The Cambrian bradoriid ? Parahoulongdongella sp. is shown to have a microreticulate (2nd order) surface sculpture. A possible organic sheet‐like origin is proposed for this reticulation. Abstract: Although the relationship between ontogeny and phylogeny has been of long‐standing interest to palaeontologists, the fossil record has provided little insight into the development of long extinct organisms. This has changed with the discovery of numerous assemblages of fossilized invertebrate embryos and larvae, but realising their evolutionary significance is hampered by a paucity of data on the relationship between ontogeny and taphonomy. We describe the results of an experimental taphonomy study of the development of the anostracan brine shrimp Artemia salina, which show that in conditions of aqueous aerobic and anaerobic autolysis and microbial decay, the developmental stages exhibit differential preservation potential. The most decay resistant developmental stage is the diapause cyst, encapulsating the gastrula, in which the gross morphology of the embryo can be maintained for 18 months or more in simple anaerobic conditions. Otherwise, the embryo shrinks within the cyst and cellular and tissue detail of breaks down as lipid droplets coalesce. Postembryonic excysted larvae decay more rapidly. The rate of decay is similar among all larval stages with the exception of the L4 larva, which resists cuticle failure for longer than later developmental stages. The larvae decay leading to liquefaction of the muscles and viscera, leaving an intact but empty and progressively shrunken and distorted cuticle that eventually loses structural integrity and collapses. Our experimental results provide an explanatory model for the phenomenal abundance of putative diapause stage embryos, in the absence of postembryonic stages, as seen in the Ediacaran Doushantuo Formation of South China and the incompleteness of fossilized developmental sequences of embryos and larvae more generally. It also cautions against the association of developmental stages in fossil deposits without additional evidence. Finally, the pattern of decay seen in larvae provides an explanation for the preservation style of Orsten‐type Lagerstätten where preservation of cuticular detail can be astonishingly fine, but extends internally to muscles and viscera only rarely.  相似文献

Population genetics of the amphibian pathogen Batrachochytrium dendrobatidis ( Bd ) show that isolates are highly related and globally homogenous, data that are consistent with the recent epidemic spread of a previously endemic organism. Highly related isolates are predicted to be functionally similar due to low levels of heritable genetic diversity. To test this hypothesis, we took a global panel of Bd isolates and measured (i) the genetic relatedness among isolates, (ii) proteomic profiles of isolates, (iii) the susceptibility of isolates to the antifungal drug caspofungin, (iv) the variation among isolates in growth and phenotypic characteristics, and (v) the virulence of isolates against the European common toad Bufo bufo . Our results show (i) genotypic differentiation among isolates, (ii) proteomic differentiation among isolates, (iii) no significant differences in susceptibility to caspofungin, (iv) differentiation in growth and phenotypic/morphological characters, and (v) differential virulence in B. bufo . Specifically, our data show that Bd isolates can be profiled by their genotypic and proteomic characteristics, as well as by the size of their sporangia. Bd genotypic and phenotypic distance matrices are significantly correlated, showing that less-related isolates are more biologically unique. Mass spectrometry has identified a set of candidate genes associated with inter-isolate variation. Our data show that, despite its rapid global emergence, Bd isolates are not identical and differ in several important characters that are linked to virulence. We argue that future studies need to clarify the mechanism(s) and rate at which Bd is evolving, and the impact that such variation has on the host–pathogen dynamic.  相似文献

The Droseraceae exhibit a range of"germination patterns which are of possible taxonomic usefulness at the sectional and higher levels. Three patterns were observed in the family: phanerocotylar ( Drosera subgen. Drosera and Regia ); cryptocotylar (subgen. Ergaleium and Rorella ); and a new category, hemi-cryptocotylar, where both cotyledon apices remain in the seed coat, but the remainder emerges forming a photosynthetic cotyledon loop ( Dionaea, Drosophyllum and Drosera adelae ). Subgenus Drosera , especially sect. Drosera , is heterogenous for germination, and other features, with Drosera indica , sect. Lasiocephala and D. neocaledonica anomalous on current placements, and in need of further study. Conversely, the merger of sects Psycophila and Stelogyne and subgenl Ptycnostigma with subgen. Drosera sect. Drosera is supported by their germination features. Further studies are needed to determine the significance of environmental effects on the germination characteristics, although because the results of this study largely agree with recent morphological and molecular studies, this suggests that germination characteristics are useful at the sectional level, potentially providing additional characters for phylogenetic investigations within the Droseraceae.  相似文献

