1. The effect of insulin (0.5, 10 and 50 munits/ml of perfusate) on glucose uptake and disposal in skeletal muscle was studied in the isolated perfused hindquarter of obese (fa/fa) and lean (Fa/Fa) Zucker rats and Osborne-Mendel rats. 2. A concentration of 0.5 munit of insulin/ml induced a significant increase in glucose uptake (approx. 2.5 mumol/min per 30 g of muscle) in lean Zucker rats and in Osborne-Mendel rats, and 10 munits of insulin/ml caused a further increase to approx. 6 mumol/min per 30 g of muscle; but 50 munits of insulin/ml had no additional stimulatory effect. In contrast, in obese Zucker rats only 10 and 50 munits of insulin/ml had a stimulatory effect on glucose uptake, the magnitude of which was decreased by 50-70% when compared with either lean control group. Since under no experimental condition tested was an accumulation of free glucose in muscle-cell water observed, the data suggest an impairment of insulin-stimulated glucose transport across the muscle-cell membrane in obese Zucker rats. 3. The intracellular disposal of glucose in skeletal muscle of obese Zucker rats was also insulin-insensitive: even at insulin concentrations that clearly stimulated glucose uptake, no effect of insulin on lactate oxidation (nor an inhibitory effect on alanine release) was observed; [14C]glucose incorporation into skeletal-muscle lipids was stimulated by 50 munits of insulin/ml, but the rate was still only 10% of that observed in lean Zucker rats. 4. The data indicate that the skeletal muscle of obese Zucker rats is insulin-resistant with respect to both glucose-transport mechanisms and intracellular pathways of glucose metabolism, such as lactate oxidation. The excessive degree of insulin-insensitivity in skeletal muscle of obese Zucker rats may represent a causal factor in the development of the glucose intolerance in this species.  相似文献

In commercial oil palm plantations in Costa Rica, we tested the hypotheses that pupation site and emergence time affect the mating success of protogynous female bagworms,Oiketicus kirbyi (Guilding) (Lepidoptera: Psychidae). Greater proportions of female than male pupae on upper leaves of oil palms and greater proportions of mated females in the upper rather than lower crown strata support the hypothesis that selection of pupation site by female larvae influences the mating success of adults. Increasing captures of males with increasing trap height further suggest that enhanced mating success of females in tree tops may be attributed either to most effective dissemination of sex pheromone on higher sites, or to males foraging predominantly in the upper strata of oil palms. As the majority of females pupated in the middle rather than upper crown of oil palms, selection of pupation site by females may be affected by additional as yet unknown factors. Emergence of females significntly preceded emergence of males. Increasing proportions of mated females throughout the emergence seasons probably resulted from an increased ‘availability’ of males. In tropical rainforests with local variations inO. kirbyi developmental time and stage, protogyny may represent an evolutionary strategy that furthers outbreeding.  相似文献

We have conducted a comprehensive study of the molecular basis of cystic fibrosis (CF) in 350 German CF patients. A screening approach based on single-strand conformation analysis and direct sequencing of genomic polymerase chain reaction products has allowed us to detect the molecular defects on 95.4% of the CF chromosomes within the coding region and splice sites of the cystic fibrosis transmembrane conductance regulator (CFTR) gene. The spectrum of sequence changes comprises 54 different mutations, including 17 missense mutations, 14 nonsense mutations, 11 frameshift mutations, 10 splice site variants and two amino acid deletions. Eleven of these mutations have not previously been described. Our results reflect the marked mutational heterogeneity of CF in a large sample of patients from a non-isolated population.  相似文献

Summary In order to facilitate the screening for the less common mutations in the cystic fibrosis (CF) gene viz., the CF transmembrane conductance regulator gene (CFTR), marker haplotypes were determined for German nonCF (N) and CF chromosomes by polymerase chain reaction analysis of four polymorphisms upstream of the CF gene (XV-2c, KM.19, MP6-D9, J44) and six intragenic polymorphisms (GATT, TUB9, M470V, T854T, TUB18, TUB20) that span the CFTR gene from exon 6 through exon 21. Novel informative sequence variants of CFTR were detected in front of exons 10 (1525-61 A or G), 19 (3601-65 C or A), and 21 (4006-200 A or G). The CF locus exhibits strong long-range marker-marker linkage disequilibrium with breakpoints of recombination between XV-2c and KM.19, and between exons 10 and 19 of CFTR. Marker alleles of GATT-TUB9 and TUB18-TUB20 were found to be in absolute linkage disequilibrium. Four major haplotypes encompass more than 90% of German N and CF chromosomes. Fifteen CFTR mutations detected on 421 out of 500 CF chromosomes were each identified on one of these four predominant 7-marker haplotypes. Whereas all analysed F508 chromosomes carried the same KM.19-D9-J44-GATT-TUB9-M470V-T854T haplotype, another frequent mutation in Germany, R553X, was identified on two different major haplotypes. Hence, a priori haplotyping cannot exclude a particular CF mutation, but in combination with population genetic data, enables mutations to be ranked by decreasing probability.  相似文献

