Acetylcholinesterase (AChE) inhibitor donepezil is widely used for the treatment of Alzheimer’s disease (AD). The mechanisms of therapeutic effects of the drug are not well understood. The ability of donepezil to reverse a known pathogenic effect of β-amyloid peptide (Abeta), namely, the impairment of hippocampal long-term potentiation (LTP), was not studied yet. The goal of the present study was to study the influence of donepezil in 0.1–10 μM concentrations on control and Abeta-impaired hippocampal LTP. Possible involvement of N-methyl-d-aspartate receptors (NMDARs) into mechanisms of donepezil action was also studied. LTP of population spike (PS) was studied in the CA1 region of rat hippocampal slices. Change of LTP by donepezil treatment had a bell-shaped dose–response curve. The drug in concentrations of 0.1 and 1 μM did not change LTP while in concentration of 0.5 μM significantly increased it, and in concentration of 5 and 10 μM suppressed LTP partially or completely. Abeta (200 nM) markedly suppressed LTP. Addition of 0.1, 0.5 or 1 μM donepezil to Abeta solution caused a restoration of LTP. N-methyl-d-aspartate (NMDA) currents were studied in acutely isolated pyramidal neurons from CA1 region of rat hippocampus. Neither Abeta, nor 0.5 μM donepezil were found to change NMDA currents, while 10 μM donepezil rapidly and reversibly depressed it. Results suggest that donepezil augments control and impaired by Abeta hippocampal LTP in NMDAR-independent manner. In general, our findings extend the understanding of mechanisms of therapeutic action of donepezil, especially at an early stage of AD, and maybe taken into account while considering the possibility of donepezil overdose.  相似文献

Artificial ribonucleases, conjugates of short oligodeoxyribonucleotides and peptides built of arginine, leucine, proline, and serine, were synthesized and assessed in terms of ribonuclease activity and specificity of RNA cleavage. A specific group of the conjugates was identified that display T1-ribonuclease-like activity and cleave RNA predominantly at G-X sequences. Circular dichroism study of the structures of the most active conjugates, free peptide (LR)4G, and oligonucleotides revealed that conjugation of oligonucleotide to the peptide results in a specific peptide folding that possibly provides ribonuclease activity to the conjugate.  相似文献

The aim of the present study is to test by molecular DNA data a hypothesis concerned to speciation by allopatry occurring in Mimagoniates microlepis, associated to the Serra do Mar mountain chain (Atlantic Rain Forest hotspot) in Southern Brazil. Overall genetic diversity and mean genetic distances were high, demonstrating both good conservation status and genetic differentiation. Neighbor-Joining (NJ) and parsimony analyses, together with population genetic parameters (ΦST, Nm, GST, and AMOVA), identified two main vicariant genetic/evolutionary stocks dividing the upper Iguaçu River samples from those of the coastal plains. Other well-supported intrinsic monophyletic clades were also identified, suggesting fast and remarkable speciation processes. In addition, the genetic, evolutionary, geographic, and phylogeographic evidences reinforced an occurring species complex. Moreover, these evolutionarily significant units (species complex) seem to be inside four natural biogeographic areas. Thus, the genesis and evolution of the Serra do Mar complex might be associated to diversification processes of M. microlepis. Such a consideration suggests that the areas including the upper Iguaçu River and the coastal plains of the states of São Paulo, Paraná, and Santa Catarina require distinct conservation policies involving one of the global biodiversity hotspots, namely, the Brazilian Atlantic Rain Forest.  相似文献

