Many advanced medical and biological devices require microscale patterning of cells, proteins, and other biological materials. This article describes the use of piezoelectric ink jet processing in the fabrication of biosensors, cell-based assays, and other microscale medical devices. A microelectromechanical system-based piezoelectric transducer was used to develop uniform fluid flow through nozzles and to prepare well-defined microscale patterns of proteins, monofunctional acrylate ester, sinapinic acid, deoxyribonucleic acid (DNA), and DNA scaffolds on relevant substrates. Our results demonstrate that piezoelectric ink jet deposition is a powerful non-contact, non-destructive additive process for developing biosensors, cell culture systems, and other devices for medical and biological applications.  相似文献

The in vitro influence of three prebiotics viz. mannitol, maltodextrin and sorbitol was evaluated on probiotic aspects like bile salt tolerance, cholesterol lowering efficiency and bacteriocin production of the strain, Pediococcus acidilactici LAB 5 which was isolated from vacuum packed fermented meat product. Optimum temperature for bacteriocin production in MRS medium was 37°C. The strain deconjugated bile salt (sodium taurocholate) to 607.66 ± 10.894 μg/ml from initial bile salt concentration 3 mg/ml. In vitro cholesterol removal capability of the strain P. acidilactici LAB 5 was 62 ± 2.742 μg/ml. Prebiotic sorbitol had a positive influence on the probiotic parameters like better cell growth, bacteriocin production and cholesterol removal capability. Anaerobic condition had influenced largely on bile salt deconjugation, cholesterol removal and bacteriocin production. Synbiotic treatment of P. acidilactici LAB 5 with sorbitol for 1 month lowered the plasma cholesterol level to 176.34 ± 12.66 μg/ml in comparison to untreated one (208.76 ± 20.27 μg/ml) in Swiss albino mice. Intestinal adherence of P. acidilactici LAB 5 was also more in synbiotic condition than only in probiotic and control treatments.  相似文献

Packaging waste forms a significant part of municipal solid waste and has caused increasing environmental concerns, resulting in a strengthening of various regulations aimed at reducing the amounts generated. Among other materials, a wide range of oil-based polymers is currently used in packaging applications. These are virtually all non-biodegradable, and some are difficult to recycle or reuse due to being complex composites having varying levels of contamination. Recently, significant progress has been made in the development of biodegradable plastics, largely from renewable natural resources, to produce biodegradable materials with similar functionality to that of oil-based polymers. The expansion in these bio-based materials has several potential benefits for greenhouse gas balances and other environmental impacts over whole life cycles and in the use of renewable, rather than finite resources. It is intended that use of biodegradable materials will contribute to sustainability and reduction in the environmental impact associated with disposal of oil-based polymers.The diversity of biodegradable materials and their varying properties makes it difficult to make simple, generic assessments such as biodegradable products are all ‘good’ or petrochemical-based products are all ‘bad’. This paper discusses the potential impacts of biodegradable packaging materials and their waste management, particularly via composting. It presents the key issues that inform judgements of the benefits these materials have in relation to conventional, petrochemical-based counterparts. Specific examples are given from new research on biodegradability in simulated ‘home’ composting systems. It is the view of the authors that biodegradable packaging materials are most suitable for single-use disposable applications where the post-consumer waste can be locally composted.  相似文献

Extraction of the pulp of ripe berries of Bryonia lacinosa with 1% aqueous acetic acid yielded a polysaccharide material, having d-glucose, d-mannose and l-arabinose in the molar ratio of. 5.00:3.01:4.00. Hydrolysis of the fully methylated polysaccharide furnished 2,3,4,6-tetra-O-methyl-d-glucose, 2,3-di-O-methyl-d-glucose, 2,3,6-tri-O-methyl-d-mannose, 2,3-di-O-methyl-d-mannose and 2,3,5,-tri-O-methyl-l-arabinose in 1:4:2:1:4 molar ratio. Partial hydrolysis of the polysaccharide furnished; mannobiose, epicellobiose, 6-O-β-l-arabinofuranosyl-d-glucose, 6-O-α-mannopyranosyl-d-mannose and epimaltose along with the component monosaccharides. On metaperiodate oxidation studies, 100 g of the polysaccharide liberated 0.055 mol of HCOOH consuming 0.7127 mol of periodate, indicating about 8.33% of the end groups. On the basis of the above results, a structure for the repeating unit of the polysaccharide has been proposed. The polysaccharide was tested for the microbial activity and was found to be active against Escherichia coli with a minimum dose of 6.25 mg/mL.  相似文献

