We herein describe a novel procedure for dentin regeneration that mimics the biological processes of tooth development in nature. The canonical Wnt signaling pathway is an important regulator of the Dentin sialophosphoprotein (Dspp) expression. Our approach mimics the biological processes underlying tooth development in nature and focuses on the activation of canonical Wnt signaling to trigger the natural process of dentinogenesis. The coronal portion of the dentin and the underlying pulp was removed from the first molars. We applied lithium chloride (LiCl), an activator of canonical Wnt signaling, on the amputated pulp surface to achieve transdifferentiation toward odontoblasts from the surrounding pulpal cells. MicroCT and microscopic analyses demonstrated that the topical application of LiCl induced dentin repair, including the formation of a complete dentin bridge. LiCl-induced dentin is a tubular dentin in which the pulp cells are not embedded within the matrix, as in primary dentin. In contrast, a dentin bridge was not induced in the control group treated with pulp capping with material carriers alone, although osteodentin without tubular formation was induced at a comparatively deeper position from the pulp exposure site. We also evaluated the influence of LiCl on differentiation toward odontoblasts in vitro. In the mDP odontoblast cell line, LiCl activated the mRNA expression of Dspp, Axin2 and Kallikrein 4 (Klk4) and downregulated the Osteopontin (Osp) expression. These results provide a scientific basis for the biomimetic regeneration of dentin using LiCl as a new capping material to activate dentine regeneration.  相似文献

Seven flavonol glycosides were isolated from the leaves ofT. apetalon. They were identified chromatographically and spectrally to be: quercetin/kaempferol 3-O-α-arabinopyranosyl-(1→6)-β-galactopyranoside (TQ and TK), quercetin/kaempferol 3-O-[2‴-O-acetyl-α-arabinopyranosyl]-(1→6)-β-galactopyranoside (TAQ and TAK), quercetin 3-O-β-glucoside (ISQ), isorhamnetin 3-O-α-arabinopyranosyl-(1→6)-β-galactopyranoside (TI) and isorhamnetin 3-O-[2‴-O-acetyl-α-arabinopyranosyl]-(1→6)-β-galactopyranoside (TAI). TQ, TAQ, TI and TAI were major constituents. This is the first report on two new isorhamnetin-type glycosides, TI and TAI. The seven flavonol glycosides identical to those ofT. apetalon were isolated and identified in the leaves ofT. kamtschaticum; TQ and TAQ were also major components, but TI and TAI were only minor components. TI and TAI were not detected in the leaves ofT. tschonoskii. These leaf-flavonoid patterns were discussed from a chemosystematic point of view. An advanced glycation end products (AGE)/a receptor for AGE (RAGE) axis plays a key role in diabetic vascular complications. Membrane type 1-matrix metalloproteinase (MT1-MMP) has been shown to function not only as a proteolytic enzyme but also as a signaling molecule. In this study, we investigated the role of MT1-MMP in the AGE/RAGE-triggered signaling pathways in cultured rabbit smooth muscle cells (SMCs) and the molecular interaction between RAGE and MT1-MMP in vitro and in vivo. In SMCs, AGE-activated Rac1 and p47(phox) within 1 min, NADPH oxidase activity and reactive oxygen species (ROS) generation within 5 min, and NF-κB phosphorylation within 15 min, thereby inducing redox-sensitive molecular expression. Silencing of RAGE by small-interfering RNA (siRNA) blocked the AGE-induced signaling pathways. AGE-induced geranylgeranyl transferase I (GGTase I) activity, Rac1·p47(phox) activation, NADPH oxidase activity, ROS generation, and molecular expression were also markedly attenuated by silencing of MT1-MMP. An inhibitor of GGTase I mimicked the effects of MT1-MMP-specific siRNA. Fluorescent immunohistochemistry revealed that MT1-MMP was partially co-localized with RAGE in SMCs, and RAGE was found to form a complex with MT1-MMP in both cultured SMCs and the aortae of diabetic rats by immunoprecipitation. Furthermore, MT1-MMP and RAGE formed a complex in the aortic atherosclerotic lesions of hyperlipidemic rabbits. We show that MT1-MMP plays a crucial role in RAGE-activated NADPH oxidase-dependent signaling pathways and forms a complex with RAGE in the vasculature, thus suggesting that MT1-MMP may be a novel therapeutic target for diabetic vascular complications.  相似文献

