global problem affects intermittent streams and, in particular, if the loss of a small-bodied top predator (1) leads to a ‘mesopredator release’, affects primary consumers and changes whole community structures, and (2) triggers a cascade effect modifying the ecosystem function. To address these questions, we studied the top-down effects of a small endangered fish species, Barbus meridionalis (the Mediterranean barbel), conducting an enclosure/exclosure mesocosm experiment in an intermittent stream where B. meridionalis became locally extinct following a wildfire. We found that top predator absence led to ‘mesopredator release’, and also to ‘prey release’ despite intraguild predation, which contrasts with traditional food web theory. In addition, B. meridionalis extirpation changed whole macroinvertebrate community composition and increased total macroinvertebrate density. Regarding ecosystem function, periphyton primary production decreased in apex consumer absence. In this study, the apex consumer was functionally irreplaceable; its local extinction led to the loss of an important functional role that resulted in major changes to the ecosystem’s structure and function. This study evidences that intermittent streams can be affected by the consequences of apex consumers’ extinctions, and that the loss of small-bodied top predators can lead to large ecosystem changes. We recommend the reintroduction of small-bodied apex consumers to systems where they have been extirpated, to restore ecosystem structure and function. 相似文献

Aim The evolutionary processes structuring the composition of communities remain unclear due to the complexity of factors active at various spatial and temporal scales. Here, we conducted ecological and evolutionary analyses of communities of caddisflies in the genus Hydropsyche (Insecta: Trichoptera) composed of ecomorphologically differentiated species. Location River ecosystems in the Iberian Peninsula and northern Morocco. Methods Nineteen environmental variables were assessed at 180 local study sites and species presence/absence at these sites was used to determine their ecological niche. The evolutionary framework for all 19 species of Hydropsyche encountered was generated by phylogenetic analysis of the mitochondrial cytochrome c oxidase subunit I gene and three nuclear genes: wingless, elongation factor 1‐alpha and 28S RNA. The phylogenetic tree was used: (1) to assess evolutionary niche conservatism by ecological trait correlation with the tree; and (2) to analyse the phylogenetic relatedness of community member species, at three spatial scales (local stream reaches, drainage basins, biogeographical regions). Results Ecological measurements grouped most species into either headwater, mid‐stream or lowland specialists, and traits presumably relevant to river zonation were found to be phylogenetically conservative. Species assemblages at local stream reaches were mostly mono‐ or dispecific. Species diversity increased at larger spatial scales, by adding species with non‐overlapping ecological niches at the level of river basins and by turnover of anciently differentiated lineages at the level of biogeographical regions. This indicates the effects of competition and niche filtering on community structure locally, and ancient ecological diversification and allopatric speciation, respectively, in building up the species pool at basin and biogeographical scales. Main conclusions The study demonstrates the importance of scale (grain size) in studying what determines community composition. Current ecological factors (i.e. competitive exclusion) in Hydropsyche were evident only when studying narrow local sites, while studies of assemblages at larger spatial scales instead demonstrated the roles of ecological niche differentiation, phylogenetic history of trait diversification and allopatric speciation. Functional traits are growing in popularity in modern ecology, but feeding studies remain primarily rooted in a taxonomic‐based perspective. However, consumers do not have any reason to select their prey using a taxonomic criterion, and prey assemblages are variable in space and time, which makes taxon‐based studies assemblage‐specific. To illustrate the benefits of the trait‐based approach to assessing food choice, we studied the feeding ecology of the endangered freshwater fish Barbus meridionalis. We hypothesized that B. meridionalis is a selective predator which food choice depends on several prey morphological and behavioral traits, and thus, its top‐down pressure may lead to changes in the functional composition of in‐stream macroinvertebrate communities. Feeding selectivity was inferred by comparing taxonomic and functional composition (13 traits) between ingested and free‐living potential prey using the Jacob's electivity index. Our results showed that the fish diet was influenced by 10 of the 13 traits tested. Barbus meridionalis preferred prey with a potential size of 5–10 mm, with a medium–high drift tendency, and that drift during daylight. Potential prey with no body flexibility, conical shape, concealment traits (presence of nets and/or cases, or patterned coloration), and high aggregation tendency had a low predation risk. Similarly, surface swimmers and interstitial taxa were low vulnerable to predation. Feeding selectivity altered the functional composition of the macroinvertebrate communities. Fish absence favored taxa with weak aggregation tendency, weak flexibility, and a relatively large size (10–20 mm of potential size). Besides, predatory invertebrates may increase in fish absence. In conclusion, our study shows that the incorporation of the trait‐based approach in diet studies is a promising avenue to improve our mechanistic understanding of predator–prey interactions and to help predict the ecological outcomes of predator invasions and extinctions. 相似文献