ST2 gene products that are members of IL-1 receptor family are expressed in various cells such as growth-stimulated fibroblasts and Th2 helper T-cells, and recently, IL-33, which belongs to IL-1 family, was identified as the ligand for ST2L, the receptor type product of the ST2 gene. Subsequently, IL-33 and ST2L have been reported to be involved in Th2 immunity and inflammation, however, their functions on non-immunological cells are still obscure. Among non-immunological adhesive cells, vascular endothelial cells were reported to express both ST2 gene products and IL-33, therefore, we investigated the expression manner of the ST2 gene in vascular endothelial cells and the effect of IL-33 on endothelial cells. ST2 gene was expressed in each of the vascular endothelial cell types tested, and the expression was growth-dependent and down-regulated when the cells were differentiated to form vascular structures on the extracellular membrane matrix. IL-33 scarcely affected the growth and tube formation of the endothelial cells, but induced IL-6 and IL-8 secretion from endothelial cells with the rapid activation of extracellular signal-regulated kinase (ERK) 1/2, so IL-33 is supposed to involve in inflammatory reaction of vascular endothelial cells through its receptor, ST2L.  相似文献

The nonviral vector based gene delivery approach is attractive due to advantages associated with molecular-level modifications suitable for optimization of vector properties. In a new class of nonviral gene delivery systems, we herein report the potential of poly(ether imine) (PETIM) dendrimers to mediate an effective gene delivery function. PETIM dendrimer, constituted with tertiary amine branch points, n-propyl ether linkers and primary amines at their peripheries, exhibits significantly reduced toxicities, over a broad concentration range. The dendrimer complexes pDNA effectively, protects DNA from endosomal damages, and delivers to the cell nucleus. Gene transfection studies, utilizing a reporter plasmid pEGFP-C1 and upon complexation with dendrimer, showed a robust expression of the encoded protein. The study shows that PETIM dendrimers are hitherto unknown novel gene delivery vectors, combining features of poly(ethylene imine)-based polymers and dendrimers, yet are relatively nontoxic and structurally precise.  相似文献

Rat liver mitoplasts free of detectable microsomal contamination can activate significant amounts of aflatoxin B1 (AFB1) into electrophilic reactive forms. The activated carcinogen binds to mitochondrial macromolecules and inhibits mitochondrial RNA and protein biosynthesis. The mitochondrial monooxygenase oxygenase system for AFB1 activation is restricted to liver and kidney as tested by enzyme assays and effects on mitochondrial RNA and protein synthesis.  相似文献

Food poisoning in man due to heat-sensitive strains of Cl. perfringens type A appeared to be mediated through enterotoxin synthesized in vivo during sporulation. A minimum of 2.0 X 10(5) vegetative cells suspended in sporulation medium was sufficient to elicit gut-loop response in rabbits. The functional disturbance in the gut as well as the structural changes were progressive and proportional to the size of the inoculum up to a dose limit of 2.0 X 10(7) vegetative cells and beyond this the changes remained steady.  相似文献

Previous studies have demonstrated the presence of super-high affinity endothelin receptors with apparent Kd's on the order of pM in different brain tissues. This study was designed to characterize, in detail, the receptors present in SCP cells, a non-transformed sheep choroid plexus cell line. Competitive binding assays with receptor-selective ligands indicated the presence of at least three classes of binding sites: a conventional receptor of the ETA subtype with a Kd = 0.4 nM that mediates an increase in intracellular levels of inositol 1,4,5-trisphosphate (IP3) in response to ET-1 and two additional sites with much higher binding affinities. The latter two sites are not coupled to the common signal transduction pathways of IP3, cAMP and cGMP. Northern blot analysis confirmed the presence of only the ETA subtype mRNA in SCP cells. It remains to determined if the multiple binding sites are distinct gene products, multiple affinity states of a single receptor molecule or a result of cooperative association of one site with either the ligand or with other proteins.  相似文献
