Female, male, and juvenile firebrats, Thermobia domestica (Packard) (Thysanura: Lepismatidae), employ a pheromone that arrests conspecifics on contact. Paper shelters placed in a T. domestica colony accumulate fecal excreta (= frass) and other insect‐derived debris. Such shelters elicit arrestment by conspecifics. However, the definitive source of the arrestment pheromone was not known. We tested the hypothesis that one or more debris components from a T. domestica shelter constitute the source of the arrestment pheromone. In dual‐choice, still‐air olfactometer experiments, scales, exuviae, antennae, caudal filaments, gregarine parasite cysts, and silk (each intact or macerated) retrieved from shelters and separated for experiments, as well as saliva, hemolymph, and fat body extracted from insects all failed to arrest female T. domestica. Similarly, paper that had been fed upon by insects did not elicit an arrestment response, eliminating insect‐altered cellulose as the arrestant pheromone. In contrast, insect‐exposed glass significantly arrested females. Moreover, females were significantly arrested by (i) loose, insect‐derived debris brushed from shelters, (ii) a frass mixture manually separated from loose debris, and (iii) specific amber‐type frass manually separated from the frass mixture. These results lead us to conclude that amber‐type frass constitutes the source of at least part of the T. domestica arrestment pheromone. 相似文献

Cocoon-spinning larvae of the codling moth, Cydia pomonella, emit a pheromone that mediates aggregation by pupation site-seeking fifth-instar larvae. It was unknown and, thus, we tested whether the aggregation pheromone induces arrestment or attraction responses. In paired straight-tube experiment 1, fifth-instars moved faster and farther upwind toward cospecific cocoons compared to blank controls. In still-air cage experiment 2, fifth-instars selected more often as first and final choices of pupation sites those with cocooning conspecifics than those without. Finally, in Y-tube olfactometer experiment 3, fifth-instars anemotactically responded to, and preferred, side arms with cocooning conspecifics to those without. Our data provide evidence that codling moth larvae are attracted to, rather than merely arrested by, larval aggregation pheromone. These results help explain reported aggregations or clumped distributions of larvae on tree trunks, which would likely not occur if they were based merely on chance encounter of cocoon-spinning larvae by foraging larvae. 相似文献

Aims We investigated the regulation of the water status in three predominant perennial C3 phreatophytes (Alhagi sparsifolia, Populus euphratica, Tamarix ramosissima) at typical sites of their occurrence at the southern fringe of the hyperarid Taklamakan Desert (north-west China).Methods In the foreland of the river oasis of Qira (Cele), we determined meteorological variables, plant biomass production, plant water potentials (Ψ L) and the water flux through the plants. We calculated the hydraulic conductance on the flow path from the soil to the leaves (k SL) and tested the effects of k SL, Ψ L and the leaf-to-air difference in the partial pressure of water vapour (Δ w) on stomatal regulation using regression analyses.Important findings Despite high values of plant water potential at the point of turgor loss, all plants sustained Ψ L at levels that were high enough to maintain transpiration throughout the growing season. In A. sparsifolia, stomatal resistance (r s; related to leaf area or leaf mass) was most closely correlated with k SL; whereas in P. euphratica, ~70% of the variation in r s was explained by Δ w. In T. ramosissima, leaf area-related r s was significantly correlated with Ψ L and k SL. The regulation mechanisms are in accordance with the growth patterns and the occurrence of the species in relation to their distance to the ground water. 相似文献

In laboratory and field experiments, stimuli were tested that might affect oviposition decisions by female peach twig borer moths, Anarsia lineatella Zeller (Lepidoptera: Gelechiidae). When given a choice between immature green peach fruits, green mature peach fruits and soft-ripe peach fruits, the latter received the fewest eggs. Fuzzy halves of peach fruits received ten times more eggs then shaved hairless halves. Volatiles from both almond and peach shoots induced more oviposition by females than by control stimuli. Similarly, volatiles from immature green peach fruits, mature green or mature hard-ripe peach fruits induced more oviposition than their respective control stimuli. In a choice experiment, volatiles from immature peach fruit stimulated three times more oviposition than those from soft-ripe peach fruit. Discrimination against mature soft-ripe peach fruits as potential oviposition sites may lie in the phenology of A. lineatella and host peach fruits. Larval development to the pupal stage takes 15–27 days. Therefore, any eggs laid on a ripe fruit 14 days before it falls from the tree will not likely develop into adult insects because developing larvae will only reach third or fourth instar before the fruit is decomposed, and only first and second instar larvae can overwinter. 相似文献

