The synthesis of acidic polysaccharides by eggs of Pomacea sp. at different stages of development was studied. By day 3 after oviposition an acidic galactan became apparent. This compound reaches its maximum concentration by day 6 and then slowly decreases in concentration, no longer being detectable by day 12. Among the sulfated glycosaminoglycans, chondroitin sulfate is the first to appear (day 10), followed by heparan sulfate and other sulfate glycosaminoglycans, which were synthesized in large amounts until hatching (around day 15). Each one of the compounds was purified and characterized by chemical analysis and enzymatic degradation. The synthesis and characterization of the sulfated glycosaminoglycans was confirmed by the use of radioactive sulfate applied to intact eggs at different stages of development. These studies suggest that the main difference between vertebrate and mollusc development regarding the acidic polysaccharides is that hyaluronic acid seems to be absent during early mollusc development. It is proposed that the acidic galactan may be synthesized from the neutral galactan, already present in the eggs in high amounts, and may replace this glycosaminoglycan in the mollusc embryogenesis.  相似文献

Autophagy plays critical roles in the maintenance of endothelial cells in response to cellular stress caused by blood flow. There is growing evidence that both cell adhesion and cell detachment can modulate autophagy, but the mechanisms responsible for this regulation remain unclear. Immunoglobulin and proline-rich receptor-1 (IGPR-1) is a cell adhesion molecule that regulates angiogenesis and endothelial barrier function. In this study, using various biochemical and cellular assays, we demonstrate that IGPR-1 is activated by autophagy-inducing stimuli, such as amino acid starvation, nutrient deprivation, rapamycin, and lipopolysaccharide. Manipulating the IκB kinase β activity coupled with in vivo and in vitro kinase assays demonstrated that IκB kinase β is a key serine/threonine kinase activated by autophagy stimuli and that it catalyzes phosphorylation of IGPR-1 at Ser220. The subsequent activation of IGPR-1, in turn, stimulates phosphorylation of AMP-activated protein kinase, which leads to phosphorylation of the major pro-autophagy proteins ULK1 and Beclin-1 (BECN1), increased LC3-II levels, and accumulation of LC3 punctum. Thus, our data demonstrate that IGPR-1 is activated by autophagy-inducing stimuli and in response regulates autophagy, connecting cell adhesion to autophagy. These findings may have important significance for autophagy-driven pathologies such cardiovascular diseases and cancer and suggest that IGPR-1 may serve as a promising therapeutic target.  相似文献

This study aims to evaluate the correlation between the presences of separated endodontic instrument inside the dental canal and the nickel (Ni) level in the urine samples of subjected patients. Same-gendered and near-aged participants were selected and were instructed to collect their urine in sterile nickel-free plastic containers. The procedures were carried out in the office, and samples were stored in low-temperature cooler for 1 day and then they were transferred to the laboratory for electrothermal atomic absorption spectrometry. The level of Ni was measured and the correlation coefficient was calculated. Data were analyzed using t tests, Pearson’s correlation coefficients, and linear regression analysis, at a level of significance P?<?0.05. The statistical analysis has showed significant difference in Ni level between endodontic and control groups (P?<?0.05). There was no correlation between Ni level in urine and the age or time period of broken instrument inside the canal; however, Ni level of urine and the age of participants in experimental group has demonstrated a positive correlation. The amount of Ni element can be increased in the urine of patients who have experienced broken endodontic instrument inside the dental canal. However, there is no positive correlation between the remaining pieces of instruments inside the canal and the elevation of nickel amount in urine during the tested time period. This issue suggested that the aging of remaining broken instrument inside the canal does not show any remarkable concern regarding the Ni elevation in the urine excreted by an individual.  相似文献

Abstract

Improvement of engineering properties of soils to meet project requirements has long been subject of interest to civil engineers. One of the environment-friendly methods that have recently been used for this purpose is the biological method. These methods that actually benefit from various sciences such as biology, biochemistry, and civil engineering, use biological products or organisms such as bacteria that are commonly found in soils. In this study, the reduction of permeability or hydraulic conductivity of Shiraz landfill base soil using microbial induced calcite precipitation (MICP) has been explored. B. sphaericus was used to treat the soil. Falling head permeability tests are conducted to measure soil samples’ permeability before and after biological treatment. The target variables were the curing time, bacterial density, optimal nutrient content, and soil unit weight. The test results demonstrated that the permeability of the samples treated with Bacillus sphaericus decreases by increasing curing time, the density of calcium chloride solution and bacterial density of samples. This study showed that the MICP can be utilized as a new environment-friendly method for reducing the soil permeability at the base and walls of the landfill to form a barrier between the waste and the groundwater and substrata.  相似文献

The analyses of the products formed from heparitin sulfates by the action of two heparitinases and a heparinase from Flavobacterium heparinum is reported. Heparitin sulfates A and B are degraded by heparitinase I yielding two disaccharides, one of them composed of N-acetylglucosamine and an unsaturated uronic, joined by α(1 → 4) linkage, and the other, with the same composition but with an O-sulfate at the hexosamine moiety. A third disaccharide is also formed from heparitin sulfate B, by the action of the same enzyme, composed of glucosamine N-sulfate and an unsaturated uronic acid joined probably by α(1 → 4) linkage. Besides these three disaccharides, heparitin sulfate B yields, by the action of heparitinase I, an oligosaccharide (with an average molecular weight of 6000) which is completely degraded by the heparitinase II yielding a disaccharide composed of glucosamine 2,6-disulfate and unsaturated uronic acid. All the disaccharides are further degraded by α-glycuronidase from Flavobacterium heparinum yielding the respective monosaccharides. Based on these and other analyses the possible structures of the heparitin sulfates are proposed.  相似文献

In a preliminary study, cryosurgery of the prostate in patients with prostatic cancer has been observed to possess a biphasic effect on the proliferative responsiveness of peripheral blood lymphocytes (PBL) to the plant mitogen, phytohaemagglutinin. Phytohaemagglutinin-stimulated PBL cultured in autologous serum obtained up to 1 week postoperatively showed reduction from their preoperative levels. In contrast, cultures in homologous serum showed increased responsiveness following cryosurgery. Although depression of lymphocyte proliferation in autologous serum was transitory, gradually returning to approximate or exceeding levels of lymphocytic reactivity observed preoperatively, such depression may facilitate reduced host surveillance to potential infectious agents and to metastatic tumour cells, leading to an alteration of tumour-host homeostasis. Further studies will be required to delineate the possible clinical significance of the present observations.  相似文献
