The activity of Ca(++)-Mg++ ATP-ase present in erythrocyte membranes was determined in basal conditions and following stimulation with calmodulin in 8 women with insulin-dependent diabetes and in 9 healthy women. The isolation of erythrocyte membranes and the determination of activity of Ca(++)-Mg(++)-ATP-ase were carried out according to the method of Gietzen et al. A decrease in the activity of Ca(++)-Mg(++)-ATP-ase in basal conditions was found in fractions with the highest erythrocyte content obtained from diabetic patients. After stimulation with calmodulin the activity of Ca(++)-Mg(++)-ATP-ase in all the fractions was lower in diabetic patients than in the controls. Low activities of the enzyme were accompanied by high values of HbA1c. The results suggest that glycosylation of the ATP-ase or/and calmodulin may be the main cause of the observed fall in the enzyme activity in diabetes. Also the disturbances concerning the cumulation of intracellular calcium may be related to the changes caused by glycosylation of Ca(++)-Mg(++)-ATP-ase or/and calmodulin.  相似文献

Tetrapod limb development has been studied extensively for decades, yet the strength and role of developmental constraints in this process remains unresolved. Mammals exhibit a particularly wide array of limb morphologies associated with various locomotion modes and behaviors, providing a useful system for identifying periods of developmental constraint and conserved developmental mechanisms or morphologies. In this study, landmark‐based geometric morphometrics are used to investigate levels and patterns of morphological diversity (disparity) among the developing forelimbs of four mammals with diverse limb morphologies: mice, opossums, horses, and pigs. Results indicate that disparity among the forelimbs of these species slightly decreases or stays the same from the appearance of the limb ridge to the bud stage, and increases dramatically from the paddle through tissue regression stages. Heterochrony exhibited by the precocial opossum limb was not found to drive these patterns of morphological disparity, suggesting that the low disparity of the middle stages of limb development (e.g., paddle stage) is driven by processes operating within the limb and is likely not a result of embryo‐wide constraint.  相似文献

Tympanal ears of female gypsy moths Lymantria dispar dispar (L.) (Lepidoptera: Erebidae: Lymantriinae) are reportedly more sensitive than ears of conspecific males to sounds below 20 kHz. The hypothesis is tested that this differential sensitivity is a result of sex‐specific functional roles of sound during sexual communication, with males sending and females receiving acoustic signals. Analyses of sounds produced by flying males reveal a 33‐Hz wing beat frequency and 14‐kHz associated clicks, which remain unchanged in the presence of female sex pheromone. Females exposed to playback sounds of flying conspecific males respond with wing raising, fluttering and walking, generating distinctive visual signals that may be utilized by mate‐seeking males at close range. By contrast, females exposed to playback sounds of flying heterospecific males (Lymantria fumida Butler) do not exhibit the above behavioural responses. Laser Doppler vibrometry reveals that female tympana are particularly sensitive to frequencies in the range produced by flying conspecific males, including the 33‐Hz wing beat frequency, as well as the 7‐kHz fundamental frequency and 14‐kHz dominant frequency of associated clicks. These results support the hypothesis that the female L. dispar ear is tuned to sounds of flying conspecific males. Based on previous findings and the data of the present study, sexual communication in L. dispar appears to proceed as: (i) females emitting sex pheromone that attracts males; (ii) males flying toward calling females; and (iii) sound signals from flying males at close range inducing movement in females, which, in turn, provides visual signals that could orient males toward females.  相似文献

Background  

Aggregated settlement of kin larvae in sessile marine invertebrates may result in a complex array of compatible and incompatible allogeneic responses within each assemblage. Each such aggregate can, therefore, be considered as a distinct self-organizing biological entity representing adaptations that have evolved to maximize the potential benefits of gregarious settlement. Entomopathogenic nematodes (EPNs) suffer from trait deterioration, a potential problem when these antagonists are transferred into artificial environments for mass production. In order to improve beneficial traits of EPN genetic selection and hybridization has been successfully carried out. Should these selected strains deteriorate during serial culturing the efforts would be in vain. Inbreeding might offer a possibility to stabilize traits but can also result in inbreeding depression. This study attempted to increase heat tolerance of Heterorhabditis bacteriophora by selective breeding for seven cycles either with nematodes propagated in vivo in Galleria mellonella or with in vitro propagated nematodes which were exposed to heat stress in monoxenic liquid culture. After release of the selection pressure, the tolerance was monitored over 15 additional reproductive cycles to compare the stability of the trait. Virulence of the selected strains was assessed to check for negative tradeoff effects. Heat tolerance was successfully increased in vivo (from 39.03 to 40.85 °C) and in vitro (from 39 to 40 °C) propagated H. bacteriophora, but could only be maintained in populations which were serially reared in liquid culture. When H. bacteriophora is cultured in vivo, reproduction by cross fertilization is possible. In in vitro culture male and female cannot mate and reproduction is solely by self-fertilizing hermaphrodite resulting in homozygous inbred lines. Trait deterioration seems to be restricted to in vivo propagated H. bacteriophora, whereas monoxenic liquid cultures handling large numbers of inbred lines provided genetically stable and virulent nematode populations. Selection using liquid culture technology is thus superior over in vivo propagation to sustain beneficial traits in H. bacteriophora not only for selective breeding but also for mass production.  相似文献
