A high level expression of the Ea(d) transgene encoding the I-E alpha-chain is highly effective in the suppression of lupus autoantibody production in mice. To explore the possible modulation of the Ag-presenting capacity of B cells as a result of the transgene expression, we assessed the ability of the transgenic B cells to activate Ag-specific T cells in vitro. By using four different model Ag-MHC class II combinations, this analysis revealed that a high transgene expression in B cells markedly inhibits the activation of T cells in an epitope-dependent manner, without modulation of the I-E expression. The transgene-mediated suppression of T cell responses is likely to be related to the relative affinity of peptides derived from transgenic I-E alpha-chains (Ealpha peptides) vs antigenic peptides to individual class II molecules. Our results support a model of autoimmunity prevention based on competition for Ag presentation, in which the generation of large amounts of Ealpha peptides with high affinity to I-A molecules decreases the use of I-A for presentation of pathogenic self-peptides by B cells, thereby preventing excessive activation of autoreactive T and B cells.  相似文献

Genotype 3 Hepatitis E virus (HEV-3) is an emerging threat for aging population. More than one third of older infected patients develops clinical symptoms with severe liver damage, while others remain asymptomatic. The origin of this discrepancy is still elusive although HEV-3 pathogenesis appears to be immune-mediated. Therefore, we investigated the role of CD8 T cells in the outcome of the infection in immunocompetent elderly subjects. We enrolled twenty two HEV-3-infected patients displaying similar viral determinants and fifteen healthy donors. Among the infected group, sixteen patients experienced clinical symptoms related to liver disease while six remained asymptomatic. Here we report that symptomatic infection is characterized by an expansion of highly activated effector memory CD8 T (EM) cells, regardless of antigen specificity. This robust activation is associated with key features of early T cell exhaustion including a loss in polyfunctional type-1 cytokine production and partial commitment to type-2 cells. In addition, we show that bystander activation of EM cells seems to be dependent on the inflammatory cytokines IL-15 and IL-18, and is supported by an upregulation of the activating receptor NKG2D and an exuberant expression of T-Bet and T-Bet-regulated genes including granzyme B and CXCR3. We also show that the inflammatory chemokines CXCL9-10 are increased in symptomatic patients thereby fostering the recruitment of highly cytotoxic EM cells into the liver in a CXCR3-dependent manner. Finally, we find that the EM-biased immune response returns to homeostasis following viral clearance and disease resolution, further linking the EM cells response to viral burden. Conversely, asymptomatic patients are endowed with low-to-moderate EM cell response. In summary, our findings define immune correlates that contribute to HEV-3 pathogenesis and emphasize the central role of EM cells in governing the outcome of the infection.  相似文献

Aim

HIV infection is associated with distortion of T-cell homeostasis and the IL-7/IL7R axis. Progressive infection results in loss of CD127+132− and gains in CD127−132+ CD4+ and CD8+ T-cells. We investigated the correlates of loss of CD127 from the T-cell surface to understand mechanisms underlying this homeostatic dysregulation.

Methods

Peripheral and cord blood mononuclear cells (PBMCs; CBMC) from healthy volunteers and PBMC from patients with HIV infection were studied. CD127+132−, CD127+132+ and CD127−132+ T-cells were phenotyped by activation, differentiation, proliferation and survival markers. Cellular HIV-DNA content and signal-joint T-cell receptor excision circles (sjTRECs) were measured.

Results

CD127+132− T-cells were enriched for naïve cells while CD127−132+ T-cells were enriched for activated/terminally differentiated T-cells in CD4+ and CD8+ subsets in health and HIV infection. HIV was associated with increased proportions of activated/terminally differentiated CD127−132+ T-cells. In contrast to CD127+132− T-cells, CD127−132+ T-cells were Ki-67+Bcl-2low and contained increased levels of HIV-DNA. Naïve CD127+132− T-cells contained a higher proportion of sjTRECs.

Conclusion

The loss of CD127 from the T-cell surface in HIV infection is driven by activation of CD127+132− recent thymic emigrants into CD127−132+ activated/terminally differentiated cells. This process likely results in an irreversible loss of CD127 and permanent distortion of T-cell homeostasis.  相似文献

Campylobacter jejuni (C. jejuni) is a leading cause of human enteritis worldwide and the most frequently reported zoonotic agent in the European Union. Despite the fact that C. jejuni is a microaerobic bacteria, known as a fragile one, it is able to survive through adverse conditions such as oxidative stress. The purpose of this study was first to test the oxidative stress resistance in 22 C. jejuni strains of various origins, and to compare adhesive and invasive abilities of four selected strains in the intestinal cell line Ht-29. Secondly, the effect of an oxidative stress on C. jejuni adhesion to Ht-29 cells was investigated. Results show that all the tested strains were able to survive after a 24-h incubation period in broth containing 10 μM of paraquat. From 12.5 μM of paraquat, bacterial strains exhibit different behaviour, and only three strains are able to survive at 25 μM of paraquat. In addition, this study revealed that the number of bound bacteria to epithelial cells increases with augmentation of paraquat concentration, suggesting a link between oxidative stress survival of C. jejuni and virulence on Ht-29 cells.  相似文献

Nothobranchius cooperi, Nagy, Watters and Bellstedt, new species, is described from seasonal streams and ephemeral pools associated with the upper Mansa River system in the middle Luapula drainage and systems draining into the low-lying area marginal to the southwestern part of Lake Bangweulu, in the Luapula province of northern Zambia. It belongs to the N. brieni species group. Males of Nothobranchius cooperi are distinguished from congeners by the following unique combination of characters: body scales with broad orange posterior margin, forming a highly irregular cross-barred pattern; anal fin fairly uniform orange-red with irregular to regular, light blue-green zone close to the base; caudal peduncle length 1.2–1.3 times its depth; prepelvic length 48.8–51.9% SL; and head depth 75–77% of head length. Genetic divergence of the mitochondrial COI and ND2 genes and nuclear S7 gene support the distinction of the new species from its closest known relative, N. rosenstocki and confirms its position in the N. brieni species group.  相似文献

Summary A study has been made on the influx and outflux of nitrogen compounds by the excised roots of barley, wheat and peas. A two way movement of nitrogen compounds was found to occur between root and external medium. Factors such as initial N content in the roots, species of plant and external concentration of N highly affect the extent to which this two-way movement proceeds. Further investigations are needed for more understanding of the nitrogen balance between plant roots and external medium.  相似文献