A pilot study for grafting of patients with vitiligo using cultured epithelial autografts containing melanocytes gave disappointing clinical results, with pigmentation achieved in only one out of five patients. Irrespective of the fate of melanocytes grafted back onto the patients, we experienced problems in identifying melanocytes within these well‐integrated keratinocyte sheets. This led us to explore the fate of these cells within these sheets in vitro and to seek to improve their number and function within the sheets. We report that the introduction of a fibroblast feeder layer can improve melanocyte number within melanocyte/keratinocyte co‐cultures initially, but at very high keratinocyte density, there is a marked loss of melanocytes (as detected by staining for S100). Additionally, we found that keratinocytes not only down‐regulate melanocyte number, but also pigmentary function; thus, it was possible to identify melanocytes that were S100 positive but tyrosinase‐related protein‐1 (TRP‐1) negative in confluent well‐integrated keratinocyte sheets. In summary, our data suggest that keratinocytes at high density initially suppress melanocyte pigmentation (as evidenced by a lack of TRP‐1 expression) and then cause a physical loss of melanocytes. The introduction of a fibroblast feeder layer can help maintain melanocyte number while keratinocytes are subconfluent, but fails to oppose the inhibitory influence of the keratinocytes on melanocyte TRP‐1 expression.  相似文献

To learn more of the role of calcium in the regulation of melanogenesis, we have used direct manipulation of medium calcium and pharmacological modulation of intracellular calcium to examine the consequences on unstimulated and cyclic AMP elevated tyrosinase activity and melanin synthesis and distribution in B16 melanoma cells. In unstimulated cells, calcium is clearly inhibitory to tyrosinase activity. However, in cells stimulated with cAMP-elevating agents the requirement for extracellular calcium was changed such that cells required a minimum of 0.4–0.6 mmol medium calcium for maximum tyrosinase response to these agents. Paradoxically, pharmacologically increasing intracellular calcium in cAMP-stimulated cells with ionophore inhibited tyrosinase activity, and the calcium-lowering agent TMB8 and the calcium channel blocker verapamil both stimulated tyrosinase activity. When melanin synthesis was measured in cAMP-stimulated cells, TMB8 was found to significantly increase the sensitivity and the maximum melanogenic response to α-MSH, suggesting the presence of at least one level of endogenous calcium inhibitory control operative in these cells. In addition, TMB8 changed the distribution of melanin between the cell and the medium such that, in the presence of α-MSH and TMB8, significantly more melanin was secreted into the medium. These data suggest that calcium is required for several steps in melanogenesis, having an apparently inhibitory effect on pre-tyrosinase activity in unstimulated cells, but also showing evidence of a positive role in cyclic AMP-stimulated tyrosinase activity, as well as a further possible inhibitory role in melanin movement or secretion.  相似文献

Subspecies lie at the interface between systematics and population genetics, and represent a unit of biological organization in zoology that is widely used in the disciplines of taxonomy and conservation biology. In this review, we explore the utility of subspecies in relation to their application in systematics and biodiversity conservation, and briefly summarize species concepts and criteria for their diagnosis, particularly from an invertebrate perspective. The subspecies concept was originally conceived as a formal means of documenting geographical variation within species based on morphological characters; however, the utility of subspecies is hampered by inconsistencies by which they are defined conceptually, a lack of objective criteria or properties that serve to delimit their boundaries, and their frequent failure to reflect distinct evolutionary units according to population genetic structure. Moreover, the concept has been applied to populations largely comprising different components of genetic diversity reflecting contrasting evolutionary processes. We recommend that, under the general lineage (unified) species concept, the definition of subspecies be restricted to extant animal groups that comprise evolving populations representing partially isolated lineages of a species that are allopatric, phenotypically distinct, and have at least one fixed diagnosable character state, and that these character differences are (or are assumed to be) correlated with evolutionary independence according to population genetic structure. Phenotypic character types include colour pattern, morphology, and behaviour or ecology. Under these criteria, allopatric subspecies are a type of evolutionarily significant unit within species in that they show both neutral divergence through the effects of genetic drift and adaptive divergence under natural selection, and provide an historical context for identifying biodiversity units for conservation. Conservation of the adaptedness and adaptability of gene pools, however, may require additional approaches. Use of the Pressure Chamber in Membrane Damage Studies   总被引:2,自引:0,他引:2  
The ability to detect membrane damage in plant leaves by thepressure chamber technique was evaluated. Membrane damage wasinduced by freezing and thawing, absorption of the host-specifictoxin victorin, ozonation, inoculation with Helminthosporiummaydis, and spraying with the phytotoxin, fusicoccin. The pressure-volumerelationships, i.e. the volume of sap expressed from a leafwith incremental increases in pressure, were compared in leaveswith intact or damaged membranes. Where membrane damage waswidespread throughout the leaf, sap was expressed at pressuresas low as one tenth of those needed in leaves with intact membranesand at low pressures the amount of water expressed from leaveswith damaged membranes was up to 10 times that from leaves withintact membranes. Further, the pressure-volume curves of healthyleaves became linear when the leaf turgor potential was reducedto zero, but were non-linear in leaves with damaged membranes.Ozone treatment, inoculation with H. maydis, or spraying withfusicoccin damaged only a proportion of the leaf cells, andpressure-volume relationships more nearly resembled those obtainedwith healthy leaves rather than those obtained with freezingand thawing and absorption of victorin. It is concluded that the pressure chamber can be used to observemembrane integrity, but its ability to detect damage may belimited to conditions in which the majority of cells in theleaf are damaged.  相似文献