This study examines in vivo the role and functional interrelationships of components regulating exit from the G1 resting phase into the DNA synthetic (S) phase of the cell cycle. Our approach made use of several key experimental attributes of the developing mouse lens, namely its strong dependence on pRb in maintenance of the postmitotic state, the down-regulation of cyclins D and E and up-regulation of the p57KIP2 inhibitor in the postmitotic lens fiber cell compartment, and the ability to target transgene expression to this compartment. These attributes provide an ideal in vivo context in which to examine the consequences of forced cyclin expression and/or of loss of p57KIP2 inhibitor function in a cellular compartment that permits an accurate quantitation of cellular proliferation and apoptosis rates in situ. Here, we demonstrate that, despite substantial overlap in cyclin transgene expression levels, D-type and E cyclins exhibited clear functional differences in promoting entry into S phase. In general, forced expression of the D-type cyclins was more efficient than cyclin E in driving lens fiber cells into S phase. In the case of cyclins D1 and D2, ectopic proliferation required their enhanced nuclear localization through CDK4 coexpression. High nuclear levels of cyclin E and CDK2, while not sufficient to promote efficient exit from G1, did act synergistically with ectopic cyclin D/CDK4. The functional differences between D-type and E cyclins was most evident in the p57KIP2-deficient lens wherein cyclin D overexpression induced a rate of proliferation equivalent to that of the pRb null lens, while overexpression of cyclin E did not increase the rate of proliferation over that induced by the loss of p57KIP2 function. These in vivo analyses provide strong biological support for the prevailing view that the antecedent actions of cyclin D/CDK4 act cooperatively with cyclin E/CDK2 and antagonistically with p57KIP2 to regulate the G1/S transition in a cell type highly dependent upon pRb. 相似文献

Differential TLR recognition of leptospiral lipid A and lipopolysaccharide in murine and human cells

Leptospira interrogans is a spirochete that is responsible for leptospirosis, a zoonotic disease. This bacterium possesses an unusual LPS that has been shown to use TLR2 instead of TLR4 for signaling in human cells. The structure of its lipid A was recently deciphered. Although its overall hexa-acylated disaccharide backbone is a classical feature of all lipid A forms, the lipid A of L. interrogans is peculiar. In this article, the functional characterization of this lipid A was studied in comparison to whole parental leptospiral LPS in terms of cell activation and use of TLR in murine and human cells. Lipid A from L. interrogans did not coagulate the Limulus hemolymph. Although leptospiral lipid A activated strongly murine RAW cells, it did not activate human monocytic cells. Results obtained from stimulation of peritoneal-elicited macrophages from genetically deficient mice for TLR2 or TLR4 clearly showed that lipid A stimulated the cells through TLR4 recognition, whereas highly purified leptospiral LPS utilized TLR2 as well as TLR4. In vitro experiments with transfected human HEK293 cells confirmed that activation by lipid A occurred only through murine TLR4-MD2 but not through human TLR4-MD2, nor murine or human TLR2. Similar studies with parental leptospiral LPS showed that TLR2/TLR1 were the predominant receptors in human cells, whereas TLR2 but also TLR4 contributed to activation in murine cells. Altogether these results highlight important differences between human and mouse specificity in terms of TLR4-MD2 recognition that may have important consequences for leptospiral LPS sensing and subsequent susceptibility to leptospirosis. 相似文献

beta(2)-Glycoprotein I (beta(2)GPI) is a highly glycosylated phospholipid-binding plasma protein comprised of four complement control protein (CCP) domains and a distinct fifth domain. The structural organisation of human and bovine beta(2)GPI in aqueous solution was studied by small-angle X-ray scattering (SAXS). Low-resolution models that match the SAXS experimental data best were independently constructed by three different ab initio 3D-reconstruction algorithms. Similar elongated S-shaped models with distinct side-arms, which were correlated to the position of the carbohydrate chains, were restored from all three algorithms. Due to an additional glycosylation site located on the CCP2 domain of bovine beta(2)GPI a small change in the characteristic SAXS parameters was observed, which coincided with results obtained from SDS-PAGE. In comparison to the human analogue the corresponding restored low-resolution models displayed a similar S-shape with less bending in the middle part. As the experimental SAXS curves fit poorly to the simulated scattering curves calculated from the crystallographic coordinates of human beta(2)GPI, the crystal structure was modified. First, additional carbohydrate residues missing from the crystal structure were modelled. Second, on the basis of the low-resolution models, the J-shaped crystal structure was rotated between CCP3 and CCP2 assuming the greatest interdomain flexibility between these domains. An S-shaped model with a tilt angle of approximately 60 degrees between CCP3 and CCP2 yielded the best fit to the experimental SAXS data. Since there is evidence that beta(2)GPI can adopt different conformations, which reveal distinct differences in autoantibody recognition, our data clearly point to a reorientation of the flexible domains, which may be an essential feature for binding of autoantibodies. 相似文献

Life history theory predicts that parents should desert a reproductive attempt if the costs of rearing this brood exceed the expected benefits. Thus, if the value of the current breeding attempt is reduced, for example through an unexpected reduction in size, parents are expected to reconsider whether it is worth continuing investing in their brood. With regard to nest desertion behaviour two predictions can be made: individuals are (1) more likely to desert if the reduction in clutch size is large and (2) less likely to desert if the reduction is at a late stage of breeding. We investigated the threshold at which nest desertion takes place by experimentally reducing great tit, Parus major, clutches to different sizes and at different stages of the incubation period. The results were in accordance with the predictions: clutch desertion rates were negatively related to the number of eggs that remained in the nest, and nest desertion was less likely nearer the end of the incubation period. In addition, we estimated the fitness consequences of nest desertion behaviour. For this purpose we made one group of birds desert in favour of a replacement clutch and another group rear a reduced brood. The latter were more likely to produce a second clutch after the first-brood fledglings had left the nest. As a consequence, the number of fledglings produced over the entire breeding season did not differ between the two experimental groups. We also counted the number of recruits and breeding adults in the following breeding season and found that the experimental groups did not differ in local recruitment and adult survival.
