AFLP analysis was performed between a pair of thermo-sensitive genic male sterile (TGMS) rice allelic mutant lines (5460S and 5460F). The reaction conditions for rice AFLP assay were optimized. The relative efficiencies for polymorphism detection of RFLP, RAPD and AFLP were compared. The results indicated that the efficiency for polymorphism detection in rice was in the order of AFLP > RAPD > RFLP, and also indicated that AFLP was a powerful DNA molecular marker technique for polymorphism detection, especially in the case of extremely low polymorphism, such as isogenic lines and allehc mutant hnes. Some of the AFLP products between the TGMS rice allehc mutant lines were cloned. Three of them were used as mixed probes to screen BAC library of rice line 5460S. 12 positive clones were screened out. In addition, the advantages and disadvantages of these three molecular marker systems were discussed.  相似文献

沿海湿地是鸟类重要的栖息地,人类活动导致的湿地生境变化对鸟类群落造成了一定的影响。在盐城自然保护区新洋港河口内,人类活动大体上导致了由陆向海方向,形成芦苇沼泽、农田、鱼塘和滩涂4种不同干扰程度的生境。分别在不同生境中设置3条长3 km的样线,对越冬鸟类的种类、数量进行了调查,运用多响应置换过程测试、指示种分析、双向聚类分析和除趋势对应分析等统计方法,研究了不同生境中冬季鸟类的指示物种、群落结构及梯度变化,结果表明:1)4种生境共记录鸟类67种,其中鱼塘生境的鸟类种类最多,为37种;滩涂生境的D'多样性指数在各生境不同时段中为最高,芦苇沼泽生境的H'多样性指数在各生境不同时段中为最高。与其余生境比较,芦苇沼泽、滩涂的均匀度也更高。生境间的差异性与鸟类群落的组成有关。2)MRPP分析表明,芦苇沼泽、农田生境的鸟类组成差异不显著(P0.05);其余组合各生境间的鸟类组成具有显著差异。灰鹤等9种鸟类可分别作为4种不同生境的指示种。3)双向聚类分析法把12种观测条件分为4大类,与生境划分非常吻合。在剩余信息40%水平上,鸟类可聚为7类。利用12种观测条件作为变量进行DCA排序的结果表明,鸟类类群在轴1上可以分为3个集群,分别为适应开阔水域环境的鸟类、依赖水生植被环境的鸟类和依赖陆地植被环境的鸟类。根据结果分析,生境异质性、干扰程度及潮水涨落等与鸟类多样性组成有密切关系;不同生境具有明显的指示种;鸟类群落结构为适应生境变化而出现一定的梯度变化。应加强芦苇沼泽的保护,在开垦农田、鱼塘和在光滩引入互花米草时应考虑到人类活动对鸟类多样性的影响。  相似文献

目的:合成单链莫内甜蛋白基因,构建其植物表达载体。方法与结果:根据已报道的单链莫内甜蛋白的氨基酸序列及甜味机理,重新设计合成了全长294bp的莫内甜蛋白基因,其中单链莫内甜蛋白氨基酸序列中的Asp69(原AspA16)突变为Asn。利用DNA重组技术,将莫内甜蛋白基因克隆到植物表达载体pBl221中,构建了莫内甜蛋白基因的植物表达载体pBI221-monellin。结论:构建了莫内甜蛋白基因的植物表达载体,为转化园艺植物以改善其口感奠定了基础。  相似文献

The subterranean termite Reticulitermes speratus usually nests in rotten wood trunks, which may also be occupied by the Japanese garden ant Lasius japonicus. Few battles were observed between them under ordinary circumstances because they inhabit separate nesting sites. However, once the termite nesting sites were artificially broken, the ant workers invaded and hunted the termites, although the termite soldiers fought against the ants. This study aims to confirm intra‐ and inter‐specific chemical interactions between the termite and ant. Solid phase microextraction–gas chromatograph (SPME‐GC) analyses revealed that R. speratus soldiers secreted caste‐specific sesquiterpene hydrocarbon when they were irritated. Both the hexane extract of the soldiers and its hydrocarbon fraction, as well as the crushed soldier bodies, attracted the soldiers but dispersed the workers when presented on the trails. We also confirmed that the soldier chemicals enhanced aggressiveness of L. japonicus, which rushed around the odor sources and hunted any termites that were present. These findings suggest that: (i) the soldier–specific secretion might serve as an alarm pheromone in termite chemical communication, in which components recruit soldiers and also warn the other colony members away; and (ii) termite communication is eavesdropped on by L. japonicus workers to locate and hunt the termites.  相似文献

