A cDNA library was prepared from the bulk mRNA of mature wheat embryos and screened with mixed 32P-labeled oligonucleotide probes that encoded parts of the partial amino-acid sequence for the Zn-containing Ec protein. Each DNA insert in 11 positives from a screen of 10(5) plaques encoded a 5' untranslated and a 3' untranslated region, in addition to an open reading frame (of 81 amino acids) which, in every case, corresponded to at least 56 of the 59 amino acids in the partial polypeptide sequence previously determined for the Ec protein. The three different mRNA sequences encoded in the cDNA probably correspond to single-copy genes in the A, B and D genomes of hexaploid wheat. A wheat genomic library was screened with 32P-labeled cDNA and gave a single positive in a screen of 5 x 10(5) plaques. A 3.1-kb genomic fragment (gf-3.1) was sequenced and a cap site for the encoded mRNA was determined by primer extension. The gf-3.1 sequence encodes an intronless mRNA for the Ec protein and contains appreciable amounts of 5' and 3' flanking sequences. In addition to a putative TATA box, two inverted-repeat sequences and one direct-repeat sequence, the 5' flank in gf-3.1 contains a sequence similar to the abscisic-acid-responsive element in other higher-plant genes but does not contain sequences similar to the metal-responsive elements in animal metallothionein genes. Consistent with these findings, RNA blotting shows that accumulation of Ec mRNA is abundant in immature embryos, undetectable in germinated embryos and can be induced by adding abscisic acid, but not by adding Zn2+ to the medium in which mature wheat embryos are germinated. The findings suggest that the wheat Ec metallothionein genes, like mammalian liver metallothionein genes, are conspicuously expressed during embryogenesis.  相似文献

Excess Copper Induces A Cytosolic Cu,Zn-Superoxide Dismutase in Soybean Root   总被引:15,自引:0,他引:15  
The induction of several proteins in soybean roots in responseto copper was investigated. The major Cu2+-induced protein of16 kDa was purified by SDS-PAGE and 2D-PAGE. This Cu2+-inducedprotein cross-reacted with antibodies against cytosolic Cu,Zn-SODand the sequence of 40 amino-terminal residues of this proteinwas found to be 70% and 62.5% homologous to the sequences ofcytosolic Cu,Zn-SOD from rice and tomato, respectively. An assayof enzymatic activity revealed that SOD activity of Cu2+-treatedsoybean roots was more than twice that in roots of non-treatedcontrol plants. Thus, treatment with copper of soybean roots appears to inducean increase in SOD activity via the synthesis of cytosolic Cu,Zn-SOD.The induction of SOD by such treatment may be the result eitherof a direct effect of copper on the gene for SOD or of an indirecteffect via an increase in levels of O2 (Received July 12, 1991; Accepted January 22, 1992)  相似文献

A lipid transfer particle (LTP) from the hemolymph of adult male locusts, Locusta migratoria, was isolated and purified. The locust LTP exhibited its capacity to catalyze the exchange of diacylglycerol between low density lipophorin (LDLp) and high density lipophorin (HDLp). Contrary to the LTP reported for the tobacco hornworm, M. sexta, the locust LTP appeared to lack the capacity to promote net transfer of diacylglycerol to form an intermediate density lipophorin, although it seems premature to conclude the complete lack of such a capacity in locust LTP. The original concentration of LTP in hemolymph is assumed to be extremely low compared to that of lipophorin; only a catalytic amount of LTP may be present in the hemolymph (e.g., only 160 micrograms of LTP was obtained from the original hemolymph containing 400 mg protein). The molecular weight of intact LTP was estimated to be about 600,000 and the LTP was comprised of three glycosylated apoproteins, apoLTP-I (mol wt 310K), apoLTP-II (mol wt 89K), and apoLTP-III (mol wt 68K). The locust LTP contained significant amounts of lipids; the total lipid content amounted to 14.4% and the lipids were comprised of 17% hydrocarbons, 44% diacylglycerol, 8% cholesterol, 13% free fatty acid, and 18% phospholipids. The above molecular properties of locust LTP are essentially similar to those reported for M. sexta LTP.  相似文献

The chemical composition of phloem sap from the uppermost internodeof rice plants (Oryza sativa L., var. Kantou), one week afteranthesis, was compared with that of phloem sap from the leafsheath of a young seedling. The pure phloem sap from rice plantswas collected by an insect laser technique. The phloem sap from the uppermost internode contained a highlevel of sucrose (573.8 mM) which was the only sugar detected.The concentrations of total amino acids, potassium and ATP were124.8 mM, 40.4 mM and 1.76 mM, respectively. The concentrationof sucrose was three times higher and the potassium level wasone third as high in the internode sap as in the phloem sapfrom the leaf sheath. The total concentration of amino acidswas almost the same, but the relative amount of each amino acidwas quite different. The ratios of levels of Glu to Gln andof levels of Asp to Asn in the phloem sap from the uppermostinternode were smaller than those in the phloem sap from theleaf sheath. The adenylate energy charge was 0.92–0.93in both types of phloem sap. The amino acid composition of the phloem sap from the uppermostinternode was compared with that of the phloem sap of the flagleaf and the endosperm sap of the same plant, one week afteranthesis. A procedure utilizing high-pressure size-exclusion chromatography that permits rapid screening for both the types of components present in and the quantity of antifreeze glycoprotein in fish serum or solution is described. The applicability of the method is demonstrated by a comparative study of five different fish species, four of which contain the antifreeze glycoprotein and one which does not contain this protein. The antifreeze glycoprotein compositions of two fish of the same species, collected at different locations or under different environmental conditions, are also compared. A linear molecular-weight versus elution-volume function is established for both standard native proteins and the antifreeze glycoproteins, but these two lines do not coincide. The differences in tertiary structure between the antifreeze glycoproteins and normal proteins are presented as an explanation for the nonequivalence of calibration lines.  相似文献

