Ionic and gating currents from voltage-gated sodium channels were recorded in mouse neuroblastoma cells using the path-clamp technique. Displacement currents were measured from whole-cell recordings. The gating charge displaced during step depolarizations increased with the applied membrane potential and reached saturating levels above 20 mV Prolonged large depolarizations produced partial immobilization of the gating charge, and only about one third of the displaced charge was quickly reversed upon return to negative holding potentials. The activation and inactivation properties of macroscopic sodium currents were characterized by voltage-clamp analysis of large outside-out patches and the single-channel conductance was estimated from nonstationary noise analysis. The general properties of the sodium channels in mouse neuroblastoma cells are very similar to those previously reported for various preparations of invertebrate and vertebrate nerve cells. To investigate the biases associated with manta tow surveys of Acanthaster planci, counts obtained by manta-towed observers were compared with counts made on SCUBA swims under a limited range of conditions. Five 10 m wide strip transects on different parts of two reefs and with different densities of A. planci were surveyed. On average, 22.7% of starfish counted on SCUBA searches were counted on manta tows over the same transect (SD=12.0%, n=69). This proportion is termed sightability. As sightability decreases with increasing transect width, we estimate that, on average, less than 5% of the A. planci present are counted on routine manta tows which are conducted over transect of undefined width. Multiple regression analysis was used to determine which of 33 variables explained most of the variation in sightability in 10 m wide transects. The most influential variables were the proportion of cryptic starfish and an index of the degree of reef complexity. A regression equation designed to improve estimates of the abundance of A. planci on routine manta tows was developed. However, as it explained only 39% of the variation in sightability, this equation is of limited value in stabilising the negative bias associated with manta tow counts. In view of the variability of this bias, the manta tow technique is not suitable for estimating absolute densities of A. planci. Manta tow surveys are appropriate for identifying gross relative differences between densities of A. planci, and thus for determining broad-scale patterns of abundance. As such, they are a cost-effective method of estimating the geographical extent of A. planci outbreaks.  相似文献

Much debate has surrounded the notion that outbreaks of the crown-of-thorns starfish (Acanthaster planci) have occurred in the geological past and hence are natural phenomena. As this debate has recently been renewed, we have reassessed statistically data presented by Frankel (1977, 1978) as evidence for the occurrence of past outbreaks. This was done using Frankel's data as well as those from extensive starfish surveys conducted prior to the commencement of his research. Our analysis of these data indicates that the occurrence of A. planci remains in recent sediments is independent of whether or not the reef from which the sample was collected had experienced a recent outbreak. Based on this premise, it is not possible to infer from Frankel's data the occurrence of past outbreaks from similar material in much older sediments. Thus while the data presented by Frankel (1977, 1978) may show that A. planci has existed within the Great Barrier Reef for at least several thousand years it does not demonstrate that outbreaks of this starfish have occurred in the geological past.  相似文献

Molecular pathways underlying the activation of dendritic cells (DCs) in response to Herpes Simplex Virus type 1 (HSV-1) are poorly understood. Removal of the HSV virion host shut-off (vhs) protein relieves a block to DC activation observed during wild-type infection. In this study, we utilized a potent DC stimulatory HSV-1 recombinant virus lacking vhs as a tool to investigate the mechanisms involved in the activation of DCs by HSV-1. We report that the release of pro-inflammatory cytokines by conventional DC (cDC) during HSV-1 infection is triggered by both virus replication-dependent and replication-independent pathways. Interestingly, while vhs is capable of inhibiting the release of cytokines during infection of human and mouse cDCs, the secretion of cytokines by plasmacytoid DC (pDC) is not affected by vhs. These data prompted us to postulate that infection of cDCs by HSV triggers a TLR independent pathway for cDC activation that is susceptible to blockage by the vhs protein. Using cDCs isolated from mice deficient in both the TLR adaptor protein MyD88 and TLR3, we show that HSV-1 and the vhs-deleted virus can activate cDCs independently of TLR signaling. In addition, virion-associated vhs fails to block cDC activation in response to treatment with TLR agonists, but it efficiently blocked cDC activation triggered by the paramyxoviruses Sendai Virus (SeV) and Newcastle Disease Virus (NDV). This block to SeV- and NDV-induced activation of cDC resulted in elevated SeV and NDV viral gene expression indicating that infection with HSV-1 enhances the cell''s susceptibility to other pathogens through the action of vhs. Our results demonstrate for the first time that a viral protein contained in the tegument of HSV-1 can block the induction of DC activation by TLR-independent pathways of viral recognition.  相似文献

