Filamentous fungi show great promise in remediation of environmental contaminants such as industrial dyes. In the current study, Aspergillus niger (Genbank ID: JF437542) decolorized 82 % of the test dye malachite green (MG; 50 mg/l) during cultivation for 24 h. The organism decolorized only 6 % of the MG at higher concentration (250 mg MG/l) during the same time period and growth was inhibited at this higher MG concentration. Exposing A. niger to different types of stress resulted in variable impacts on ability to decolorize MG. CaCl2 had the largest positive impact on decolorization. A. niger cultures treated with CaCl2 (1 M) decolorized 46 % of the MG (250 mg/l) in 1 h compared to 6 % in untreated control cultures. CaCl2 also increased catalase production in A. niger which strongly supported a direct relationship between stress response and decolorizing ability. Spectrophotometric measurement confirmed MG decolorization while Fourier transform infrared spectroscopy suggested that biodegradation of MG occurred. Cultures treated with CaCl2 accumulated fewer toxic MG by-products than untreated cultures. CaCl2-induced stress increased the permeability and conductivity of the fungal cell membrane. An observed increase in medium [H+] also suggested a change in Ca2+/H+ exchange capacity in the fungal cell. Calcium ions had a pronounced effect on membrane properties and this may have had an important impact on signal transduction. We conclude that A. niger decolorizes MG and that CaCl2 enhances this process; the CaCl2 effect appears to be associated with stress response.  相似文献

Acoustic telemetry and scuba revealed immediate and high post-stocking mortality of razorback sucker Xyrauchen texanus in Lake Mohave, U.S.A., a Colorado River impoundment. At the conclusion of this 6 month study, only three of 19 (16%) study fish remained active. Concurrently, 20 X. texanus implanted with acoustic transmitters and held in a hatchery raceway remained healthy throughout the experiment and no transmitters were shed. Post-stocking mortality was probably due to predation by striped bass Morone saxatilis , a piscivorous non-native fish.  相似文献

This first pan-Mediterranean analysis of genetic diversity in wild narrow-leafed lupin revealed strong East–West genetic differentiation of populations, an historic eastward migration, and signatures of genetic adaptation to climatic variables.

Abstract

Most grain crops suffer from a narrow genetic base, which limits their potential for adapting to new challenges such as increased stresses associated with climate change. Plant breeders are returning to the wild ancestors of crops and their close relatives to broaden the genetic base of their crops. Understanding the genetic adaptation of these wild relatives will help plant breeders most effectively use available wild diversity. Here, we took narrow-leafed lupin (Lupinus angustifolius L.) as a model to understand adaptation in a wild crop ancestor. A set of 142 wild accessions of narrow-leafed lupin from across the Mediterranean basin were subjected to genotyping-by-sequencing using Diversity Arrays Technology. Phylogenetic, linkage disequilibrium and demographic analyses were employed to explore the history of narrow-leafed lupin within the Mediterranean region. We found strong genetic differentiation between accessions from the western and eastern Mediterranean, evidence of an historic West to East migration, and that eastern Mediterranean narrow-leafed lupin experienced a severe and recent genetic bottleneck. We showed that these two populations differ for flowering time as a result of local adaptation, with the West flowering late while the East flowers early. A genome-wide association study identified single nucleotide polymorphism markers associated with climatic adaptation. Resolving the origin of wild narrow-leafed lupin and how its migration has induced adaptation to specific regions of the Mediterranean serves as a useful resource not only for developing narrow-leafed lupin cultivars with greater resilience to a changing climate, but also as a model which can be applied to other legumes.