We studied fatty acid (FA) composition of littoral microalgae in the fast-flowing oligotrophic river, the Yenisei, Siberia, monthly for 3 years. Seasonal dynamics of species composition had similar patterns in all the studied years. In springs, a pronounced dominance of filamentous green algae occurred, in summer and autumn diatoms were abundant, and in late autumn and winter epilithic biofilms consisted primarily of cyanobacteria and detritus. In general, FA composition of the algal periphytic community was dominated by 16:0, 16:1ω7, 20:5ω3, 14:0, and 18:3ω3 throughout the studied period. Several groups of FAs, which had peculiar seasonal dynamics, were differentiated by statistical analysis based on a method of correlation graphs. The seasonal changes in FA composition could be partly explained by the seasonal succession of species composition of the community. Besides, we found that populations of both diatom and green algae grown in summer at a higher water temperature were lower in polyunsaturated fatty acids than those in spring, at a lower temperature. Hence, we suppose that the regular seasonal dynamics of FA composition of the studied littoral microalgae was driven both by changes in species composition and by temperature adaptations of the algal populations. The highest content of essential polyunsaturated FAs, eicosapentaenoic and docosahexaenoic acids, in the spring “psychrophilic” populations of diatoms could make them of the higher nutritive value for zoobenthic primary consumers.  相似文献

We studied the kinetics of NADPH-dependent reduction of human CYP3A4 incorporated into Nanodiscs (CYP3A4-ND) and proteoliposomes in order to probe the effect of P450 oligomerization on its reduction. The flavin domain of cytochrome P450-BM3 (BMR) was used as a model electron donor partner. Unlike CYP3A4 oligomers, where only 50% of the enzyme was shown to be reducible by BMR, CYP3A4-ND could be reduced almost completely. High reducibility was also observed in proteoliposomes with a high lipid-to-protein ratio (L/P = 910), where the oligomerization equilibrium is displaced towards monomers. In contrast, the reducibililty in proteoliposomes with L/P = 76 did not exceed 55 ± 6%. The effect of the surface density of CYP3A4 in proteoliposomes on the oligomerization equilibrium was confirmed with a FRET-based assay employing a cysteine-depleted mutant labeled on Cys-468 with BODIPY iodoacetamide. These results confirm a pivotal role of CYP3A4 oligomerization in its functional heterogeneity. Furthermore, the investigation of the initial phase of the kinetics of CYP3A4 reduction showed that the addition of NADPH causes a rapid low-to-high-spin transition in the CYP3A4-BMR complex, which is followed by a partial slower reversal. This observation reveals a mechanism whereby the CYP3A4 spin equilibrium is modulated by the redox state of the bound flavoprotein.  相似文献

An increase of xenodiversity in plankton and benthos in the eastern Gulf of Finland was observed from 1998 to 2004. Nonindigenous species account for 4.8% of all species found and up to 96% of total biomass. Invasive benthic omnivores, the alien amphipods Gmelinoides fasciatus and Pontogammarus robustoides and the predaceous fish Perccottus glenii with their versatile diets strongly affect the community structure. Invasive sessile seston-feeders that directly (through grazing and water clearance) and indirectly (through recycling of nutrients) interact with other ecosystem components, are mainly represented by the zebra mussel Dreissena polymorpha, which affect the structure of benthic and planktonic communities as well as benthic-pelagic coupling. The invasive predatory cladocerans Cercopagis pengoi and Evadne anonyx and larvae of D. polymorpha are only temporary components in the zooplankton, which is limiting their overall effect. Alien benthic bioturbators, the polychaetes Marenzelleria neglecta and the oligochaete Tubificoides pseudogaster account for a high proportion of total abundance and biomass but their effects on native species need further research.  相似文献

The aim of the study was to advance our knowledge regarding the activation process of the ventricular myocardium in birds in which Purkinje fibres penetrate into the ventricular wall to reach the epicardium. A depolarization pattern of the left ventricular free wall was studied in chickens (Gallus gallus) during ventricular paced beats. Duration of the activation process of the left ventricular free wall is significantly increased during ventricular ectopic excitation as compared with sinus rhythm. Its lowest increase occurs during subendocardial pacing of the middle part of the left ventricle, but its greatest increase is observed during subepicardial pacing of the left ventricular base. Multifocality and mosaicity of depolarization of the left ventricular free wall myocardium in chicken are expressed in a considerably less degree during ventricular paced beats in comparison with sinus rhythm. During ventricular paced beats, excitation of the left ventricular free wall is mostly due to the successive spreading of the depolarization wave from pacing sites.  相似文献

