This study is the first report to suggest a morphological phylogenetic framework for the seven varieties of Ficus deltoidea Jack (Ficus: Moraceae) from the Malay Peninsula of Malaysia. Several molecular-based classifications on the genus Ficus had been proposed, but neither had discussed the relationship between seven varieties of F. deltoidea to its allies nor within the varieties. The relationship between seven varieties of F. deltoidea is still debated due to the extreme morphological variabilities and ambiguous boundaries between taxa. Thus, the correct identification of these varieties is important as several morphological characters are variety-specific. To test the monophyly and further resolved the relationship in F. deltoidea, a morphological phylogenetic analysis was conducted based on herbarium specimens representing the seven varieties of F. deltoidea that were collected from the Malay Peninsula of Malaysia, by using related species of the genus Ficus; F. grossularioides, F. ischnopoda and F. oleifolia as the outgroups. Parsimony and neighbour-joining analyses indicated that F. deltoidea is monophyletic, in that the seven varieties of F. deltoidea nested into two clades; clade subspecies deltoidea (var. deltoidea, var. bilobata, var. angustifolia, var. kunstleri and var. trengganuensis) and clade subspecies motleyana (var. intermedia and var. motleyana). 相似文献

Gastrulating chick embryos in culture were treated with ethanol solutions, following which the mesoderm cells migrating from the primitive streak were examined by scanning electron microscopy. Morphometric analysis of cell shape showed that mesoderm cells from treated embryos were significantly more rounded and therefore less well spread than controls, and showed fewer filopodial contacts with the overlying basement membrane. This result was only obtainable for cells leading the migration from the primitive streak, since the following cells in the mesodermal mass apparently did not show this difference. The ethanol concentration required to obtain a reliable effect was 5%, while lower concentrations produced highly variable results. The mesoderm cells were also examined for their in vitro responsiveness to ethanol by investigating their adhesiveness and cytoskeleton. No effect was observed on cell-glass adhesion as judged by interference reflection microscopy using up to 1% ethanol. This concentration did, however, disrupt the actin cytoskeleton of cultured cells when stained with NBD-phallacidin, but lower concentrations were ineffective. Olfactory bulb (OB) projection neurons receive sensory input from olfactory receptor neurons and precisely relay it through their axons to the olfactory cortex. Thus, olfactory bulb axonal tracts play an important role in relaying information to the higher order of olfactory structures in the brain. Several classes of axon guidance molecules influence the pathfinding of the olfactory bulb axons. Draxin, a recently identified novel class of repulsive axon guidance protein, is essential for the formation of forebrain commissures and can mediate repulsion of diverse classes of neurons from chickens and mice. In this study, we have investigated the draxin expression pattern in the mouse telencephalon and its guidance functions for OB axonal projection to the telencephalon. We have found that draxin is expressed in the neocortex and septum at E13 and E17.5 when OB projection neurons form the lateral olfactory tract (LOT) rostrocaudally along the ventrolateral side of the telencephalon. Draxin inhibits axonal outgrowth from olfactory bulb explants in vitro and draxin-binding activity in the LOT axons in vivo is detected. The LOT develops normally in draxin−/− mice despite subtle defasciculation in the tract of these mutants. These results suggest that draxin functions as an inhibitory guidance cue for OB axons and indicate its contribution to the formation of the LOT. 相似文献

The effect of inoculation with Pythium aphanidermatum was studied on total phenol (TP), salicylic acid (SA), chlorophylls and carotenoid contents of leaves and plant growth characteristics of five tobacco cultivars, namely RK-10 P3, RK-12 P3, RK-13 P4, RK-18 P8 and RK-26 P3, to assess cultivar response at biochemical and morphological levels. Root rot measured at 0–5 scale was 2.66 on cv. RK-10 P3, followed by 2. 33 on cv. RK-18 P8, 1.33 on cv. RK-26 P3 and 1.0 on cv. RK-13 P4. The cv. RK-12 P3 did not develop measurable root rot. The rhizosphere population of root rot fungus increased over time, being highest on the cv. RK-10 P3 (P ≤ 0.001), followed by cvs. RK-18 P8, RK-26 P3, RK-13 P4 and RK-12 P3. Inoculation with the fungus resulted in 5% (cv. RK-10 P3), 10.3% (cv. RK-18 P8, P ≤ 0.05), 10.9% (cv. RK-26 P3, P ≤ 0.05), 16.4% (cv. RK-13 P4, P ≤ 0.01) and 41.5% (cv. RK-12 P3, P ≤ 0.001) increase in the TP content of leaves. SA concentration in tobacco leaves increased marginally (0.8%–3%) in cvs. RK-10 P3, RK-18 P8 and RK-26 P3, but considerably (16%–17%, P ≤ 0.01) in cv. RK-13 and RK-12 P3 in comparison to uninoculated plants. Total chlorophyll content of leaves in response to inoculation with P. aphanidermatum decreased by 27% and 23% in tobacco cvs. RK-10 P3 and RK-18 P8 (P ≤ 0.001) and 17.6 (P ≤ 0.01) and 10.6% (P ≤ 0.05) in cv. RK-26 P3 and RK-13 P4, respectively. Reduction in chlorophylls a and b was 20% and 15% in cv. RK-10 P3 and 20% and 11% in cv. RK-18 P8. Total carotenoid contents of tobacco leaves decreased significantly in cvs. RK-10 P3 and RK-18 P8 (P ≤ 0.05). Significant and greater decrease in plant growth variables was recorded in the cultivars in which increase in TP and SA was lower and decrease in chlorophyll and carotenoids was greater. This study has revealed that greater synthesis of TP and SA may provide resistance in tobacco plants against P. aphanidermatum. The cv. RK-12 P3, in which greatest increase in the SA (17%) and TP (41.5%) was recorded, did not exhibit a significant decrease in plant growth variables and leaf pigments (P ≤ 0.05). 相似文献

Verbascum species are widely used in folk medicine because of their broad range of biological activities. Harpagoside, an iridoid glycoside isolated from some Verbascum plants is known for its anti-inflammatory properties. In the present study, the effect of five extracts of Verbascum species and harpagosides were evaluated in mouse models of acute and chronic inflammation. The results demonstrate that Verbascum phoeniceum extract strongly inhibits COX-1 (60.2% inhibition vs PMA-stimulated cells) and COX-2 (44.8% inhibition) expression stimulated peritoneal macrophages resulting in reduced paw swelling in carrageenan-induced oedema (55.5% inhibition vs PBS-treated mice). Harpagoside ameliorated the development of zymosaninduced arthritis and reduced pathological changes in joints as shown by the decreased histological score for cell infiltration in synovial cavity (3.5±0.2 in vs 2.0±0.16), cartilage loss (2.5±0.3 vs 1.8±0.5) and bone resorption (2.4±0.2 vs 1.8±0.4). Molecular docking simulations of harpagoside suggest that it may function with increased specific affinity towards COX-1 than COX-2. The potential of harpagoside to be applied as an effective agent for treating joint-related disorders is discussed. 相似文献