The aim of this study was to investigate biohydrogen production from biofuel-reforming processes using new multi-component bulk-type cobalt-based catalysts. The addition of different components to improve the catalytic performance was studied. Monometallic cobalt catalyst and catalysts containing Ru (ca. 1%) and/or Na (ca. 0.5%) were characterized and tested in the 623-673 K temperature range in ethanol steam reforming (ESR) with a steam/carbon ratio (S/C) of 3. The catalysts showed a high performance for hydrogen production and, except for H(2) and CO(2), only small amounts of by-products were obtained, depending on the temperature and the catalyst used. The catalyst containing both Ru and Na (Co-Ru(Na)) showed the best catalytic behavior in ESR. It operated stably for at least 12 days under cycles of oxidative steam reforming of glycerol/ethanol mixtures (S/C=2) and activation under O(2). 相似文献

In the past, we proposed to develop a heterologous recombinant BCG prime-recombinant modified vaccinia virus Ankara (MVA) boost dual pediatric vaccine platform against transmission of breast milk HIV-1 and Mycobacterium tuberculosis (Mtb). In this study, we assembled an E. coli-mycobacterial shuttle plasmid pJH222.HIVACAT expressing HIV-1 clade A immunogen HIVA. This shuttle vector employs an antibiotic resistance-free mechanism based on Operator-Repressor Titration (ORT) system for plasmid selection and maintenance in E. coli and lysine complementation in mycobacteria. This shuttle plasmid was electroporated into parental lysine auxotroph (safer) strain of BCG to generate vaccine BCG.HIVACAT. All procedures complied with Good Laboratory Practices (GLPs). We demonstrated that the episomal plasmid pJH222.HIVACAT was stable in vivo over a 20-week period, and genetically and phenotypically characterized the BCG.HIVACAT vaccine strain. The BCG.HIVACAT vaccine in combination with MVA.HIVA induced HIV-1- and Mtb-specific interferon γ-producing T-cell responses in newborn and adult BALB/c mice. On the other hand, when adult mice were primed with BCG.HIVACAT and boosted with MVA.HIVA.85A, HIV-1-specific CD8+ T-cells producing IFN-γ, TNF-α, IL-2 and CD107a were induced. To assess the biosafety profile of BCG.HIVACAT-MVA.HIVA regimen, body mass loss of newborn mice was monitored regularly throughout the vaccination experiment and no difference was observed between the vaccinated and naïve groups of animals. Thus, we demonstrated T-cell immunogenicity of a novel, safer, GLP-compatible BCG-vectored vaccine using prototype immunogen HIVA. Second generation immunogens derived from HIV-1 as well as other major pediatric pathogens can be constructed in a similar fashion to prime protective responses soon after birth. 相似文献

In Spain, a national project known as GUADALMED, focusing on Mediterranean streams, has been carried out from 1998 to 2005
to implement the European water framework directive (WFD) requirements. One of the main objectives of the second phase of
the project (2002–2005) was to develop a predictive system for the Spanish Mediterranean aquatic macroinvertebrate communities.
A combined-season (spring, summer, and autumn) predictive model was developed by using the latest improvements on the selection
of best predictor variables. Overall model performance measures were used to select the best discriminant function (DF) models,
and also to evaluate their biases and precision. The final predictive model was based on the best five DF models. Each one
of these models involved eight environmental variables. Final observed (O), expected (E), and O/E values for the number of
macroinvertebrate families (NFAM) and two biotic indices (IBMWP and IASPT) were calculated by averaging their values, previously
weighted by the quality of each DF model. Regression analyses among the final O and E values for the calibration dataset showed
a high proximity to the ideal theoretical model, where the final E values explained 73–84% of the variation present in the
macroinvertebrate communities of the Spanish Mediterranean watercourses. The ANOVA performed among the reference (calibration
and validation) and test datasets showed clear differences for the O/E values. Finally, the assessments carried out by the
predictive model were sensitive to anthropogenic pressure present in the study area and allowed the definition of five ecological
status classes according to the WFD requirements.

