Continous production of 1-kestose by β-fructofuranosidase immobilized onshirasu porous glass

Summary 1-Kestose was produced continously and selectively from 40% (w/v) sucrose solution at fast flow rate by a column packed with an immobilized -fructofuranosidase onshirasu porous glass.     

N-linked sugar chains of mouse B16 melanoma cells and their low-metastasizing variant selected by wheat germ agglutinin.

The N-linked sugar chains of metastasizing mouse B16 melanoma cells (F1) and their wheat germ agglutinin-resistant variant (Wa4-b1) showing a dramatic decrease in metastasizing and tumorigenic potentials were liberated from their membrane glycoproteins by hydrazinolysis, and their structures were comparatively analysed. The results indicated that both cell lines contain high-mannose-type and bi-, tri- and tetraantennary complex-type oligosaccharides, and their ratios are similar. However, outer chain moieties of tri- and tetraantennary oligosaccharides of the variant are extensively fucosylated, resulting in the formation of X-antigenic determinants, Gal beta 1----4(Fuc alpha 1----3)GlcNAc. Oligosaccharides containing X-antigenic determinants amounted to 71% of the total complex-type oligosaccharides. Fucosylation occurs on every N-acetyllactosamine unit and the number of the determinants ranges from one to three in triantennary oligosaccharides and one to four in tetraantennary oligosaccharides. The determinants occur predominantly in non-sialylated forms, although some are in sialylated forms. Oligosaccharides containing non-sialylated X-antigenic determinants and those containing sialylated X-antigenic determinants are approximately in the ratio 6:1. Since no significant difference was found in the extent of branching of complex-type oligosaccharides of the two cell lines, it is suggested that non-fucosylated outer chains are important for the expression of metastasizing potential by the tumour cells.     

The amino acid sequence of sarcoplasmic calcium-binding protein obtained from sandworm, Perinereis vancaurica tetradentata

Isocitrate dehydrogenase kinase and isocitrate dehydrogenase phosphatase were purified over 1000-fold from Escherichia coli ML308 by procedure involving fractionation with (NH4)2SO4 and chromatography on DEAE-cellulose, blue-dextran-Sepharose and Sephadex G150. The kinase and phosphatase activities copurified, in agreement with the observation [Laporte, D.C. and Koshland, D.E. (1982) Nature (Lond.) 300, 458-460] that a single protein bears both activities. Isocitrate dehydrogenase kinase catalysed the phosphorylation of homogeneous active isocitrate dehydrogenase with a stoichiometry of just under one phosphate group incorporated per subunit. This almost completely inactivated the dehydrogenase. There was a good correlation between phosphorylation and inactivation. Analysis of a partial acid hydrolysate of phosphorylated isocitrate dehydrogenase showed that the only phosphoamino acid present was phosphoserine. Isocitrate dehydrogenase phosphatase catalysed the release of 32P from 32P-phosphorylated isocitrate dehydrogenase; it required either ADP or ATP for activity. In the presence of ADP, or ATP plus an inhibitor of the kinase, the phosphatase catalysed full reactivation of isocitrate dehydrogenase and there was a good correlation between reactivation and the release of phosphate. In the presence of ATP alone the phosphatase catalysed the release of 32P from phosphorylated isocitrate dehydrogenase but the activity of the dehydrogenase remained low, indicating that the kinase and phosphatase were active simultaneously in these conditions. The active and inactive forms of isocitrate dehydrogenase can be resolved by non-denaturing gel electrophoresis; the two forms of the enzyme were interconverted by phosphorylation and dephosphorylation in vitro. The extent of the interconversion correlated well with the changes in isocitrate dehydrogenase activity.     

Presence of functional receptors for atrial natriuretic peptide in human pheochromocytoma

Pheochromocytoma, a catecholamine-secreting adrenomedullary tumor, has been shown to contain the functional receptor for human atrial natriuretic peptide(h-ANP). Release of catecholamines from tissue slices of pheochromocytoma was inhibited by h-ANP in a dose-dependent manner. Binding assays using 125I-ANP revealed a single class of high affinity binding sites for ANP. When covalently tagged with 125I-ANP and electrophoresed under non-reducing and reducing conditions, the receptor migrated as a 140-kDa band and a 70-kDa band, respectively, reflecting its disulfide-linked subunit structure. The presence of ANP receptor in pheochromocytoma was further demonstrated by immunohistochemistry; the tumor was positively stained with an antireceptor antiserum. The antiserum was also useful to establish the zona glomerulosa localization of ANP receptor in the normal human adrenal gland.     

Comparative study of the asparagine-linked sugar chains of human erythropoietins purified from urine and the culture medium of recombinant Chinese hamster ovary cells

The asparagine-linked sugar chains of human erythropoietin produced by recombinant Chinese hamster ovary cells and naturally occurring human urinary erythropoietin were liberated by hydrazinolysis and fractionated by paper electrophoresis, lectin affinity chromatography, and Bio-Gel P-4 column chromatography. Both erythropoietins had three asparagine-linked sugar chains in one molecule, all of which were acidic complex type. Structural analysis of them revealed that the sugar chains from both erythropoietins are quite similar except for sialyl linkage. All sugar chains of erythropoietin produced by recombinant Chinese hamster ovary cells contain only the NeuAc alpha 2----3Gal linkage, while those of human urinary erythropoietin contain the NeuAc alpha 2----6Gal linkage together with the NeuAc alpha 2----3Gal linkage. The major sugar chains were of fucosylated tetraantennary complex type with and without N-acetyllactosamine repeating units in their outer chain moieties in common, and small amounts of 2,4- and 2,6-branched triantennary and biantennary sugar chains were detected. This paper proved, for the first time, that recombinant technique can produce glycoprotein hormone whose carbohydrate structures are common to the major sugar chains of the native one.     