Cyclometalated ruthenium(II) complexes, [Ru(II)(C~N)(N~N)(2)]PF(6) [HC~N=2-phenylpyridine (Hphpy) or 2-(4'-tolyl)pyridine; N~N=2,2'-bipyridine, 1,10-phenanthroline, or 4,4'-dimethyl-2,2'-bipyridine], are rapidly oxidized by H(2)O(2) catalyzed by plant peroxidases to the corresponding Ru(III) species. The commercial isoenzyme C of horseradish peroxidase (HRP-C) and two recently purified peroxidases from sweet potato (SPP) and royal palm tree (RPTP) have been used. The most favorable conditions for the oxidation have been evaluated by varying the pH, buffer, and H(2)O(2) concentrations and the apparent second-order rate constants ( k(app)) have been measured. All the complexes studied are oxidized by HRP-C at similar rates and the rate constants k(app) are identical to those known for the best substrates of HRP-C (10(6)-10(7) M(-1) s(-1)). Both cationic (HRP-C) and anionic (SPP and RPTP) peroxidases show similar catalytic efficiency in the oxidation of the Ru(II) complexes. The mediating capacity of the complexes has been evaluated using the SPP-catalyzed co-oxidation of [Ru(II)(phpy)(bpy)(2)]PF(6) and catechol as a poor peroxidase substrate as an example. The rate of enzyme-catalyzed oxidation of catechol increases more than 10000-fold in the presence of the ruthenium complex. A simple routine for calculating the rate constant k(c) for the oxidation of catechol by the Ru(III) complex generated enzymatically from [Ru(II)(phpy)(bpy)(2)](+) is proposed. It is based on the accepted mechanism of peroxidase catalysis and involves spectrophotometric measurements of the limiting Ru(II) concentration at different concentrations of catechol. The calculated k(c) value of 0.75 M(-1) s(-1) shows that the cyclometalated Ru(II) complexes are efficient mediators in peroxidase catalysis.  相似文献

Mild acid degradation of the lipopolysaccharide of the bacterium Idiomarina zobellii, type strain KMM 231T, with aq 2% HOAc at 100 degrees C, yielded an oligosaccharide, which represents one repeating unit of the O-polysaccharide. A polysaccharide was obtained by mild base degradation of the lipopolysaccharide. The following structure of the O-polysaccharide was elucidated by 1H and 13C NMR spectroscopy of the oligosaccharide and base-degraded lipopolysaccharide, including COSY, TOCSY, ROESY, 1H, 13C HSQC, HSQC-TOCSY and HMBC experiments: [-->3)-alpha-D-Quip4N-(1-->4)-alpha-D-GlcpA-(1-->6)-alpha-D-GlcpNAc-(1-->4)-alpha-L-GulpNA-(1-->3)-beta-D-FucpNAc-(1-->] The O-polysaccharide is distinguished by the presence of two unusual amino sugars, 4-amino-4,6-dideoxy-D-glucose (D-Qui4N) and 2-amino-2-deoxy-L-guluronic acid (L-GulNA), both having the free amino group. The unexpectedly high acid lability of the glycosidic linkage of 2-acetamido-2,6-dideoxy-D-galactose (D-FucNAc) could be associated with the presence of a free amino group adjacent to the site of attachment of FucNAc to Qui4N.  相似文献

A fucoidan fraction consisting of L-fucose, sulfate, and acetate in a molar proportion of 1:1.21:0.08 was isolated from the brown seaweed Fucus distichus collected from the Barents Sea. The 13C NMR spectrum of the fraction was typical of regular polysaccharides containing disaccharide repeating units. According to 1D and 2D 1H and 13C NMR spectra, the fucoidan molecules are built up of alternating 3-linked alpha-L-fucopyranose 2,4-disulfate and 4-linked alpha-L-fucopyranose 2-sulfate residues: -->3)-alpha-L-Fucp-(2,4-di-SO3-)-(1-->4)-alpha-L-Fucp-(2SO3-)-(1-->. The regular structure may be only slightly masked by random acetylation and undersulfation of several disaccharide repeating units.  相似文献
