The role of melanization and cuticular lipids in water conservation has been studied in many Drosophila species (Diptera: Drosophilidae). Nevertheless, a comparative approach to larval and adult stages of ecologically diverse, wild Drosophila species is still required. Based upon abdominal cuticular melanization patterns, wild‐caught Drosophila species were categorized as (1) melanic, (2) fixed‐melanic, or (3) non‐melanic. At the interspecific level, the ecological significance of melanization and cuticular lipids was determined by the inverse association of melanization and cuticular water loss in melanic species, and of cuticular lipids and cuticular water loss in fixed‐melanic and non‐melanic species. Interestingly, higher amounts of cuticular lipids were also evident in fixed as well as non‐melanic species, as compared to melanic species at larval stages, which is consistent with their differences in reduced water loss rates. Moreover, fixed‐melanic and non‐melanic species exhibited comparatively higher (ca. 1.8–2.0 fold) desiccation resistance. Thus, cuticular lipids provide a better waterproofing mechanism than melanization. Furthermore, acclimation to dehydration stress in adults improved desiccation resistance in melanic species, whereas such effects were lacking in fixed‐melanic and non‐melanic species. However, there were no changes in cuticular components as a consequence of desiccation acclimation. Thus, our results indicate that melanic, fixed‐melanic, and non‐melanic Drosophila species differ in the evolved physiological mechanisms of water conservation to adapt to dry conditions.  相似文献

Aims

Increasing evidence indicates that plant responses to ions (uptake/transport, inhibition, and alleviation of inhibition) are dependent upon ion activities at the outer surface of root-cell plasma membranes (PMs) rather than activities in the bulk-phase rooting medium.

Methods

A web-accessible computer program was written to calculate the electrical potential (ψ) at the outer surface of root-cell PMs (ψ PM). From these values of ψ PM, activities of ion I with charge Z ({I Z}) can be calculated for the outer surface of the PM ({I Z}PM). In addition, ψ and {I Z} in the Donnan phase of the cell walls (ψ CW and {I Z}CW) can be calculated.

Results

By reanalysing published data, we illustrate how this computer program can assist in the investigation of plant-ion interactions. For example, we demonstrate that in saline solutions, both Ca deficiency and Na uptake are more closely related to {Ca2+}PM and {Na+}PM than to {Ca2+}b and {Na+}b (activities in the bulk-phase media). Additional examples are given for Zn and P nutrition, Ni toxicity, and arsenate uptake.

Conclusions

The computer program presented here should assist others to develop an electrostatic view of plant-ion interactions and to re-evaluate some commonly-held views regarding mechanisms of ion transport, toxicity, competition among ions, and other phenomena.  相似文献

The recessive labile locus mapped on chromosome 5HL causes irregular spikelet fertility and controls floret development as well as row-type in barley.

Abstract

The labile-barley displays a variable number of fertile spikelets at each rachis internode (0–3 fertile spikelets/rachis internode) which is intermediate between that observed in two- or six-rowed types. Previous re-sequencing of Vrs1 in 219 labile-barley (Hordeum vulgare L. convar. labile) accessions showed that all carried a six-rowed specific allele. We therefore hypothesized that this seemingly random reduction in spikelet fertility is most likely caused by the labile (lab) locus, which we aimed to phenotypically and genetically define. Here, we report a detailed phenotypic analysis of spikelet fertility in labile-barleys in comparison to two- and six-rowed genotypes using scanning electron microscopy analysis. We found that the first visible morphological deviation occurred during the stamen primordium stage, when we regularly observed the appearance of arrested central floral primordia in labile but not in two- or six-rowed barleys. At late stamen and early awn primordium stages, lateral florets in two-rowed and only some in labile-barley showed retarded development and reduction in size compared with fully fertile lateral florets in six-rowed barley. We used two F2 mapping populations to generate whole genome genetic linkage maps and ultimately locate the lab locus as a recessive Mendelian trait to a 4.5–5.8 cM interval at approximately 80 cM on chromosome 5HL. Our results will help identifying the role of the lab gene in relation to other spikelet fertility factors in barley.  相似文献

Transferring the knowledge bases between related species may assist in enlarging the yield potential of crop plants. Being cereals, rice and wheat share a high level of gene conservation; however, they differ at metabolic levels as a part of the environmental adaptation resulting in different yield capacities. This review focuses on the current understanding of genetic and molecular regulation of yield-associated traits in both crop species, highlights the similarities and differences and presents the putative knowledge gaps. We focus on the traits associated with phenology, photosynthesis, and assimilate partitioning and lodging resistance; the most important drivers of yield potential. Currently, there are large knowledge gaps in the genetic and molecular control of such major biological processes that can be filled in a translational biology approach in transferring genomics and genetics informations between rice and wheat.  相似文献

