Human TAS2 receptors (hTAS2Rs) perceive bitter tastants, but few studies have explored the structure-function relationships of these receptors. In this paper, we report our trials on the large-scale preparations of hTAS2Rs for structural analysis. Twenty-five hTAS2Rs were expressed using a GFP-fusion yeast system in which the constructs and the culture conditions (e.g., the signal sequence, incubation time and temperature after induction) were optimized by measuring GFP fluorescence. After optimization, five hTAS2Rs (hTAS2R7, hTAS2R8, hTAS2R16, hTAS2R41, and hTAS2R48) were expressed at levels greater than 1 mg protein/L of culture, which is a preferable level for purification and crystallization. Among these five bitter taste receptors, hTAS2R41 exhibited the highest detergent solubilization efficiency of 87.1% in n-dodecyl-β-d-maltopyranoside (DDM)/cholesteryl hemisuccinate (CHS). Fluorescence size-exclusion chromatography showed that hTAS2R41 exhibited monodispersity in DDM/CHS without aggregates, suggesting that hTAS2R41 is a good target for future crystallization trials.  相似文献

A novel telomerase-associated protein was isolated from porcine testis. The 115-kDa protein, purified with telomerase activity, was molecular cloned using human cDNA library, and identified as MOV10. The expression levels of both MOV10 mRNA and MOV10 protein in cancer cells were 2-3 times higher than that of the normal cells, and MOV10 mRNA was highly expressed in human testis and ovary. The anti-MOV10 antibody precipitated the telomerase activity from cancer cell extracts, and inhibited the telomerase activity in vitro. Sf9-expressed MOV10 protein bound to G-rich strand of both single- and double-stranded telomere-sequenced DNA, but not to single C-rich strand. ChIP assay showed the binding of MOV10 to telomere region in vivo. These data suggest that MOV10 is involved in the progression of telomerase-catalyzing reaction via the interaction of telomerase protein and telomere DNA.  相似文献

Zic family proteins have five C2H2-type zinc finger (ZF) motifs. We physicochemically characterized the folding properties of Zic ZFs. Alteration of chelation with zinc ions and of hydrophobic interactions changed circular dichroism spectra, suggesting that they caused structural changes. The motifs were heat stable, but electrostatic interactions had little effect on structural stability. These results highlight the importance of chelating interactions and hydrophobic interactions for the stability of the folding structure of Zic ZF proteins.  相似文献

Glis3 is a member of the Gli-similar subfamily. GLIS3 mutations in humans lead to neonatal diabetes, hypothyroidism, and cystic kidney disease. We generated Glis3-deficient mice by gene-targeting. The Glis3−/− mice had significant increases in the basal blood sugar level during the first few days after birth. The high levels of blood sugar are attributed to a decrease in the Insulin mRNA level in the pancreas that is caused by impaired islet development and the subsequent impairment of Insulin-producing cell formation. The pancreatic phenotypes indicate that the Glis3-deficient mice are a model for GLIS3 mutation and diabetes mellitus in humans.  相似文献

The purpose of this study was the enrichment and phylogenetic identification of bacteria that dechlorinate 4,5,6,7-tetrachlorophthalide (commercially designated “fthalide”), an effective fungicide for rice blast disease. Sequential transfer culture of a paddy soil with lactate and fthalide produced a soil-free enrichment culture (designated the “KFL culture”) that dechlorinated fthalide by using hydrogen, which is produced from lactate. Phylogenetic analysis based on 16S rRNA genes revealed the dominance of two novel phylotypes of the genus Dehalobacter (FTH1 and FTH2) in the KFL culture. FTH1 and FTH2 disappeared during culture transfer in medium without fthalide and increased in abundance with the dechlorination of fthalide, indicating their growth dependence on the dechlorination of fthalide. Dehalobacter restrictus TEA is their closest relative, with 97.5% and 97.3% 16S rRNA gene similarities to FTH1 and FTH2, respectively.4,5,6,7-Tetrachlorophthalide (commercially designated “fthalide”) is an effective fungicide for rice blast disease, which inhibits melanin biosynthesis and the formation of the mature appressorial cells of the rice blast pathogen on the host plant (5, 16). Fthalide has been reported to be reductively dechlorinated in soil (16) and compost (28), although its fates in paddy soil and the fthalide-dechlorinating bacteria are unknown. Besides fthalide, polychlorinated aromatic compounds are known to be reductively dechlorinated by the bacteria of several phyla. Six strains of Desulfitobacterium spp. of the phylum Firmicutes (2, 3, 6, 10, 23, 29) and Desulfomonile tiedjei DCB-1 of the phylum Proteobacteria (21) can dechlorinate polychlorinated phenols. Three strains of the phylum Chloroflexi can dechlorinate a variety of compounds, including polychlorinated phenols, benzenes, biphenyls, or dibenzo-p-dioxins: Dehalococcoides ethenogenes 195 (9, 19), Dehalococcoides sp. strain CBDB1 (1, 4), and strain DF-1 of Chloroflexi, collectively called the “o-17/DF-1 group” (18). Dehalococcoides spp. utilize hydrogen as an electron donor and acetate as a carbon source for growth coupled to the reductive dechlorination of chlorinated compounds (1, 12, 13, 19, 26). In contrast, Desulfitobacterium spp. can dechlorinate chlorinated compounds not only with hydrogen, but also organic acids, such as formate, pyruvate, lactate, or butyrate (3, 10, 23). Strain DF-1 can utilize hydrogen and formate for the dechlorination of polychlorinated biphenyls (PCBs) (18).In this study, bacteria that dechlorinate fthalide were enriched from a paddy soil with sequentially transferred cultures using a soil-free medium supplemented with single organic acids. Acetate, formate, lactate, and butyrate were used in this study because they are frequently used in the enrichment of dechlorinators and release hydrogen at different concentrations (8, 11, 14). Fthalide-dechlorinating bacteria in the enriched culture were phylogenetically identified based on the 16S rRNA gene with PCR-denaturing gradient gel electrophoresis (DGGE), a 16S rRNA gene clone library, and quantitative real-time PCR (qPCR).  相似文献

The ribosomal stalk protein L12 is essential for events dependent on the GTP-binding translation factors. It has been recently shown that ribosomes from Thermus thermophilus contain a heptameric complex L10.(L12)2.(L12)2.(L12)2, rather than the conventional pentameric complex L10.(L12)2.(L12)2. Here we describe the reconstitution of the heptameric complex from purified L10 and L12 and the characterization of its role in elongation factor G-dependent GTPase activity using a hybrid system with Escherichia coli ribosomes. The T. thermophilus heptameric complex resulted in a 2.5-fold higher activity than the E. coli pentameric complex. The structural element of the T. thermophilus complex responsible for the higher activity was investigated using a chimeric L10 protein (Ec-Tt-L10), in which the C-terminal L12-binding site in E. coli L10 was replaced with the same region from T. thermophilus, and two chimeric L12 proteins: Ec-Tt-L12, in which the E. coli N-terminal domain was fused with the T. thermophilus C-terminal domain, and Tt.Ec-L12, in which the T. thermophilus N-terminal domain was fused with the E. coli C-terminal domain. High GTPase turnover was observed with the pentameric chimeric complex formed from E. coli L10 and Ec-Tt-L12 but not with the heptameric complex formed from Ec-Tt-L10 and Tt.Ec-L12. This suggested that the C-terminal region of T. thermophilus L12, rather than the heptameric nature of the complex, was responsible for the high GTPase turnover. Further analyses with other chimeric L12 proteins identified helix alpha6 as the region most likely to contain the responsible element.  相似文献