The modelling of ion uptake by plants requires the measurement of kinetic and growth parameters under specific conditions. The objective of this study was to evaluate the effect of nine NH inf4 sup+ :NO inf3 sup− ratios on onions (Allium cepa L.). Twenty-eight to 84 day-old onion plants were treated with NH inf4 sup+ :NOf3/sup− ratios ranging from 0 to 100% of each ionic species in one mM solutions in a growth chamber. Maximum N influx (Imax) was assessed using the N depletion method. Except at an early stage, ionic species did not influence significantly Imax, the Michaelis constant (Km) and the minimum concentration for net uptake (Cmin). Imax for ammonium decreased from 101 to 59 pmole cm-2 s-1 while Imax for nitrate increased from 26 to 54 pmole cm-2 s-1 as the plant matured. On average, Km and Cmin values were 14.29 μM, and 5.06 μM for ammonium, and 11.90 μM and 4.54 μM for nitrate, respectively. In general, the effect of NH4 +:NO3 - ratios on root weight, shoot weight and total weight depended on plant age. At an early stage, maximum plant growth and N uptake were obtained with ammonium as the sole source of N. At later stages, maximum plant growth and N uptake were obtained as the proportion of nitrate increased in the nutrient solution. The was no apparent nutrient deficiency whatever NH4 +:NO3 - ratio was applied, although ammonium reduced the uptake of cations and increased the uptake of phosphorus. The effects of cinnamyl alcohol dehydrogenase (CAD, EC.1.1.1.195) down-regulation on lignin profiles of plants were analysed in four selected transgenic lines of tobacco (Nicotiana tabacum L. cv. Samsun) exhibiting different levels of CAD activity (8–56% of the control). A significant decrease in thioacidolysis yields (i.e. yield of β-O-4 linked monomers) and in the ratio of syringyl to guaiacyl monomers (S/G) was observed for three transgenic lines and the most drastic reduction (up to 50%) was correlated with the lowest level of CAD activity. Higher lignin extractability by mild alkali treatment was confirmed, and, in addition to a tenfold increase in C6-C1 aldehydes, coniferyl aldehyde was detected by high-performance liquid chromatography in the alkali extracts from the xylem of transgenic plants. In-situ polymerisation of cinnamyl aldehydes in stem sections of untransformed tobacco gave a xylem cell wall coloration strikingly similar to the reddish-brown coloration of the xylem of antisense CAD-down-regulated plants. Overall, these data provide new arguments for the involvement of polymerised cinnamyl aldehydes in the formation of the red-coloured xylem of CAD-down-regulated plants. Glutathione S-transferases (GSTs) are a family of multi-functional enzymes that play critical roles in the detoxification of xenobiotics and the protection of tissues against oxidative damage. GSTs are important enzymes in plant responses to a number of environmental stresses including herbicides and pathogen attack. Ocs elements are a group of related, 20 bp promoter elements which have been exploited by some plant pathogens to express genes in plants. Ocs elements have also been found to regulate the expression of a plant GST promoter. An Arabidopsis GST gene, called GST6 has been isolated. GST6 expression is under tissue-specific control and is induced following treatment with auxin, salicylic acid and H2O2. The GST6 promoter contains a binding site for two Arabidopsis o cs element b inding f actors (OBF), that has some sequence homology to ocs element sequences. Interestingly, OBP1 (O BF b inding p rotein), a DNA-binding protein that was isolated by screening an Arabidopsis cDNA library with a labeled OBF protein as a probe, binds next to the OBF-binding site on the GST6 promoter. OBP1 was able to significantly stimulate the binding of OBF proteins to the GST6 promoter, raising the possibility that interactions between the OBP1 and OBF proteins may be important for GST6 expression.  相似文献

Objectives

We examined whether mobility, migrant status, and risk environments are associated with sexually transmitted infections (STIs) and HIV risk behaviors (e.g. sex trading, multiple partners, and unprotected sex).

Methods

We used Respondent Driven Sampling (RDS) to recruit external male migrant market vendors from Kyrgyzstan, Uzbekistan, and Tajikistan as well internal migrant and non-migrant market vendors from Kazakhstan. We conducted multivariate logistic regressions to examine the effects of mobility combined with the interaction between mobility and migration status on STIs and sexual risk behaviors, when controlling for risk environment characteristics.

Results

Mobility was associated with increased risk for biologically-confirmed STIs, sex trading, and unprotected sex among non-migrants, but not among internal or external migrants. Condom use rates were low among all three groups, particularly external migrants. Risk environment factors of low-income status, debt, homelessness, and limited access to medical care were associated with unprotected sex among external migrants.

Conclusion

Study findings underscore the role mobility and risk environments play in shaping HIV/STI risks. They highlight the need to consider mobility in the context of migration status and other risk environment factors in developing effective prevention strategies for this population.  相似文献

Temperature-sensitive (ts) mutants of the T4 phage rII gene were islated and used in temperature shift experiments that revelaed two different expressions for the normal rII (rII+) gene function in vivo: (i) an early expression (0 to 12 min postinfection at 30 C) that prevents restriction of T4 growth in Escherichia coli hosts lysogenic for gamma phage, and (ii) a later expression (12 to 18 min postinfection at 30 C) that results in restriction of T4 growth when the phage DNA ligase (gene 30) is missing. The earlier expression appeared to coincide with the period of synthesis of the protein product of the T4 rIIA cistron, whereas the later expression occurred after rIIA protein synthesis had stopped. The synthesis of the protein product of the rIIB cistron continues for several minutes after rIIA protein synthesis ceases (O'Farrell and Gold, 1973). The two rII+ gene expressions might require different molar ratios of the rIIA and rIIB proteins. It is possible that the separate expressions of rII+ gene function are manifestations of different associations between the two rII proteins and other T4-induced proteins that are synthesized or activated at different times after phage infection.  相似文献

Amber (am) mutants of the two closely linked sites, B22 and C125, in bacteriophage T4 gene 43 [deoxyribonucleic acid (DNA) polymerase] synthesize in the nonpermissive (su(-)) Escherichia coli host gene 43 products which are devoid of DNA polymerase activity, but which retain a 3'-exonuclease activity. Diethylaminoethyl-cellulose chromatographic analysis of DNA polymerase and deoxyribonuclease activities from extracts of su(-) cells infected with single- and double-am mutants of T4 gene 43 showed that the exonuclease activity which is observed with amB22 is not seen with double mutants carrying, in addition to amB22, am mutations which map to the clockwise side of the B22 site on the circular genetic map of T4. Similarly, am mutations which map to the clockwise side of the C125 site abolish the exonuclease activity which is observed with an am mutant (amE4335) of this site. It was concluded that in these double mutants termination signals to the clockwise side of amB22 and amE4335 are encountered before the amB22 and amE4335 signals during translation of the messenger ribonucleic acid from T4 gene 43. Thus, it seems that the T4 DNA polymerase is synthesized in vivo in a direction which corresponds to a counterclockwise reading of gene 43.  相似文献
