A recombinase-based in vivo expression technology (RIVET) approach with Xanthomonas campestris pv. vesicatoria (Xcv) revealed that lipA, annotated as putative secreted lipase, is expressed during the interaction between this pathogen and tomato. Here, the tnpR and uidA reporter genes were used to show that lipA is strongly induced in XVM2 minimal medium and during the early stages of tomato infection by Xcv. A mutant strain impaired in lipA was generated by insertional mutagenesis. This mutant grew in a similar manner to the wild-type in rich medium, but its growth was significantly compromised in a medium containing olive oil as a single carbon source. The lipolytic activity of the extracellular fraction of the lipA mutant was reduced significantly relative to that of the wild-type strain, thus confirming that lipA indeed encodes a functional secreted enzyme with lipolytic activity. A plasmid carrying a wild-type copy of lipA complemented the lipA mutant for extracellular lipolytic activity. Dip inoculation experiments with tomato lines Hawaii 7998 (H7998) and Micro Tom showed that the lipA mutant grew to a lesser extent than the wild-type in tomato leaves. Following leaf syringe infiltrations, the mutant strain induced disease symptoms that were less severe than those induced by the wild-type strain, supporting a significant role of lipA in the pathogenicity of Xcv. 相似文献

Inhaled oxygen-15-labeled carbon dioxide (CO2*) is hydrated in the alveolar capillary blood to produce oxygen-15-labeled water (H2O*). This allows noninvasive delivery of a traceable indicator into the pulmonary circulation. Removal of oxygen-15 marker from the lung is a function of pulmonary perfusion. Two techniques were evaluated for computing cardiac output (CO) following single bolus inhalation of CO2*: 1) continuous monitoring of arterial blood activity through an external detector and 2) noninvasive positron imaging of oxygen-15-label washout from the chest and simultaneous emergence of activity in arterial blood. In seven mongrel dogs studied using technique 1, 46 determinations of CO were made from 1.2 to 8.0 l/min and compared with simultaneous indocyanine green dye-dilution determination. Correlation coefficient was 0.90 with slope of linear regression of 1.05. In 12 mongrel dogs studied using technique 2, 23 determinations of CO were made from 0.9 to 9.2 l/min and compared with simultaneous indocyanine green dye determination. Correlation coefficient was 0.985 (P less than 0.001) with slope of linear regression of 0.898. This noninvasive technique (2) for determination of CO is independent of assumptions regarding regional ventilation or perfusion of the lung and appears valid in animal studies. 相似文献

Natural killer (NK) cells have long been considered as potential agents for adoptive cell therapy for solid cancer patients. Until today most studies utilized autologous NK cells and yielded disappointing results. Here we analyze various modular strategies to employ allogeneic NK cells for adoptive cell transfer, including donor-recipient HLA-C mismatching, selective activation and induction of melanoma-recognizing lysis receptors, and co-administration of antibodies to elicit antibody-dependent cell cytotoxicity (ADCC). We show that NK cell activation and induction of the relevant lysis receptors, as well as co-administration of antibodies yield substantial anti-cancer effects, which are functionally superior to HLA-C mismatching. Combination of the various strategies yielded improved effects. In addition, we developed various clinically-compatible ex vivo expansion protocols that were optimized according to fold expansion, purity and expression of lysis receptors. The main advantages of employing allogeneic NK cells are accessibility, the ability to use a single donor for many patients, combination with various strategies associated with the mechanism of action, e.g. antibodies and specific activation, as well as donor selection according to HLA or CD16 genotypes. Summary A brief literature review on the incidence of fungi from various sites in the body is presented. Throat swabs were taken from 468 patients attending the Johannesburg General Hospital between March 1968 and February 1969. The fungi obtained comprised yeasts only and no filamentous species. Interactions between microorganisms can have a crucial effect on their population dynamics. Typically, interactions are mediated through the environment by molecules and proteins that are products of cell metabolism and physiology; they therefore reflect the internal dynamics of the single cell. In this work we aim to integrate single-cell properties of gene expression that affect indirect interactions between microorganisms under challenging conditions, into a quantitative model of population dynamics. Specifically we address the problem of a microbial population secreting a protein that can actively extract a growth-limiting resource, such as a simple sugar or iron, from the environment. The genes coding for the protein can undergo random epigenetic transitions between active and silenced states, and can be repressed by the product of their reaction. We model cooperative and competitive interactions between protein producing and non-producing phenotypes by nonlinear dynamical systems and analyze them both in terms of asymptotic states and of transient dynamics. Our model shows that phenotypic transitions allow a stable coexistence of the two phenotypes, and enables us to make predictions regarding the conditions required for such coexistence and the typical timescales of transient dynamics. It also shows how repression by the reaction product induces a feedback at the population-environment level that can result in limit cycle dynamics. The current study explored the influence of musical expertise, and specifically training in improvisation on creativity, using the framework of the twofold model, according to which creativity involves a process of idea generation and idea evaluation. Based on the hypothesis that a strict evaluation phase may have an inhibiting effect over the generation phase, we predicted that training in improvisation may have a “releasing effect” on the evaluation system, leading to greater creativity. To examine this hypothesis, we compared performance among three groups - musicians trained in improvisation, musicians not trained in improvisation, and non-musicians - on divergent thinking tasks and on their evaluation of creativity. The improvisation group scored higher on fluency and originality compared to the other two groups. Among the musicians, evaluation of creativity mediated how experience in improvisation was related to originality and fluency scores. It is concluded that deliberate practice of improvisation may have a “releasing effect” on creativity. 相似文献
