The Gram-negative bacteria, Salmonella, cause a broad spectrum of clinical diseases in humans, ranging from asymptomatic carriage to life-threatening sepsis. We have designed an experimental model to study the contribution of genetic factors to the persistence of Salmonella Enteritidis during the late phase of infection in 129S6/SvEvTac and C57BL/6J mice. C57BL/6J mice cleared the bacteria from their reticuloendothelial system within a period of 42 days, whereas the 129S6 mice still presented a high bacterial load. Using this model, we have identified ten Salmonella Enteritidis susceptibility loci (Ses1, Ses1.1, and Ses3Ses10) associated with bacterial persistence in target organs of 129S6/SvEvTac mice using a two-locus epistasis QTL linkage mapping approach. Significant statistical interactions were detected between Ses1 on chromosome 1 and Ses5 on chromosome 7 and between Ses1 and Ses4 on chromosome X. In this study, we functionally validated the genetic architecture of Salmonella persistence in 129S6 mice using single- (129S6.B6-Ses1.2 that combines Ses1 and Ses1.1 loci, 129S6.B6-Ses4, and 129S6.B6-Ses5) and double-congenic mice (129S6.B6-Ses1.2/Ses4 and 129S6.B6-Ses1.2/Ses5). These experiments demonstrate functional interactions between Ses1.2 and Ses4 or Ses5 that improve Salmonella Enteritidis clearance, validating the critical role played by gene–gene interactions in the contribution to bacterial clearance heritability. Improved bacterial clearance in double-congenic mice could be explained by the impact of Ses4 and Ses5 in combination with Ses1.2 on TH polarization since a TH2 bias (decreased Ifng and increased Il4 mRNA levels and reduced IgG2a immunoglobulins in the serum) was observed in 129S6.B6-Ses1.2/Ses5 mice and a TH17 (high Il17 expression) bias in 129S6.B6-Ses1.2/Ses4.  相似文献

The African freshwater oyster Etheria elliptica, which is of great economic importance throughout the continent, is facing overharvesting in many fisheries in West Africa. Its reproductive traits (sex ratio, size at sexual maturity, oocyte diameter and fecundity) were studied at four stations located along the Pendjari River, northern Benin, in April 2013. Histological techniques were implemented to identify sex and gonad development stages. Oocyte sizes were measured based on the histological images and mean oocyte diameter was 38 µm. Fecundity, estimated by counting the developing eggs, averaged 106 724 eggs and increased significantly with shell size. The average sex ratio was approximately 1:1. Hermaphrodites were rare. Males reached sexual maturity at 57.6 mm dorsoventral height, earlier than females at 64.7 mm. The majority of the oyster specimens were in ripe-spawning stages, indicating that reproductive activities partly took place towards the end of the dry season. Both mature and immature individuals were subjected to harvesting at all the investigated fishing sites. The mean size of exploited oysters was 60.6 mm, lower than the size of females at maturity. Management strategies must enforce the minimum size of 65 mm required for harvesting, particularly in open access sites where juveniles are mostly harvested.  相似文献

The possible involvement of membrane-bound calcium in the mechanism of action of leukotriene B4 was examined using the fluorescent chelate probe, chlortetracycline. Leukotriene B4 was found to cause a rapid release of membrane-bound calcium at physiologically relevant concentrations. This effect of leukotriene B4 is stereospecific and its magnitude is decreased upon the transformation of leukotriene B4 into its omega-hydroxy and omega-carboxy metabolites. The pool of calcium affected by leukotriene B4 appears to be the same as that released by other chemotactic factors such as formyl-methionyl-leucyl-phenylalanine (f-Met-Leu-Phe). Similarly, preincubation with f-Met-Leu-Phe results in a decreased responsiveness of the cells to the addition of leukotriene B4. These results extend further the analogy between the mechanism of action of peptidic and lipid chemotactic factors, and emphasize the central role of the intracellular redistribution of calcium, as inferred and monitored by chlortetracycline fluorescence and steady-state isotopic flux studies, in neutrophil activation.  相似文献

Effect of PCMBS on water transfer across biological membranes   总被引:4,自引:0,他引:4  
P-chloromercuriphenylsulfonate, PCMBS, and 5, 5′ dithiobis-(2-nitrobenzoic acid), DTNB at a concentration of 1 mM are found to inhibit the rate of water transport across human red cell membrane. In addition PCMBS inhibits the rates of transport of small hydrophilic but not hydrophobic nonelectrolytes. Other sulfhydryl reagents such as N-ethylmaleimide and iodoacetamide have no significant effect on the rate of water transfer in these cells. The results suggest that there are at least two populations of membrane bound SH-groups which differ in their topical location which participate in the control of water transfer. One is located closer to the outer surface of the membrane, and thus is readily accessible to PCMBS while the other component is probably located in the membrane interior. These two populations can be dissociated by pH. The effect of PCMBS on water transfer can be greatly influenced by pH and temperature. The main effect of temperature and pH is on the permeability of the membrane to the drug. The same concentration of PCMBS is also found to inhibit to a lesser degree water transfer across other biological membranes.  相似文献

Platelet activating factor has been found to increase the intracellular level of free calcium (as monitored by the fluorescent calcium indicator quin-2) and to stimulate the turnover of the polyphosphoinositides in rabbit neutrophils. Calcium mobilization induced by platelet activating factor, in contrast to previous reports with chemotactic factors, is unaffected by pertussis toxin; on the other hand, stimulated polyphosphoinositol hydrolysis and granule enzyme secretion are potently antagonized under the same conditions. The calcium, as well as the secretory responses to the lipid mediator are largely dependent on the presence of extracellular calcium. Internal contributions to the quin-2 signal are only detectable at relatively high concentrations of platelet activating factor. Calcium mobilization and secretion stimulated by platelet activating factor are inhibited following a short incubation with phorbol 12-myristate 13-acetate. These results are discussed in terms of the possibility that platelet activating factor activates neutrophils via dual pathways, the first involving direct interaction with phorbol ester inhibitable calcium channels and the other the stimulation in a manner dependent on a guanine nucleotide binding protein of the phospholipase C specific for polyphosphoinositides.  相似文献

We have monitored the intracellular pH of rabbit neutrophils by following the distribution of the weak acid, 5,5-Dimethyloxazolidine-2, 4-dione. The synthetic formyl-methionyl chemotactic factors were found to induce complex and specific changes in the intracellular pH of the neutrophils, a rapid drop followed by a slower and more sustained rise. These effects are receptor mediated. The relationship of these events to the physiology of the neutrophils is discussed.  相似文献
