BOLA‐DRB3 gene polymorphisms influence bovine leukaemia virus infection levels in Holstein and Holstein × Jersey crossbreed dairy cattle 下载免费PDF全文

Bovine leukemia virus (BLV) infections, causing persistent lymphocytosis and lethal lymphosarcoma in cattle, have reached high endemicity on dairy farms. We observed extensive inter‐individual variation in the level of infection (LI) by assessing differences in proviral load in peripheral blood. This phenotypic variation appears to be determined by host genetics variants, especially those located in the BoLA‐DRB3 MHCII molecule. We performed an association study using sequencing‐based typed BOLA‐DRB3 alleles from over 800 Holstein and Holstein × Jersey cows considering LI in vivo and accounting for filial relationships. The DBR3*0902 allele was associated with a low level of infection (LLI) (<1% of circulating infected B‐cells), whereas the DRB3*1001 and DRB3*1201 alleles were related to a high level of infection (HLI). We found evidence that 13 polymorphic positions located in the pockets of the peptide‐binding cleft of the BOLA‐DRB3 alleles were associated with LI. DRB3*0902 had unique haplotypes for each of the pockets: Ser13‐Glu70‐Arg71‐Glu74 (pocket 4), Ser11‐Ser30 (pocket 6), Glu28‐Trp61‐Arg71 (pocket 7) and Asn37‐Asp57 (pocket 9), and all of them were significantly associated with LLI. Conversely, Lys13‐Arg70‐Ala71‐Ala74 and Ser13‐Arg70‐Ala71‐Ala74, corresponding to the DRB3*1001 and *1201 alleles respectively, were associated with HLI. We showed that the specific amino acid pattern in the DRB3*0902 peptide‐binding cleft may be related to the set point of a very low proviral load level in adult cows. Many scholars of industrial ecology have focused on the institutional and organizational challenges of building and maintaining regional industrial symbiosis through the synergistic integration of material and energy flows. Despite the promise that these intellectual developments hold for the future dematerialization of industrial production, they rarely address the actual regulatory obstacles of turning wastes into raw materials. In this article we introduce a potential future industrial symbiosis around the Gulf of Bothnia between Finland and Sweden, and assess the regulatory bottlenecks related to waste by‐product consideration. We find that although the Gulf of Bothnia region has technological and economic potential for industrial symbiosis, the regulatory support for this is insufficient. We suggest a common pool resource‐based governance system that could utilize market and regulatory mechanisms in a regional‐level cross‐border system of governance. Importantly, the suggested governance system would protect the users of potential raw materials from unpredictable waste regulation, market risks related to large‐scale material flows, and societal risks of hazardous waste treatment. 相似文献

We have determined the complete nucleotide sequence of the small- subunit
ribosomal RNA genes for the ciliate protozoans Stylonychia pustulata and
Oxytricha nova. The sequences are homologous and sufficiently similar that
these organisms must be closely related. In a phylogeny inferred from
comparisons of several eukaryotic small-subunit ribosomal RNAs, the
divergence of the ciliates from the eukaryotic line of descent is seen to
coincide with the radiation of the plants, the animals, and the fungi. This
radiation is preceded by the divergence of the slime mold, Dictyostelium
discoideum.

A human malignant continuous cell line, named NG97, was recently established in our laboratory. This cell line has been serially
subcultured over 100 times in standard culture media presenting no sign of cell senescence. The NG97 cell line has a doubling
time of about 24 h. Immunocytochemical analysis of glial markers demonstrated that cells are positive for glial fibrillary
acidic protein (GFAP) and S-100 protein, and negative for vimentin. Under phase-contrast microscope, cultures of NG97 showed
cells with variable morphological features, such as small rounded cells, fusiform cells (fibroblastic-like cells), and dendritic-like
cells. However, at confluence just small rounded and fusiform cells can be observed. At scanning electron microscopy (SEM)
small rounded cells showed heterogeneous microextentions, including blebs and filopodia. Dendritic-like cells were flat and
presented extensive prolongations, making several contacts with small rounded cells, while fusiform cells presented their

