In order to gain further insight into the partly-characterized carotenoid biosynthetic pathway in corn (Zea mays L.), we cloned cDNAs encoding the enzymes carotenoid isomerase (CRTISO) and β-carotene hydroxylase (BCH) using endosperm mRNA isolated from inbred line B73. For both enzymes, two distinct cDNAs were identified mapping to different chromosomes. The two crtiso cDNAs (Zmcrtiso1 and Zmcrtiso2) mapped to unlinked genes each containing 12 introns, a feature conserved among all crtiso genes studied thus far. ZmCRTISO1 was able to convert tetra-cis prolycopene to all-trans lycopene but could not isomerize the 15-cis double bond of 9,15,9′-tri-cis-ζ-carotene. ZmCRTISO2 is inactivated by a premature termination codon in B73 corn, but importantly the mutation is absent in other corn cultivars and the active enzyme showed the same activity as ZmCRTISO1. The two bch cDNAs (Zmbch1 and Zmbch2) mapped to unlinked genes each coding sequences containing five introns. ZmBCH1 was able to convert β-carotene into β-cryptoxanthin and zeaxanthin, but ZmBCH2 was able to form β-cryptoxanthin alone and had a lower overall activity than ZmBCH1. All four genes were expressed during endosperm development, with mRNA levels rising in line with carotenoid accumulation (especially zeaxanthin and lutein) until 25 DAP. Thereafter, expression declined for three of the genes, with only Zmcrtiso2 mRNA levels maintained by 30 DAP. We discuss the impact of paralogs with different expression profiles and functions on the regulation of carotenoid synthesis in corn.  相似文献

Many proteins displayed differential expression (either up- or down-regulation) when proteome of migrating and non-migrating epithelium was assessed using 2-DE and ESI-Q-TOF MS/MS. From the up-regulated set, we have identified for the first time a 69-kDa albumin precursor protein with four peptides sequences and 70-kDa heat shock protein (hsp70) with one peptide in the active phase of cell migration (48 h) during the healing process. Western blot analysis was used to further characterize these proteins at different phases (24, 48 and 72 h) of healing. An increase in the mRNA expression (measured using RT-PCR) in the active migration phase (48 h) for albumin precursor and hsp70 was also observed. Furthermore, co-immunoprecipitation studies with anti-albumin precursor and anti-hsp70 antibodies, followed by immunoblotting with anti-fibronectin antibody demonstrated a novel and biologically relevant interaction between albumin precursor protein and fibronectin in corneal epithelial wound healing but not with hsp70. The increased gene and protein expression of albumin and hsp70 during the active phase of cell migration (48 h) in the corneal epithelium suggests their possible role in corneal wound healing. These findings may have broader implications for developing therapeutic strategies for treating wound healing disorders.  相似文献

The temporal profile of the phosphorescence of singlet oxygen endogenously photosensitized by photosystem II (PSII) reaction centre (RC) in an aqueous buffer has been recorded using laser excitation and a near infrared photomultiplier tube. A weak emission signal was discernible, and could be fitted to the functional form a[exp( - t/t2 ) - exp( - t/t1 )] a[\exp ( - t/\tau_{2} ) - \exp ( - t/\tau_{1} )] , with $ a > 0 $ a > 0 and $ \tau_{2} > \tau_{1} $ \tau_{2} > \tau_{1} . The value of t2 \tau_{2} decreased from 11.6 ± 0.5 μs under aerobic conditions to 4.1 ± 0.2 μs in oxygen-saturated samples, due to enhanced bimolecular quenching of the donor triplet by oxygen, whereas that of t1 \tau_{1} , identifiable with the lifetime of singlet oxygen, was close to 3 μs in both cases. Extrapolations based on the low amplitude of the emission signal of singlet oxygen formed by PSII RC in the aqueous buffer and the expected values of t1 \tau_{1} and t2 \tau_{2} in chloroplasts indicate that attempts to analyse the temporal profile of singlet oxygen in chloroplasts are unlikely to be rewarded with success without a significant advance in the sensitivity of the detection equipment.  相似文献

Beta2-adrenergic receptor (beta2AR) gene polymorphisms have been reported to be associated with various asthma-related traits in different racial/ethnic populations. However, it is unknown whether beta2AR genetic variants are associated with asthma in African Americans. In this study, we have examined whether there is association between beta2AR genetic variants and asthma in African Americans. We have recruited 264 African American asthmatic subjects and 176 matched healthy controls participating in the Study of African Americans, Asthma, Genes and Environments (SAGE). We genotyped seven known and recently identified beta2AR SNP variants, then tested genotype and haplotype association of asthma-related traits with the beta2AR SNPs in our African American cohort with adjustment of confounding effect due to admixture background and environmental risk factors. We found a significant association of the SNP -47 (Arg-19Cys) polymorphism with DeltaFEF(25-75), a measure of bronchodilator drug responsiveness, in African American asthmatics after correction for multiple testing (P = 0.001). We did not observe association of the SNP +46 (Arg16Gly) variant with asthma disease diagnosis and asthma-related phenotypes. In contrast to previous results between the Arg16Gly variant and traits related to bronchodilator responsiveness, our results indicate that the Arg-19Cys polymorphism in beta upstream peptide may play an important role in bronchodilator drug responsiveness in African American subjects. Our findings highlight the importance of investigating genetic risk factors for asthma in different populations.  相似文献

The objective of this study was to explore the possibility of a recurrent increase in the ovulation rate of Malpura sheep, a non-prolific breed, by immunization against inhibin-based peptide immunogens over a period of 3 years. Adult ewes (4–7 years of age) and weighing between 28 and 38 kg were randomly allocated equally to three treatment groups. The immunization of the ewes was initiated during the autumn breeding season. Ewes were divided into three groups (n = 5 ewes/group) and actively immunized against the synthetic peptides from the αC [bIα(1–29)-Tyr30] (Group I) or αN [bI-43-Tyr152(153-167)Cys168] (Group II) area of the bovine inhibin α-subunit, conjugated to ovalbumin or against ovalbumin (control). Each ewe received a primary immunization of 400 μg immunogen and 3 booster injections, 200 μg immunogen each at 4-week intervals. Estrous was synchronized in all the ewes by administering two doses of PGF2α at 10-day intervals for three consecutive years. Ovaries of ewes were examined each year between days 4 and 6 of the synchronized cycle, with the aid of the laparoscope to determine the ovulation rate. Active immunization significantly (p < 0.05) increased the ovulation rate. The overall ovulation rate, irrespective of the treatment period was 5.2 ± 0.44, 2.3 ± 0.38 and 0.9 ± 0.11 in Group I, Group II and the control, respectively. Although the beneficial effect of immunization on ovulation rate persisted for the entire period of the study, the interaction between immunization treatment and the time period was non-significant. The results clearly indicate that the active immunization against inhibin peptides can induce multiple ovulations in Malpura ewes and its effect on multiple ovulations is sustained for a prolonged period of time after the initial immunization.  相似文献
