We have determined the complete nucleotide sequence of the small- subunit ribosomal RNA genes for the ciliate protozoans Stylonychia pustulata and Oxytricha nova. The sequences are homologous and sufficiently similar that these organisms must be closely related. In a phylogeny inferred from comparisons of several eukaryotic small-subunit ribosomal RNAs, the divergence of the ciliates from the eukaryotic line of descent is seen to coincide with the radiation of the plants, the animals, and the fungi. This radiation is preceded by the divergence of the slime mold, Dictyostelium discoideum.   相似文献

Temperature-controlled open-top chambers for global change research   总被引:6,自引:0,他引:6  
To enable experiments on the interactive effects of elevated atmospheric CO2 and increased air temperature on physiological processes in trees to be carried out, we altered the standard design of open-top chambers by replacing blowers with evaporative coolers and in-line heaters, with a feedback control system to maintain ambient or elevated air temperatures within the chambers. Ambient and elevated (+ 4 °C) temperature regimes were attained consistently and reliably throughout the growing season, with high reproducibility between chambers. From May through December the average of nearly 300,000 temperature measurements was 18.5 °C in ambient air, 18.9 ± 0.6 °C in six ambient chambers, and 22.4 ± 0.9 °C in six elevated temperature chambers. The difference in soil temperature between ambient and elevated chambers was 1.2 °C. Absolute humidity (vapour pressure) in the chambers was higher than that of ambient air, but it was generally similar between temperature treatments. Vapour pressure deficit therefore was higher in elevated temperature chambers than in ambient chambers, and this difference is considered an inseparable part of the temperature treatment. The addition of a temperature control system to open-top chambers removes what has been an important flaw in this important tool for global change research.  相似文献

Maximum ATPase activities in the cell wall fraction of English ryegrass (Lolium perenne L.) roots were stimulated by foru discrete millimole ratios of (Na++ K+); 40:0, 35:5, 5:35, and 0:40. The optimal pH for stimlation was found to be 6.5. Contrary to data in the literature, Mg2+ inhibited all stimulatory ratios of (Na++ K+) when plants were cultured on an adequate nutrient solution. When grown on a dilute solution, Mg2+ enhanced (Na++ K+)-stimulated ATPase activity in this membrane preparation. The single optimal combined concentration of (Na++ K+) for all stimulatory ratios was 40 MM. The ratios of (Na++ K+) which stimulated ATPase activity in the cell wall fraction varied with position along the root axis such that all rarely existed simultaneously nor did any exist in the terminal millimetre of the root. Both cell wall and microsomal fractions showed stimulation by (Na++ K+) at all the above ratios indicating the possible presence of plasma membrane fragments in both fractions. Only the 35:5 ratio was stimulations were found in the supernatant. Implications of ion-stimulated ATPase involvement in ion transport were drawn from the appearance of ATPase activity at a 40:0 ratio of (Na++ K+) and the disappearance of stimulations at 35:5, 5:35, and 0:40 ratios when plants were moved from a strong (35 mM total concentration) to a dilute (0.75 mM) nutrient solution.  相似文献

The presence of boron in the germination medium markedly increasedthe RNA level and a-amylase activity in the embryo and endospermtissue of dormant 2-month-old seed. In non-dormant 14-month-oldseed the presence of boron slightly increased the level of RNA,but had virtually no effect on the -amylase activity. Gibberellicacid (GA2) in the germination medium also increased the -amylaseactivity in the 2-month-old seed; the greatest activity wasobtained when both boron and GA2 were together in the germinationmedium. Changes in starch, sucrose, and reducing sugar levelswere obtained in the seed, in the presence of boron. Indicationsof changes in RNA level, a-amylase activity, and sugar translocationin non-germinating seed occur between the dormant and non-dormantstate. Increasing growth irradiance and CO2 generally decreases foliarnutrient concentration on a dry weight basis and increases foliarstarch concentration. However, the extent to which starch concentrations‘dilute’ foliar nutrient concentrations when thelatter are expressed on a dry weight basis is not known. Todetermine the importance of differential starch accumulationin calculating nutrient concentrations on a dry weight basis,leaf nutrient and starch concentrations were measured in Chrysanthemum? morifolium ‘Fiesta’ (Ramat.) cuttings grown atthree irradiance levels and two CO2 levels for eight weeks inboth winter and spring. On a dry weight basis, foliar concentrationsof most nutrients were lower in both seasons as a result ofthe elevated CO2 and irradiance levels, and total dry weightswere higher. Per cent starch was greater at the high CO2 levelin both seasons but was only greater at higher irradiances inthe winter experiment. When starch was subtracted from the leafdry weights, the differences between CO2 and irradiance treatmentsdisappeared with respect to N, P, K, Ca, Mg, S, and B but notfor Fe, Mn, Zn, and Cu. The lymphoblastomas occurring in childhood are divided for purposes of discussion into lymphocytoma cutis, mycosis fungoides, lymphosarcoma, Hodgkin's disease, and leukemia. The cutaneous lesions may be either specific (as a result of the infiltration of the skin with specific cells of the conditions) or toxic (non-specific). With the possible exception of mycosis fungoides, the cutaneous manifestations are not diagnostic. Three cell types are present in tubules of the digestive glandof the marine prosobranch Maoricrypta monoxyla (Lesson). Histochemistry,and feeding and starvation experiments established that themain type, the digestive cell, is involved in endocytotic uptakeof food material from the lumen. Digestion of this materialoccurs within vacuoles, and indigestible material (indicatedby the dye trypan blue) accumulates in basal residual bodiescontaining lipofuscin pigment. Another cell type, the cryptcell, appears to secrete a glycoprotein, probably enzymaticin function. The third cell type contains large vacuoles butit has not been Established whether the contents are secretoryor excretory. The tubules undergo a cycle of digestive activity not relatedto the tidal cycle as in some marine molluscs, but probablyan indirect result of the feeding regime. The cycle begins withimmature tubules in which some endocytosis occurs. These developinto absorbing tubules involved mainly in food uptake. In maturetubules intracellular digestion occurs. At a later stage thetubules fragment to produce spherules which may conserve usefulmaterial, and finally they, disintegrate completely. Eighty per cent of the dividing cells in the epithelium occurin crypts, which are therefore considered to be the main sitesof epithelial renewal.
