Previous work from our laboratory has shown that both cyclic AMP and calcium/calmodulin appear to be involved in the regulation of melanogenesis in murine B16 melanoma cells. In these cells as in murine Cloudman S91 cells, melanogenic responsiveness to melanocyte-stimulating hormone (MSH) varies with cell density in culture. Our objective in this study was to learn more about the intracellular systems involved in the control of melanogenesis, particularly the role played by calcium. The melanogenic response to αMSH was compared to the response to drugs affecting intracellular free calcium and calmodulin over a range of cell densities in B16F1 cells. αMSH-stimulated melanin production was extremely density-dependent but αMSH-stimulated cyclic AMP production was independent of cell density. The melanogenic response to agents that increased intracellular calcium (A23187) or inhibited intracellular calmodulin varied with cell density. A drug (TMB8) that lowered intracellular free calcium, however, increased melanogenesis independently of cell density. At high cell density it was found that an elevation in calcium decreased melanogenesis, whereas agents that reduced calcium or inhibited calmodulin activity increased melanogenesis. At low cell density, however, the inhibitory response to A23187 was lost and in some experiments even stimulated melanogenesis. These data suggest that the calcium/calmodulin signalling system has an inhibitory influence on melanogenesis, and its expression, which depends upon cell density, may also modulate the response to stimulatory agents such as αMSH.  相似文献

We compiled a large database of 58 059 point locality records for 70 species and 434 subspecies of heliconiine butterflies and used these data to test evolutionary hypotheses for their diversification. To study geographical patterns of diversity and contact zones, we mapped: (1) species richness; (2) mean molecular phylogenetic terminal branch length; (3) subspecies richness and the proportion of specimens that were subspecific hybrids, and (4) museum sampling effort. Heliconiine species richness is high throughout the Amazon region and peaks near the equator in the foothills and middle elevations of the eastern Andes. Mean phylogenetic terminal branch length is lowest in the eastern Andes and tends to be low in species‐rich areas. By contrast, areas of high subspecies richness, where subspecies overlap in range and/or hybridize, are concentrated along the course of the Amazon River, with the eastern Andes slopes and foothills relatively depauperate in terms of local intraspecific phenotypic diversity. Spatial gradients in heliconiine species richness in the Neotropics are consistent with the hypothesis that species richness gradients are driven at least in part by variation in speciation and/or extinction rates, resulting in observed gradients in mean phylogenetic branch length, rather than via evolutionary age or niche conservatism alone. The data obtained in the present study, coupled with individual case studies of recently evolved Heliconius species, suggest that the radiation of heliconiine butterflies occurred predominantly on the eastern slopes of the Andes in Colombia, Ecuador, and Peru, as well as in the upper/middle Amazon basin. © 2012 The Linnean Society of London, Biological Journal of the Linnean Society, 2012, 105 , 479–497.  相似文献

ABSTRACT We designed a novel approach to determining extent of distribution and area of occupancy for wolverines (Gulo gulo) by using aerial surveys of tracks in snow and hierarchical spatial modeling. In 2005 we used a small, fixed-wing aircraft with pilot and one observer to search 575 of 588 survey units for wolverine tracks in approximately 60,000 km2 of boreal forest in northwestern Ontario, Canada. We used sinuous flight paths to scan open areas in the forest in the 100-km2 survey units. We detected tracks in 138 (24%) of the 575 sampled units. There was strong evidence of occurrence (probability of occurrence >0.80) in 30% of the 588 survey units, weak evidence of occurrence (0.50–0.80) in 12%, weak evidence of absence (0.20–0.50) in 15%, and strong evidence of absence (< 0.20) in 43%. Wolverine range comprised 59% of the study area and area of occupancy was 33,400 km2. With information on probability of occurrence and core areas of occupation for wolverines in our study area, resource managers and others can examine factors that influence wolverine distribution patterns and use this information to formulate best management practices that will maintain wolverines on the landscape in the face of increasing resource development. Comparing future survey results with those of our 2005 survey will provide an objective way to assess the efficacy of management practices.  相似文献