The fixation of inorganic carbon has been documented in all three domains of life and results in the biosynthesis of diverse organic compounds that support heterotrophic organisms. The primary aim of this study was to assess carbon dioxide fixation in high-temperature Fe(III)-oxide mat communities and in pure cultures of a dominant Fe(II)-oxidizing organism (Metallosphaera yellowstonensis strain MK1) originally isolated from these environments. Protein-encoding genes of the complete 3-hydroxypropionate/4-hydroxybutyrate (3-HP/4-HB) carbon dioxide fixation pathway were identified in M. yellowstonensis strain MK1. Highly similar M. yellowstonensis genes for this pathway were identified in metagenomes of replicate Fe(III)-oxide mats, as were genes for the reductive tricarboxylic acid cycle from Hydrogenobaculum spp. (Aquificales). Stable-isotope (13CO2) labeling demonstrated CO2 fixation by M. yellowstonensis strain MK1 and in ex situ assays containing live Fe(III)-oxide microbial mats. The results showed that strain MK1 fixes CO2 with a fractionation factor of ∼2.5‰. Analysis of the 13C composition of dissolved inorganic C (DIC), dissolved organic C (DOC), landscape C, and microbial mat C showed that mat C is from both DIC and non-DIC sources. An isotopic mixing model showed that biomass C contains a minimum of 42% C of DIC origin, depending on the fraction of landscape C that is present. The significance of DIC as a major carbon source for Fe(III)-oxide mat communities provides a foundation for examining microbial interactions that are dependent on the activity of autotrophic organisms (i.e., Hydrogenobaculum and Metallosphaera spp.) in simplified natural communities.  相似文献

The organosulfur compound dimethylsulfoniopropionate (DMSP) is produced by phytoplankton and is ubiquitous in the surface ocean. Once released from phytoplankton, marine bacteria degrade DMSP by either the cleavage pathway to form the volatile gas dimethylsulfide (DMS) or the demethylation pathway, yielding methanethiol (MeSH), which is readily assimilated or oxidized. The enzyme DmdB, a methylmercaptopropionate (MMPA)-coenzyme A (CoA) ligase, catalyzes the second step in the demethylation pathway and is a major regulatory point. The two forms of DmdB present in the marine roseobacter Ruegeria pomeroyi DSS-3, RPO_DmdB1 and RPO_DmdB2, and the single form in the SAR11 clade bacterium “Candidatus Pelagibacter ubique” HTCC1062, PU_DmdB1, were characterized in detail. DmdB enzymes were also examined from Ruegeria lacuscaerulensis ITI-1157, Pseudomonas aeruginosa PAO1, and Burkholderia thailandensis E264. The DmdB enzymes separated into two phylogenetic clades. All enzymes had activity with MMPA and were sensitive to inhibition by salts, but there was no correlation between the clades and substrate specificity or salt sensitivity. All Ruegeria species enzymes were inhibited by physiological concentrations (70 mM) of DMSP. However, ADP reversed the inhibition of RPO_DmdB1, suggesting that this enzyme was responsive to cellular energy charge. MMPA reversed the inhibition of RPO_DmdB2 as well as both R. lacuscaerulensis ITI-1157 DmdB enzymes, suggesting that a complex regulatory system exists in marine bacteria. In contrast, the DmdBs of the non-DMSP-metabolizing P. aeruginosa PAO1 and B. thailandensis E264 were not inhibited by DMSP, suggesting that DMSP inhibition is a specific adaptation of DmdBs from marine bacteria.  相似文献