In Mediterranean climate areas, the great seasonal variability in temperature and rainfall is considered to be an evolutionary
pressure that constrains plant and animal communities and their biological traits. Droughts alter habitat availability (changes
of flow alter riffle-pool sequences), although habitat characteristics may also exacerbate drought to some extent. Using a
simple quantitative index based on the proportion of conglomerate bedrock versus gravel and cobbles, pools versus riffles
and winter versus summer flow, we show how habitat characteristics (in terms of substratum and flow) may influence the permanency
of a stream site and how flow permanence constrains macroinvertebrate community structure and biological traits. Annual and
seasonal macroinvertebrate richness, and the EPT (Ephemeroptera, Plecoptera and Trichoptera) and OCH (Odonata, Coleoptera
and Hemiptera) metrics differed between permanent, intermittent and ephemeral sites, but not between permanent and intermittent
sites. In contrast, distinct biological traits were observed in the three flow categories, although permanent sites presented
few significant traits which was attributed to the stability of the habitat. Intermittent sites were dominated by taxa with
pool-like strategies, while ephemeral sites were characterized by fauna with life-history adaptations to floods and droughts.
In contrast to most traits (e.g., dissemination, reproduction, substrate relation), which were more constrained by local flow
and substrate characteristics, life-cycle characteristics did not differ significantly among flow categories. This pattern
can be explained by the features of the Mediterranean climate and particularly its high seasonal predictability, which serves
as a large-scale filter of life-cycle traits, independently of local hydromorphological characteristics. Our findings indicate
that drought is related to habitat characteristics and that local habitat variability favours organisms with certain traits,
while other traits are independent of habitat variability on this scale and are probably affected by other large-scale habitat
characteristics.

Data on macroinvertebrates of selected reference sites were compiled from a long-term monitoring programme carried out in the Mediterranean Catalan Basins (NE Spain) that permitted analysis for nine years, from 1996 to 2004, using a homogeneous data collection procedure. This study aims to analyse the differences in composition and structure of macroinvertebrate communities at family level in five Mediterranean river types, and the values of biological quality metrics (IBMWP and IASPT indices, taxon richness and EPT) in reference conditions. Also differences between seasons (spring vs. summer) and between dry and wet periods were analysed. The dry and wet periods were determined using the Standardised Precipitation Index (SPI). A total of 29 reference sites were selected out of 184 sampling sites analysed, and 171 reference samples were available (from 1996 to 2004), of which 88 were sampled in dry periods, whereas 83 correspond to wet periods. Differences on community composition at family level were appreciated, clustering the rivers in three different groups: (1) rivers with a continuous flow regime located in siliceous zones; (2) rivers with a continuous flow regime located in calcareous zones; and (3) temporary rivers regardless of geology. Moreover, our results explain that the characteristics of hydrological periods (dry and wet) characterize the differences between communities better than just the season. The analysis of four biological quality metrics reveals clear differences between values obtained from dry and wet periods concerning taxon richness, EPT values and IBMWP biological indices, whereas the IASPT index does not show significant differences. The median taxonomic richness in wet periods is 32 macroinvertebrate families per sample while in dry periods this value falls to 22. Reference values of IBMWP index, the total number of taxa, and EPT metric are different between dry and wet periods in spring samples, while these differences are not relevant for IASPT index except for temporary streams. Hydrological specific conditions should additionally be considered in order to better calculate biological reference conditions, and to properly apply biological quality metrics used to establish the ecological status in Mediterranean rivers, especially in temporary ones. The use of the dry–wet period classification according to the climate characteristic results is a more accurate application of the Water Framework Directive in Mediterranean rivers. Implications of future climate change should be also considered from our results. 相似文献