Intermediate filaments have been isolated from rabbit intradural spinal nerve roots by the axonal flotation method. This method was modified to avoid exposure of axons to low ionic strength medium. The purified filaments are morphologically 75-80 percent pure. The gel electrophoretogram shows four major bands migrating at 200,000, 145,000, 68,000, and 60,000 daltons, respectively. A similar preparation from rabbit brain shows four major polypeptides with mol wt of 200,000 145,000, 68,000, and 51,000 daltons. These results indicate that the neurofilament is composed of a triplet of polypepetides with mol wt of 200,000, 145,000, and 68,000 daltons. The 51,000-dalton band that appears in brain filament preparations as the major polypeptide seems to be of glial origin. The significance of the 60,000- dalton band in the nerve root filament preparation is unclear at this time. Antibodies raised against two of the triplet proteins isolated from calf brain localize by immunofluorescence to neurons in central and peripheral nerve. On the other hand, an antibody to the 51,000-dalton polypeptide gives only glial staining in the brain, and very weak peripheral nerve staining. Prolonged exposure of axons to low ionic strength medium solubilizes almost all of the triplet polypeptides, leaving behind only the 51,000- dalton component. This would indicate that the neurofilament is soluble at low ionic strength, whereas the glial filament is not. These results indicate that neurofilaments and glial filaments are composed of different polypeptides and have different solubility characteristics. 相似文献

Ca2+-entry via L-type Ca2+ channels (DHPR) is known to trigger ryanodine receptor (RyR)-mediated Ca2+-release from sarcoplasmic reticulum (SR). The mechanism that terminates SR Ca2+ release is still unknown. Previous reports showed evidence of Ca2+-entry independent inhibition of Ca2+ sparks by DHPR in cardiomyocytes. A peptide from the DHPR loop II-III (PepA) was reported to modulate isolated RyRs. We found that PepA induced voltage-dependent “flicker block” and transition to substates of fully-activated cardiac RyRs in planar bilayers. Substates had less voltage-dependence than block and did not represent occupancy of a ryanoid site. However, ryanoids stabilized PepA-induced events while PepA increased RyR2 affinity for ryanodol, which suggests cooperative interactions. Ryanodol stabilized Imperatoxin A (IpTxA) binding but when IpTxA bound first, it prevented ryanodol binding. Moreover, IpTxA and PepA excluded each other from their sites. This suggests that IpTxA generates a vestibular gate (either sterically or allosterically) that prevents access to the peptides and ryanodol binding sites. Inactivating gate moieties (“ball peptides”) from K+ and Na+ channels (ShakerB and KIFMK, respectively) induced well resolved slow block and substates, which were sensitive to ryanoids and IpTxA and allowed, by comparison, better understanding of PepA action. The RyR2 appears to interact with PepA or ball peptides through a two-step mechanism, reminiscent of the inactivation of voltage-gated channels, which includes binding to outer (substates) and inner (block) vestibular regions in the channel conduction pathway. Our results open the possibility that “ball peptide-like” moieties in RyR2-interacting proteins could modulate SR Ca2+ release in cells. 相似文献

The zebrafish genome contains at least five msx homeobox genes, msxA, msxB,
msxC, msxD, and the newly isolated msxE. Although these genes share
structural features common to all Msx genes, phylogenetic analyses of
protein sequences indicate that the msx genes from zebrafish are not
orthologous to the Msx1 and Msx2 genes of mammals, birds, and amphibians.
The zebrafish msxB and msxC are more closely related to each other and to
the mouse Msx3. Similarly, although the combinatorial expression of the
zebrafish msx genes in the embryonic dorsal neuroectoderm, visceral arches,
fins, and sensory organs suggests functional similarities with the Msx
genes of other vertebrates, differences in the expression patterns preclude
precise assignment of orthological relationships. Distinct duplication
events may have given rise to the msx genes of modern fish and other
vertebrate lineages whereas many aspects of msx gene functions during
embryonic development have been preserved.

The reproductive biology of Seseli farrenyi (Apiaceae), a very narrow endemic to Cape Creus (Catalonia, Spain), including flowering timing patterns, quantity and quality of pollination services (type and frequency of pollinators, pollen carryover, pollen deposition on stigmas and reproductive success measured as fruit set), and breeding system was studied. Given the decline of population size detected in the last twenty years, we also analyzed the effects of fragmentation on pollination mechanisms. Protandry along with strong synchrony of floral development within umbels and sequential inflorescence emission within individual stalks, produces sexual phase alternation that promotes a strong outcrossing despite its non-specific pollination system and its (at least partial) self-compatibility. This pronounced xenogamy is supported by results of the insect exclusion test, hand-pollination experiments, and high P/O ratio. S. farrenyi flowers received visits from at least 28 species of insects, including wasps, small bees, ants, flies, syrphid flies, beetles and stink bugs, with different pollen carry-overs. Heterospecific pollen on stigmas decreased notably during the season (50% to 2.5%), averaging 12%. In the small population the stigmatic pollen loads and seed set decreased, but there was no effect of pollinator visitation rates. It was more affected by the composition of pollinators and their efficiency. The wind had a considerable effect on the plant. Some conservation measures are proposed. 相似文献