Construction of rat aldolase C expression plasmid and the hybrid formation between rat aldolase C and human aldolase A or B co-expressed in Escherichia coli

Rat aldolase C cDNA was inserted in an Escherichia coli expression vector to construct the rat aldolase C expression plasmid, pRAC42. This plasmid produces active rat aldolase C in the transfected E. coli host cells. The characteristics of the purified enzyme, e.g. mol. wt, electrophoretic mobilities and kinetic parameters, are indistinguishable from those of authentic rat brain aldolase C. Three different tetrameric hybrid forms, C3A, C2A2 and CA3, in addition to C4 and A4, were found to be produced in the host cell when E.coli was co-transfected with expression plasmids for rat aldolase C and for human aldolase A. Similarly, the hybrid forms, C3B, C2B2 and CB3, in addition to C4 and B4, were also produced in the cells when co-transfected with the plasmids for rat aldolase C and for human aldolase B.     

Troop composition data of wild Japanese macaques reviewed by multivariate methods

The troop composition (numbers of adult males,X ₁, adult females,X ₂, juveniles,X ₃, and infants,X ₄) of the Japanese macaque,Macaca fuscata, was examined using principal component analysis and discriminant analysis with 35 data sets from its entire distribution range.X ₂,X ₃ andX ₄ showed an equally high, positive correlation with one another. The variations of the troop composition variables were reinterpreted by a component representing the troop “size” and those representing “shape.” The data sets were sorted into three habitat zone groups from north to south. The functions discriminating between the habitat zone groups indicated thatX ₂ andX ₄ largely suffice for the discrimination. Examination ofX ₂ andX ₄ revealed that the troops in the south have a greaterX ₄/X ₂ ratio; however, further examination of this result indicated a relatively high offspring/female ratio only in the disturbed middle habitat zone but no conclusive latitudinal difference of birth rate. The results were discussed in relation to socioecology of the species. Order of authorship determined by a flip of a coin.     

Antibodies against type II phospholipase A2 prevent renal injury due to ischemia and reperfusion in rats

This study was performed to determine the involvement of type II phospholipase A₂ (PLA₂-II) in renal injury caused by ischemia and reperfusion. Ischemia and reperfusion significantly elevated levels of blood urea nitrogen and serum creatinine in rats. These increases were significantly reduced by i.v. administration of rabbit IgG F(ab′)2 fragments against rat PLA₂-II. Increased levels of acid-stable PLA₂ activity in the kidney were caused by ischemia and reperfusion, and were suppressed by administration of anti-PLA₂-II F(ab′)2. Increased levels of myeloperoxidase activity, a marker of neutrophil infiltration, in the kidney were also reduced after anti-PLA₂-II F(ab′)2 treatment. These results suggest that PLA₂-II plays a pivotal role in pathogenesis of ischemia and reperfusion injury through induction of neutrophil infiltration.     

Effect of hypertension on fibronectin expression in the rat aorta

Interactions between extracellular fibronectin and vascular cells are thought to influence the phenotype of those cells. To determine if changes in fibronectin expression accompany the phenotypic changes of vascular tissue characteristic of experimental hypertension, steady state mRNA levels for fibronectin were determined in aortae of normotensive and hypertensive rats. A 3-6-fold increase in fibronectin mRNA was observed in aortic tissue of hypertensive rats following 3 weeks of treatment with deoxycorticosterone and salt, whereas if rats were treated only with deoxycorticosterone or salt alone, no changes occurred. The changes were reversed by normalization of blood pressure. The increases observed were localized to aorta and not to the periaortic tissue. Angiotensin II infusion using osmotic minipumps also caused an increase in fibronectin expression. Age-dependent increases in aortic fibronectin mRNA occurred in several rat strains, and the combined effects of hypertension and aging were greater than either variable alone. A clear distinction between the expression of fibronectin mRNA and that for collagen or tropoelastin were found in hypertensive and aging models. Aortic fibronectin was also increased in the hypertensive rats as determined by Western blot analysis. The findings indicate that elevation in blood pressure increases fibronectin expression in rat aorta and suggest that such changes may influence the aortic cellular responses to hypertension.     

Aggression toward large carnivores by wild chimpanzees of Mahale Mountains National Park, Tanzania

In the Mahale Mountains National Park of Tanzania, a group of about 33 chimpanzees were observed to surround a leopard den containing a mother and at least one cub and to drag out and kill the cub. This is the first report of chimpanzees or any other primate species killing their potential predator's offspring. The incident suggests that chimpanzees, without any weapons, can manage to defend themselves against a carnivore of at least up to leopard size, and implicates how the early hominids may have reacted against their potential predators.