Abstract

The global effort to identify new sources of resistance to wheat stem rust, caused by Puccinia graminis f. sp. tritici race group Ug99 has resulted in numerous studies reporting both qualitative genes and quantitative trait loci. The purpose of our study was to assemble all available information on loci associated with stem rust resistance from 21 recent studies on Triticum aestivum L. (bread wheat) and Triticum turgidum subsp. durum desf. (durum wheat). The software LPmerge was used to construct a stem rust resistance loci consensus wheat map with 1,433 markers incorporating Single Nucleotide Polymorphism, Diversity Arrays Technology, Genotyping-by-Sequencing as well as Simple Sequence Repeat marker information. Most of the markers associated with stem rust resistance have been identified in more than one population. Several loci identified in these populations map to the same regions with known Sr genes including Sr2, SrND643, Sr25 and Sr57 (Lr34/Yr18/Pm38), while other significant markers were located in chromosome regions where no Sr genes have been previously reported. This consensus map provides a comprehensive source of information on 141 stem rust resistance loci conferring resistance to stem rust Ug99 as well as linked markers for use in marker-assisted selection.  相似文献

In the current study, an attempt was made to encapsulate assembly pheromone using natural polymer, chitosan. Chitosan beads were prepared by incorporating assembly pheromone in conjunction with an acaricide, namely, deltamethrin. In the in vitro bioassay, the test beads attracted and killed 79 % of unfed larvae, 88 % of unfed nymphs and 61 % of unfed adults of the brown dog tick, Rhipicephalus sanguineus, in 24 h of exposure. Field trials were carried out to attract and kill the pre-parasitic environmental stages. The beads were dispersed onto specially designed devices and they were placed in infested kennels. D-Ribitol-5-phosphate (Rbt-5-P) is an important metabolite in the pentose phosphate pathway and an integral part of bacterial cell wall polysaccharides, specifically as polyribosyl ribitol phosphate (PRP) in Haemophilus influenzae type b (Hib). The major objective of this study was to investigate whether an antibody specific to Rbt-5-P can recognize the PRP of Hib. D-Ribose-5-phosphate was reacted with proteins in the presence of sodium cyanoborohydride to obtain Rbt-5-P epitopes; 120 h reaction resulted in conjugation of ~30 and ~17 moles of Rbt-5-P/mole of BSA and OVA, respectively, based on decrease in amino groups, MALDI–TOF analyses, an increase in apparent molecular weight (SDS–PAGE) and glycoprotein staining. Immunization of rabbits with Rbt-5-P–BSA conjugate generated antibodies to Rbt-5-P as demonstrated by dot immunoblot and non-competitive ELISA. Homogeneous Rbt-5-P-specific antibody was purified from Rbt-5-P–BSA antiserum subjected to caprylic acid precipitation followed by hapten–affinity chromatography; its affinity constant is 7.1?×?108 M?1. Rbt-5-P antibody showed 100 % specificity to Rbt-5-P, ~230 %, 10 % and 3.4 % cross-reactivity to FMN, riboflavin and FAD, respectively; the antibody showed ~4 % cross-reactivity to D-ribitol and <3 % to other sugars/sugar alcohols. Rbt-5-P–specific antibody recognized Hib conjugate vaccines containing PRP which was inhibited specifically by Rbt-5-P, and also detected Hib cell-surface capsular polysaccharides by immunofluorescence. In conclusion, Rbt-5-P–protein conjugate used as an immunogen elicited antibodies binding to an epitope also present in PRP and Hib bacteria. Rbt-5-P–specific antibody has potential applications in the detection and quantification of free/bound Rbt-5-P and FMN as well as immunological recognition of Hib bacteria and its capsular polysaccharide.  相似文献

The marine red alga Gracilaria crassa was investigated for its proximate composition, minerals, fatty acids, amino acids, and agar content to decipher its nutritional implications. The growth performance and pigments were studied under different combinations of temperature and salinity. On a dry weight basis the total lipid content was 1.30?±?0.05 %, protein was 5.18?±?0.64 %, carbohydrate was 42.0?±?1.2 %, ash was 43.18?±?1.15 %, and agar content was 21.52?±?0.73 %. Appreciable amounts of macro-, micro-nutrients (K?>?Na, Ca, Mg, and Fe), and essential amino acids (Ileu, His, Thr, Leu, and Lys) were found. Palmitic, stearic acid, and arachidonic acid were major fatty acids detected. The alga showed maximum daily growth rate (DGR %) 5.8?±?0.09 % at 25 °C, 35 ‰ salinity. The highest content of pigment R-phycoerythrin (444.7?±?1.9 μg g?1 fresh weight (FW) basis) was obtained at 25 ‰ salinity at 35 °C while that of R-phycocyanin (476.3?±?2.3 μg g?1 DW) at 30 ‰ salinity at 30 °C. This study revealed that this alga can be utilized as a potential source for food and feed. The data generated on best growth conditions will be very useful for farming of G. crassa in open sea. This alga could be used for production of natural colorants at defined control condition.  相似文献