German patients with cystic fibrosis (CF) were screened for molecular lesions in exon 13 of the cystic fibrosis transmembrane conductance regulator (CFTR) gene by single strand conformation polymorphism (SSCP) and chemical cleavage of mismatch analyses. Direct sequencing of four samples that displayed the same SSCP pattern and that were susceptible to cleavage of heteroduplexes by osmium tetroxide revealed, in all cases, a deletion of a single T residue at nucleotide position 2143 within codon 671 of the CFTR gene. As a result, leucine codon 671 is changed into a termination codon. In total, the 2143delT mutation was confirmed in 6 out of 271 German non-F508 CF chromosomes by artificial restriction fragment length polymorphism analysis, indicating that this frameshift mutation accounts for about 2% of German non-508 mutations. The 6 pancreas insufficient patients who are compound heterozygous for 2143-delT suffer from the typical features of pulmonary and gastrointestinal CF disease. The 2143delT mutation completes the panel of the more frequent CFTR mutations that reside on the F508 haplotype and that contribute to its overpresentation among German non-F508 alleles that are associated with severe forms of disease.  相似文献

Most antibodies known to interact with beta-adrenergic receptors do not exhibit subtype selectivity, nor do they provide quantitative immunoprecipitation. A monoclonal antibody, G27.1 raised against a synthetic peptide corresponding to the C-terminus of the beta 2-adrenergic receptor of hamster, is selective for the beta 2 subtype. G27.1 provides nearly quantitative immunoprecipitation of the beta 2-adrenergic receptor from hamster lung that has been photoaffinity-labeled and solubilized with sodium dodecyl sulfate. Immunoprecipitation is completely blocked by nanomolar concentrations of the immunizing peptide. This antibody interacts with beta 2-adrenergic receptors from three rodent species, but not with those from humans. When C6 glioma cells, which contain both beta 1- and beta 2-adrenergic receptors, are photoaffinity-labeled in the absence or presence of subtype-selective antagonists, subtype-selective photoaffinity-labeling results. G27.1 can immunoprecipitate beta 2-, but not beta 1-, adrenergic receptors from these cells. Similar results were obtained following subtype-selective photoaffinity-labeling of membranes from rat cerebellum and cerebral cortex. The beta-adrenergic receptors from C6 glioma cells and rat cerebral cortex exist as a mixture of two molecular weight species. These species differ in glycosylation, as shown by endoglycosidase F digestion of crude and immunoprecipitated receptors.  相似文献

Daphnids with higher density than the surrounding water body have to push water downwards to hold their position in the water column. This swimming current is mainly the result of the movement of the second antennae and only to a small extent due to the filter current. The water structures under the Daphnia are very variable depending on the swimming behavior. We focused on the hop and sink behavior, where the downward-pushed water masses form a wake which can be described as a mushroom-shaped vortex. The volume of this structure increases linearly with time. The wake volume is much larger than the zooplankton itself which can be important for the perceptibility by mechanically sensing carnivorous predators. In water with a density gradient of 10 kg m-4 the wake length WL can be described by WL = 1.64 (0.096) x L1.58(±0.14) with the length of the Daphnia L in millimeters. The wake length, measured after 5 s, remained constant for gradients up to 1 kg m-4. Above this value, the wake length declines in good agreement with the value expected from theory with WL = 10.66 (± 0.21) x (d/dz)-0.265 (± 0.010). Since the intra- and inter-specific 'communication' (e.g. mate seeking, nutrient partition between phytoplankton and bacteria, predator avoidance) can be expected to be bound to the hydrodynamic properties of the wakes, different wake forms and sizes most likely have an important impact on the information sent out.   相似文献

This article examines the health complaints of settled and nomadic Ngisonyoka Turkana of northwest Kenya. Samples of 152 nomadic and 124 settled men, aged 14 and over, were surveyed about their health status. The general pattern of disease reported concurs with previous studies of health among Turkana; that is, the primary complaints are respiratory tract infections and eye infections. The settled Turkana reported more severe complaints and higher rates of infectious disease than the nomads, including a significantly higher frequency of cold with cough, eye infection, and chest infection. Although the settled males as a group had slightly higher body mass index and other measures of body fat than the nomadic group, none of these indicators of body composition were predictive of health complaints. Observed differences in health patterns are possibly related to differences in dietary composition, exposure to pathogens associated with population density and environmental pollution, physical activity patterns, and psychosocial stress.  相似文献

A biodegradable attracticide formulation containing the pyrethroid insecticide permethrin and the sex pheromone of the ash leaf cone roller, Caloptilia fraxinella (Ely) (Lepidoptera: Gracillariidae), was tested for attractiveness and toxicity to males of this invasive pest of horticultural ash, Fraxinus spp. Marsh. (Oleaceae). Trap capture in attracticide‐baited traps was equal to capture in traps baited with rubber septa lures releasing the known attractive pheromone. Pheromone response by male C. fraxinella was not affected by the presence of permethrin as equal numbers of males were captured in traps baited with the attracticide formulation with and without permethrin. Attraction to the attracticide containing both pheromone and insecticide active ingredients did not vary with dose as 10, 50, and 100 mg droplets of the formulation attracted similar numbers of male moths in a field experiment. Equal numbers of male moths were captured in traps baited with droplets that had aged for 0–5 weeks in a laboratory fume hood before deployment in the field. The permethrin constituent of the attracticide formulation was toxic to male moths treated through tarsal contact and resulted in 73 and 100% mortality at 24 and 72 h post‐treatment, respectively. Leg autotomy occurred in over 80% of males 24 h post‐treatment to formulations containing permethrin. Mortality of moths was greater 24 h after treatment with a fresh attracticide source as compared to a 5‐week‐old source. However, mortality as a result of treatment with fresh and aged attracticide droplets was equal at 48, 72, and 96 h post‐treatment. This formulation has the potential to become a useful tool in an integrated pest‐management system for C. fraxinella on horticultural ash.  相似文献