The Mexican rice borer, Eoreuma loftini (Dyar) (Lepidoptera: Crambidae), is an economic pest of sugarcane (Saccharum spp. hybrids) and other graminaceous host crops, and it attacks grassy weeds. Oviposition preference has been known to be for plants with leaves that form folds. This study is the first to associate the nutritional quality of crop and forage plant hosts with Mexican rice borer injury. Three experiments were conducted to determine the levels of selected biochemical nutrients, including free amino acids and sugars, in four grass weeds [barnyardgrass, Echinochloa crus‐galli (L.); broadleaf signalgrass, Urochloa platyphylla (Munro ex C. Wright); johnsongrass, Sorghum halepense (L.) Pers.; and Vasey's grass, Paspalum urvillei Steud.], a forage grass [sudangrass, Sorghum bicolor (L.) Moensch spp. drummondi (Nees ex Steud.) de Wet & Harlan], three crop species [maize, Zea mays L.; sorghum, Sorghum bicolor (L.) Moench; and sugarcane, Saccharum spp.], and a sorghum‐sudangrass forage hybrid (all Poaceae). Of 16 free amino acids detected among plants in the first two experiments, only high accumulations of free histidine in sudangrass and maize were associated with increased infestations by Mexican rice borer larvae. In all three experiments, high levels of fructose were associated with heightened Mexican rice borer infestation. Ramifications of these findings on the potential dispersal of this invasive pest in the USA and possible applications of fructose in baits are discussed.  相似文献

Mortalin/GRP75, the mitochondrial heat shock protein 70, plays a role in cell protection from complement-dependent cytotoxicity (CDC). As shown here, interference with mortalin synthesis enhances sensitivity of K562 erythroleukemia cells to CDC, whereas overexpression of mortalin leads to their resistance to CDC. Quantification of the binding of the C5b-9 membrane attack complex to cells during complement activation shows an inverse correlation between C5b-9 deposition and the level of mortalin in the cell. Following transfection, mortalin-enhanced GFP (EGFP) is located primarily in mitochondria, whereas mortalinΔ51-EGFP lacking the mitochondrial targeting sequence is distributed throughout the cytoplasm. Overexpressed cytosolic mortalinΔ51-EGFP has a reduced protective capacity against CDC relative to mitochondrial mortalin-EGFP. Mortalin was previously shown by us to bind to components of the C5b-9 complex. Two functional domains of mortalin, the N-terminal ATPase domain and the C-terminal substrate-binding domain, were purified after expression in bacteria. Similar to intact mortalin, the ATPase domain, but not the substrate-binding domain, was found to bind to complement proteins C8 and C9 and to inhibit zinc-induced polymerization of C9. Binding of mortalin to complement C9 and C8 occurs through an ionic interaction that is nucleotide-sensitive. We suggest that to express its full protective effect from CDC, mortalin must first reach the mitochondria. In addition, mortalin can potentially target the C8 and C9 complement components through its ATPase domain and inhibit C5b-9 assembly and stability.  相似文献

The mechanical properties of human brain tissue are the subject of interest because of their use in understanding brain trauma and in developing therapeutic treatments and procedures. To represent the behavior of the tissue, we have developed hyperelastic mechanical models whose parameters are fitted in accordance with experimental test results. However, most studies available in the literature have fitted parameters with data of a single type of loading, such as tension, compression, or shear. Recently, Jin et al. (Journal of Biomechanics 46:2795−2801, 2013) reported data from ex vivo tests of human brain tissue under tension, compression, and shear loading using four strain rates and four different brain regions. However, they do not report parameters of energy functions that can be readily used in finite element simulations. To represent the tissue behavior for the quasi-static loading conditions, we aimed to determine the best fit of the hyperelastic parameters of the hyperfoam, Ogden, and polynomial strain energy functions available in ABAQUS for the low strain rate data, while simultaneously considering all three loading modes. We used an optimization process conducted in MATLAB, calling iteratively three finite element models developed in ABAQUS that represent the three loadings. Results showed a relatively good fit to experimental data in all loading modes using two terms in the energy functions. Values for the shear modulus obtained in this analysis (897−1653 Pa) are in the range of those presented in other studies. These energy-function parameters can be used in brain tissue simulations using finite element models.  相似文献