ß-Conglycinin, the 7S seed storage protein of soybean(Glycine max [L.] Merr.), is comprised mainly of three subunits,designated , ' and ß. Expression of the gene encodingthe ß subunit is unique because its expression hasbeen shown to be down-regulated by exogenously applied L-methioninein immature soybean cotyledon cultures in vitro. Arabidopsisthaliana strain carrying a mto1-1 mutation overaccumulates solublemethionine. By using this mutant, we analyzed the effects ofmethionine on expression of the ß subunit gene invivo. Reciprocal crosses were made between the mto1-1 mutantand a transgenic A. thaliana strain, designated SNTß3,which carries a ß-glucuronidase (GUS) reporter geneunder the control of the promoter region of the ßsubunit gene. Analysis of GUS activity in F1 seeds indicatedthat the GUS activity was dramatically repressed when the mto1-1mutant plants were used as female parents. We constructed astrain which carries both the transgene and mto1-1 mutationin the homozygous state. Analyses of the GUS activity in seedsof this double homozygous strain indicated that the GUS activitywas repressed to 2.5% of control by introduction of the mto1-1mutation. Endothelin (ET) is one of the active endogenous substances regulating the functions of astrocytes. In the present study, we examined effects of ET on cyclooxygenase (COX) expression in cultured astrocytes. ET-3 (100 nM) caused transient increases in the expression of both COX2 mRNA and protein, but not those of COX1, in cultured astrocytes. ET-induced COX2 mRNA expression was suppressed by 5 microg/ml actinomycin D, 30 microM BAPTA/AM, inhibitors of protein kinase C (1-100 nM staurosporin and 100 microM H-7), 2 microM dexamethasone, and prolonged treatment with 100 nM phorbol 12-myristate 13-acetate. ET-3 stimulated production of prostaglandin (PG) E2 in cultured astrocytes. The effect of ET-3 on the PGE2 production was diminished by actinomycin D. Indomethacin and NS398, a selective COX2 inhibitor, comparably decreased both the basal and the ET-stimulated PGE2 production. Proliferation of cultured astrocytes was stimulated by 100 nM ET-3, and the increased proliferation was reduced by co-addition of 1 microM PGE2. Treatment with 1 microM PGE2 caused astrocytic morphological changes accompanied by disappearance of stress fibers, a prominent structure of organized cytoskeletal actin in cultured astrocytes. In the presence of 10 nM ET-3, PGE2 did not show an effect on astrocytic actin organization. The present study shows that ET is an inducer of astrocytic COX2 and suggests that ET-induced PGE2 production through COX2 may be involved in the regulation of astrocytic functions.  相似文献

Oxidative folding of o-conotoxin MVIIC, a highly basic 26-amino acid peptide with three disulfide bonds, predominantly gave two products with mismatched disulfide bonds in 0.1M NH4OAc buffer (pH 7.7) at 21°C both in the presence and absence of redox reagents such as reduced and oxidized glutathione. A low reaction temperature (5°C) and a high salt concentration in buffer such as 2M (NH4)2SO4 were necessary to obtain the correctly folded biologically active product. The folding reaction was found to proceed via a two-stage pathway of (I) the formation and (II) the rearrangement of the mismatched disulfide bonds. Both the reaction temperature and the salt strongly affected the equilibrium between mismatched and correctly formed disulfide bonds in the second stage. Such an effect of salts on the rearrangement reaction could be explained by anion binding at a low concentration and the salting out effect at a high concentration by analyzing the rank order of their effectiveness. The anion-binding effect was also confirmed by examining the folding of the tetra-acetylated peptide at the Lys side chains. CD study suggested that the yield of the biologically active product was correlated with its conformational change as functions of temperature and salt concentration. © 1996 John Wiley & Sons, Inc.  相似文献

Structure-activity relationship of endothelin: importance of charged groups   总被引:6,自引:0,他引:6  
Endothelin (ET)-related peptides including ET-1 (1-39) were synthesized, and their constricting activity in rat pulmonary artery rings and pressor activity in unanesthetized rat were measured to elucidate their structure-activity relationship. The vasoconstrictor activities of ET-2, ET-3 and sarafotoxin S6b were one-half, one-60th and one-third that of ET-1, respectively. Such differences in biological activities should mainly arise from sequence heterogeneity at the N-terminal portion, especially at positions 4 to 7. All of the blocked ETs at the amino or carboxyl termini showed greatly decreased activities. A monocyclic analog, in which Cys3 and Cys11 were replaced by Ala, showed one-third the activity of ET-1; however, its deamino dicarba analog was almost completely inactive. Significant activities were retained even with replacement of amino acids at positions Ser4, Ser5, Leu6, Met7, Lys9, Tyr13, and Trp21 by Ala, Ala, Gly, Met(0), Leu, Phe, and Tyr or Phe, respectively. On the other hand, replacement of Asp8, Glu10 and Phe14 by Asn, Gln and Ala, respectively, resulted in complete loss of the biological activity. These results indicated that two disulfide bonds in ET molecule were not essential for the expression of vasoconstricting activity. Both terminal amino and carboxyl groups, carboxyl groups of Asp8 and Glu10, and the aromatic group of Phe14 seemed to be contributing, more or less, to the expression of the biological activities.  相似文献