We developed a numerical simulation of mating experiment to study effects of phototactic gamete behavior and density on fertilization success, using the C++ programming language, and pseudo-parallelization methods with input parameters based on experimental data. In our experiments, we found that gametes with positive phototaxis are favored, particularly in shallow water, because they can search for potential mates on the two-dimensional (2-D) water surface rather than randomly in three dimensions. We also found evidence that sperm (male gametes) limitation might not be the dominant selective force in the evolution of isogamous or slightly anisogamous marine green algae because almost all of female gametes can be fertilized on the 2-D water surface meaning they might not be under sperm limited conditions. Gamete density also appears to affect mating success seriously. These findings were produced by some technical progress made recently to rapidly and correctly count the numbers of zygotes formed calculating the locations of huge numbers of male and female gametes in the test tank. Both gamete behavior and density might be determined by environmental conditions of habitat, particularly the depth of water.  相似文献

Melanin-concentrating hormone receptor 1 (MCHR1) antagonists have been studied as potential agents for the treatment of obesity. Initial structure-activity relationship studies of in-house hit compound 1a and subsequent optimization studies resulted in the identification of tetrahydroisoquinoline derivative 23, 1-(2-acetyl-1,2,3,4-tetrahydroisoquinolin-7-yl)-4-[4-(4-chlorophenyl)piperidin-1-yl]butan-1-one, as a potent hMCHR1 antagonist. A homology model of hMCHR1 suggests that these compounds interact with Asn 294 and Asp 123 in the binding site of hMCHR1 to enhance binding affinity. Oral administration of compound 23 dose-dependently reduced food intake in diet-induced obesity (DIO)-F344 rats.  相似文献

In this paper, we investigate structured population model of marine invertebrate whose life stage is composed of sessile adults and pelagic larvae, such as barnacles contained in a local habitat. First we formulate the basic model as an Cauchy problem on a Banach space to discuss the existence and uniqueness of non-negative solution. Next we define the basic reproduction number R0 to formulate the invasion condition under which the larvae can successfully settle down in the completely vacant habitat. Subsequently we examine existence and stability of steady states. We show that the trivial steady state is globally asymptotically stable if R0 < or = 1, whereas it is unstable if R0 > 1. Furthermore, we show that a positive (non-trivial) steady state uniquely exists if R0 > 1 and it is locally asymptotically stable as far as absolute value of R0 - 1 is small enough.  相似文献

We used PCR-based cDNA subtraction to screen for genes up-regulated during mouse hair morphogenesis. One gene selected was predominantly expressed at the tip of developing hair follicles and encoded a protein characterized by the presence of twelve tandem repeats of approximately 120 amino acids and a novel N-terminal domain containing an Arg-Gly-Asp cell-adhesive motif. Immunohistochemistry demonstrated that the protein encoded by this gene, named QBRICK, was localized at the basement membrane zone of embryonic epidermis and hair follicles, in which it was more enriched at the tip rather than the stalk region. Cell adhesion assays showed that QBRICK was active in mediating cell-substratum adhesion through integrins containing alphav or alpha8 chain, but not integrin alpha5beta1. Immunohistochemistry showed that QBRICK colocalized with alphav-containing integrins in the interfollicular region, but with the alpha8-containing integrin at the tip region of developing hair follicles. These results, together, indicate that QBRICK is an adhesive ligand of basement membrane distinctively recognized by cells in the embryonic skin and hair follicles through different types of integrins directed to the Arg-Gly-Asp motif.  相似文献