1. To study the bottom‐up linkages in arctic lakes, we treated one side of a partitioned lake with inorganic nitrogen and phosphorus for a 6‐week period each summer for 6 years starting in the summer of 1985. We took a variety of weekly measurements to determine the impact of the nutrient loading on the lake and continued weekly measurements for 2–6 years after the cessation of nutrient loading to observe the recovery of the treated side. The loading rates (2.91 mmol N m?2 day?1 and 0.23 mmol P m?2 day?1) were five times the calculated loading rates for Toolik Lake, located nearby. 2. In all 6 years of nutrient addition, phytoplankton biomass and productivity were greater in the treated sector than the reference sector. In the first 4 years of nutrient addition there was no flux of phosphorus from the mineral‐rich sediments. This changed in the last 2 years of nutrient addition as phosphorus was released to the lake. 3. The response of the animal community to increased plant production was mixed. One of the four macro‐zooplankton species (Daphnia longiremis) increased in number by about twofold in the first 5 years. However, the copepod Cyclops scutifer showed no response during the treatment phase of the study. The benthic invertebrate response was also mixed. After a 2‐year lag time the snail Lymnaea elodes increased in the treated lake sector but chironomids did not. 4. Ecosystem response to fertilisation was not controlled solely by nutrient addition because phosphorus was not recycled from the sediments until the last 2 years of nutrient addition. Phytoplankton still showed the effects of nutrient addition in the recovery period and the hypolimnion of the treated sector was still anaerobic starting at 6 m in 1996.  相似文献

Exceptionally well-preserved remains of phosphatized siphuncles were discovered in four specimens of a Permian prolecanitid ammonoid Akmilleria electraensis (Plummer and Scott) from Buck Mountain, Nevada. These structures occur as truncated segments within the siphuncular tube. The outer surface of the siphuncle is sculptured by numerous equally spaced longitudinal ridges and furrows; the ridges represent an infolded basement membrane of epithelial cells which corresponds to the distal ends of individual canaliculi between epithelial cells. In cross-section, the siphuncle of A. electraensis consists of a large central vein, possibly two pairs of arteries, porous connective tissue with reticulate hemocoelic spaces, and a thin epithelium. In the presence of two pairs of arteries and porous connective tissue, the siphuncle of A. electraensis is more like that of Nautilus pompilius than that of Spirula spirula, which has nine arteries and dense connective tissue. However, Nautilus possesses relatively smaller and more numerous epithelial cells around the siphuncle than does Akmilleria. These observations strongly suggest that the siphuncular epithelium of Akmilleria served as the salt-­concentrating organ for buoyancy regulation of the living animal, just as in Nautilus and Spirula.  相似文献

In a 3 year study of the cattle egret at a heronry of 3500 nests in south-east Queensland, 34% of eggs in 711 nests observed failed to survive to become fledglings (11 day old chicks). Fledgling mortality seemed much lower. The main sources of mortality were crow predation on eggs, failure of the incubated egg to develop fully, starvation of nestlings and tick infestation of nestlings. These were of about equal importance overall but their individual magnitudes varied considerably among the three seasons. The pattern of infestation of the cattle egret chick with the tick Argas (Persicargas) robertsi and the effect of this on chick health are described. An account is given of the other sources of morbidity also. The heronry appeared to be producing a net surplus of young despite these losses.  相似文献

Abstract .Unilaterally antennectomized Heliothis virescens (F.) males flying close to the central axis of a plume of sex pheromone display no significant differences in behaviour compared to sham-operated males in course angles, track angles, airspeed and groundspeed. This demonstrates that right/left antennal information is not necessary for normal orientation movements in response to pheromone, but rather that it is 'blended' within the moth's central nervous system before pheromone-mediated manoeuvres are made. However, some unilaterally antennectomized moths (36%) make repetitive, asymmetrical, saw-tooth-shaped tracks during pheromone-mediated upwind progress, whereas control moths never make such tracks. Unilaterally antennectomized moths made such tracks on the side of the plume contralateral to the missing antenna. We hypothesize that these occasional asymmetrical tracks in unilaterally ablated males are the result of reiterative asymmetrical pheromone stimulation of a higher probability on track legs going toward rather than away from the long axis of the plume on males with a single antenna remaining on the 'away from axis' side. Combined with a greater propensity for treated moths to lock onto the plume away from the central axis on one side rather than the other, repetitive successive asymmetrical track legs (resulting in a saw-tooth-shaped track) are commonly observed in these moths. Control moths do also make asymmetric successive track legs but they rarely are repeated and thus are not readily observed.  相似文献

The Cambrian bradoriid ? Parahoulongdongella sp. is shown to have a microreticulate (2nd order) surface sculpture. A possible organic sheet‐like origin is proposed for this reticulation.