On the basis of field observations and museum diagnosis of two specimens collected from Ngoc Linh in the Western Highlands (sometimes termed Central Highlands) of Vietnam, we here describe a new species of Actinodura. This species is most closely related to A. ramsayi of Burma, China, Laos, Thailand and Vietnam. Among other differences, it has a distinctive black crown - a feature otherwise unknown in the genus Actinodura , of which it is the southernmost known taxon. It occurs in montane evergreen forest from at least about 1100–2400 m asl and is likely to occur widely in this habitat and at this altitude elsewhere in the Western Highlands and in adjacent Laos. We present notes on the taxonomic affinities, ecology, behaviour and conservation of this new species.
A partir d'observations effectuées sur le terrain et de l'analyse en laboratoire de deux spécimens collectés au Mont Ngoc Linh dans le massif de Kontum, nous décrivons ici une nouvelle espèce du genre Actinodura. Cette espèce est la plus proche de A. ramsayi de Chine, Birmanie, Laos, Thaïlande et Viêt-nam. Elle possède une calotte noire distincte, un caractère jusqu'à présent inconnu du genre Actinodura dont elle occupè l'atire la plus mAeridionale connue. Cette espèce vit dans la forêt sempervirente de montagne entre 1100 m et 2400 m d'altitude et se trouve probablement dans un habitat et à une altitude similaires dans les montagnes environnantes du massif de Kontum et du Laos adjacent. Nous présentons ici des informations sur la taxonomie, l'écologie, le comportement et la conservation de cette nouvelle espèce.  相似文献

A typical marl lake of the Upper Great Lakes region has very few quantitatively important aquatic macrophytes. The macrophytes, however, dominate the total primary production of the lake. Submersed vegetation is extremely sparse on the shallow (less than I m) marl bench that characterizes the littoral of these lakes, and is completely dominated by one. little-known species (Scirpus subterminalis Torr.) between 1 and 7 m. A detailed investigation of the spatial and seasonal distribution of macrophytic species and biomass showed that S. subterminalis strongly dominated the lake (79% of total biomass). S. suhterminalis represented an almost pure stand (to 200 g m?2 mean annual ash-free dry weight) at all times of the year at intermediate depths of macrophytic growth (1–6 m). Two species of Chara (of eight varieties and forms) were present in significant quantities (12% of total biomass; to 100 g m?2) but were severely limited to shallow depths (0-S-l m) and protected areas. Several annual submersed angiosperms were present (9% of total biomass), but only two species were quantitatively important. Potamogeton illinoensis Morong. and P. praelongus Wulfen formed brief summer peaks (less than 100 g m?2) at 3 and 4–6 m, respectively. A striking feature of the seasonal biomass distribution of Scirpus subterminalis was the higher, viable biomass (to 150g m?2) throughout the winter under ice cover. Cyclic fluctuations of the S. subterminalis populations were discerned at different depths, each with different periodicities. The population at 2 m exhibited a fall peak; that at 4 m had a summer maximum. The lowest overall biomass of S. subterminalis occurred in the 2 m population in June. Chara populations at 0–2 m also exhibited a relatively constant biomass throughout the year. The appearance of Nitella at 7 m in July-October and of Chara at 5 m in September-October was interpreted as an interaction between light, thermal, and carbon stratification. Estimates of macrophytic productivity of perennial (‘evergreen’) species populations whose biomass remains relatively constant throughout the year were made employing several different methods of calculation and turnover factors. All methods resulted in productivity estimates in good agreement with the conservative value of 178 g m?2 year?1 for the entire lake. In comparison to the other components (phyto-planktonic, epiphytic and epipelic algae) of the primary production of Lawrence Lake, the aquatic macrophytes constituted a major portion (anuual mean 82·77 g C m?2 year?1 or 48·3 %) of the total production of the lake. The low diversity but relatively high quantitative importance of macrophytes in marl lakes is attributed to an adverse dissolved inorganic and organic chemical milieu which inhibits phytoplanktonic production and allows only certain adapted macrophytes to develop strongly. The phenomenon of perennial biomass levels throughout the year is believed to be much more common than previously suspected and has iikely resulted from adaptations of submersed macrophytes to ameliorated conditions of water and temperatures relative to the terrestrial situation in winter.  相似文献

该文报道了国产杜鹃花科(Ericaceae)一新记录种,即埃氏越橘(Vaccinium eberhardtii Dop)。该种隶属于越橘属(Vaccinium)南烛组(Vaccinium sect. Bracteata),与同属于该组的南烛(V. bracteatum)和海岛越橘(V. wrightii)近缘,但与前者的区别在于花序梗、花梗、萼筒无毛,花冠球状坛形,药室背部明显具2个距,与后者的区别在于花梗较短,短于苞片,药室背后的距直立,长超过药管的一半。据文献记录,该种原来仅分布于越南中部和泰国,此次发现该种的分布范围可以北延至中国的广西东兴市的沿海地区,不仅丰富了越橘属分类、分布的数据,也为今后该属的分类学修订提供了新资料。凭证标本保存在中国科学院华南植物园标本馆(IBSC)和中国科学院广西植物研究所标本馆(IBK)中。  相似文献
