Biochemical Studies on “Bakanae” Fungus. Part 55 Synthesis of Gibberellins

In order to clarify the structure of ring A of gibberellins, thirteen lactones of cyclohexan series, of which eight were new, were prepared to examine their infrared spectra. So far; the experiment is concerned, γ-lactones show the characteristic absorption band in the rang 1775~1782 cm^?1 in dioxane, while 5-ones in the range 1730/~1762 cm^?1. Since the absorptio band due to lactone carbonyl of gibberellins occurs at the range 1777~1786 cm^?1 in dioxane, the lactone ring of gibberellins seems to be γ.     

Isolation and Chemical Characterization of a Mating Pheromone Produced by Saccharomyces kluyveri

The pullulanase gene (pul) of Klebsiella aerogenes was transferred in vivo to Escherichia coli by using RP4:: Mu cts. The pul gene was expressed in E. coli, although the level of pullulanase activity in E. coli was lower than that in K. aerogenes, and the Pul⁺ transconjugants were relatively unstable in an unselective medium. Production of pullulanase, which is used to make maltose from starch, was induced in E. coli by pullulan, waxy maize amylopectin, soluble starch and maltose. When the transconjugant cells of E. coli were grown with pullulan or maltose, most pullulanase was produced intracellularly, whereas K. aerogenes produced pullulanase extracellularly. Retransfer of the pul_k gene from E. coli to K. aerogenes by conjugation resulted in an increase of the production of extracellular pullulanase.     

Synthesis of dl-3-Isobutyroxy-β-ionone and dl-Dehydrovomifoliol

3-Isobutyroxy-β-ionone (III) is the proposed structure of quiesone, the naturally occurring inhibitor of the germination of Peronospora tabacina conidia. This was synthesized as a racemate and shown to possess qualitatively identical biological activity as quiesone itself. Employing an intermediate of this synthesis, dl-dehydrovomifoliol (VII) was also synthesized.     

New Syntheses of (R)-(+)-3-Acetoxy-2,6-dimethyl-l,5-heptadiene,the Pheromone of the Comstock Mealybug

L-Phenylalanine was converted to optically impure (R)-(+)-2,6-dimethyl-1,5-heptadien-3-ol 2 (19% e.e.) .(R)-(+)-2 (96% e.e.) was prepared by a kinetic resolution of (±)-2. Acetylation of the pure (R)-(+ )- 2 gave the pheromone of the Comstock mealybug ( Pseudococcus comstockii KUWANA) [(R)-(+)-1].     

Isolation and Identification of a Sexual Hormone in Yeast

5-Fluorotryptophan (5FT), indolmycin (IM), 4-fluorotryptophan and 7-azatryptophan were found on screening to be tryptophan antagonists among various chemically synthesized and naturally occurring tryptophan analogues for the isolation of l-tryptophan (l-Trp) producing mutants of Bacillus subtilis K.

From among 5FT resistant mutants, potent l-Trp producers were obtained using an improved isolation medium. Growth of the isolated 5FT-resistant l-Trp producer, AJ 11709, was inhibited by IM. From among 5FT and IM resistant mutants, the best strain, AJ 11979, which produced 9.0 g/liter of l-Trp from 13% glucose on 120hr cultivation, was selected.     

Determination of Stereochemistry at C-16 of Dihydrogibberellin A1 Methyl Ester (Methyl Tetrahyclrogibberellate) and Its 16-Epimer

Stereochemistry at C–16 of dihydrogibberellin A₁ methyl ester (methyl tetrahydrogib-berellate, III) and its 16-epimer was elucidated. NMR analysis employing a shift reagent, Eu(thd)₃ or Eu(fod)₃, was found to be very effective for settling this stereochemical problem.     

Purification and Properties of Lytic, β-1, 3-Glucanase from Flavobacterium dormitator var. glucanolyticae

An endo β-1, 3-glucanase which is able to disrupt the cells of living yeast has been purified in homogeneous state from the culture filtrate of Flavobacterium dormitator var. glucanolyticae. The molecular weight of the enzyme was estimated to be 17,000 ~ 22,000. The mode of enzyme action has been suggested to be a “random” type of β-1, 3-glucanase. The enzyme preferes larger chains saccharides as substrate for its action, however, smaller oligosaccharides such as laminaritriose and laminaribiose are also decomposed by the enzyme. The Km values of the enzyme for laminarin, laminarihexaose, and laminaritetraose were determined to be 0.26, 1.18, and 2.00 g/liter, respectively. The ability of this enzyme to disrupt the cells of living yeast is its remarkable point, since endo β-1, 3-glucanase of a smaller oligosaccharide-producing type from most sources has been recognized to be inactive (or very weakly active) on living yeast cells.     

A Stereoselective Synthesis of Antimycinone A3 and Its Stereoisomers

Antimycinone A₃, which is a neutral fragment of mild alkaline hydrolysate of antimycin A₃, and its stereoisomers were synthesized stereoselectively from methyl trans-2-n-butylpent-3-enoate or methyl cis-2-n-butylpent-3-enoate, and natural antimycinone A₃ was proved to possess Hα-Hβ and Hβ-Hγ trans configuration.     

Quantitative Analysis of Sugars in Plant Extracts by Ion-exchange Chromatography,with Special Reference to the Examination of Conditions for Preparing the Sample Sugar Solutions

Critical examinations were made on the conditions for preparing the sugar solutions to be analyzed by ion-exchange chromatography of sugar-borate complexes by the method of Khym and Zill. A procedure was proposed which gave the best recovery of sugars with minimum hydrolysis of sucrose. By means of this procedure, sugar solutions were prepared from potato tubers which had been stored at a high (30°C) or low temperature (6°C). Results of the chromatographic separation and determination of component sugars showed that main sugars present in potato tubers were sucrose, glucose, and fructose. Maltose and pentoses could not be detected. The contents of sucrose, glucose, and especially, fructose were far greater in potatoes stored at a low temperature than in those stored at a high temperature.
