Phenylalanyl-tRNA synthetase and phenylalanine ammonia-lyase activities were demonstrated in partially purified extracts of pine (Pinus elliottii) suspension cultures. The optimum pH for the phenylalanyl-tRNA synthetase reaction was 7.5 and the optimum ATP and Mg2+ concentrations were 1.0 and 15 mM respectively. Pine, calf liver and yeast tRNA were inadequate substitutes for pea tRNA in the synthetase reaction mixtures. The optimum pH for the phenylalanine ammonia-lyase reaction was 9.0. The Km for phenylalanine was approximately 6.6 × 10?5M. The activity of both enzymes in the partially purified extracts was unstable on storage.  相似文献

The type and two other specimens of Ephalophis greyi are described. The venom gland musculature is of the Demansia type, with an isolated muscle running from the quadrate to the rear of the venom gland. The only other hydrophiine with the Demansia pattern of venom gland musculature, Hydrophis mertoni Roux, is referred, for this and other reasons, to the genus Ephalophis as E. mertoni (Roux). Some notes on a skull of E. mertoni are presented, along with a revised diagnosis of Ephalophis. The Demansia pattern of venom gland muscles seen in Ephalophis is primitive for the Hydrophiinae and other members of that subfamily have secondarily taken on a Glyphodon type of musculature of the venom gland by loss of the muscle slip from quadrate to gland. The Demansia group of terrestrial elapids, particularly the genus Drepanodontis Worrell, stand close to the origin of the Hydrophiinae. A classification of the Hydrophiinae into three generic groups is presented, with Ephalophis, Aipysurus , and Emydocephalus referred to an Aipysurus group.  相似文献

SYNOPSIS. Isospora petrochelidon sp. n. (Protozoa: Eimeriidae) is described from the cliff swallow, Petrochelidon pyrrhonota , from Douglas County, Colorado. Oocysts were passed primarily between 4 and 9 p.m. Sporogony was completed by 24 hr in 3% aqueous potassium dichromate at 21–28 C. The ellipsoid to ovoid oocysts averaged 25.2 × 22.2 μm. The oocyst wall was smooth, colorless, and single-layered. The 2 polar granules were disc-shaped, each adjoining a sporocyst. The lemon-shaped sporocysts averaged 18.4 × 10.8 μm. There was no membrane enclosing the sporocyst residuum and sporozoites. Two fledgling Isospora -free swallows fed sporulated oocysts began passing them on day 4 and died with massive infections on day 5. Data from these birds indicate that schizogony occurred during nighttime hours, gametogony during the day. Endogeny was found throughout the intestine distal to the gizzard, with schizogony confined to the convoluted glands of the subvillar mucosa and gametogony to the villar epithelium. Sporocysts released from mechanically ruptured oocysts excysted in 2% pancreatin within 5 min. Sporulated oocysts fed to a pigeon squab, duckling and cockerel produced no infections.  相似文献

All amphibians are thought to possess two distinct types of dermal gland: mucous and granular. Mucous glands typically produce an acidic, flocculent and carbohydrate positive secretion that keeps skin moist and helps maintain homeostasis. In this study, we use a battery of histochemical tests to identify these traditional mucous glands in the dermis of the plethodontid salamander Ensatina eschscholtzii . The periodic acid-Schiff, alcian blue, toluidine blue and carmine reactions reveal the absence of both acidic glycosaminoglycans and traditional mucous glands. Instead, E. eschscholtzii produces neutral mucus in specialized granular glands, which we name mucus-producing granular glands (MPGGs). 1. We evaluated the population genetic structure of the common New Zealand amphipod Paracalliope fluviatilis using eight allozyme loci, and the mitochondrial cytochrome oxidase c subunit I (COI) gene locus. Morphological analyses were also conducted to evaluate any phenotypic differences. Individuals belonging to P. fluviatilis were collected from a total of 14 freshwater fluvial habitats on the North and South Islands, New Zealand. 2. We found evidence for strong genetic differentiation among locations (Wright's FST > 0.25), and fixed differences (non‐shared alleles) at two of the eight allozyme loci indicating the possibility of previously unknown species. Analysis of a 545‐bp fragment of the COI locus was mostly congruent with the allozyme data and revealed the same deeply divergent lineages (sequence divergences up to 26%). 3. Clear genetic breaks were identified between North Island and South Island populations. North Island populations separated by <100 km also showed genetic differences between east and west draining watersheds (sequence divergence >12%). Accordingly, present‐day dispersal among hydrologically isolated habitats appears minimal for this taxon. 4. Although population differences were clearly shown by allozyme and mtDNA analyses, individuals were morphologically indistinguishable. This suggests that, as in North American and European taxa (e.g. Hyalella and Gammarus), morphological conservatism may be prevalent among New Zealand's freshwater amphipods. We conclude that molecular techniques, particularly the COI gene locus, may be powerful tools for resolving species that show no distinctive morphological differences.  相似文献

Recent research in whole-plant stomatal physiology, conducted largely with potted plants in controlled environments, suggests that stomatal conductance ( g s) might be more closely linked to plant chemical variables than to hydraulic variables. To test this in a field situation, seasonal g s was examined in relation to a number of plant and environmental variables in 11 temperate, deciduous forest tree species. Stomatal conductance was generally better correlated with environmental variables (air temperature, vapor pressure deficit, PPFD) than with plant variables, and slightly better correlated with plant hydraulic variables (shoot water and osmotic potentials) than with plant chemical variables (xylem sap ABA concentration, xylem sap pH). We examined a model, developed previously for maize, which describes regulation of g s by xylem sap ABA concentration with leaf water status acting to modify stomatal sensitivity to the ABA signal. This model explained slightly more variation in seasonal g s in the forest trees than did single plant variables but not more variation than most single environmental variables. Response surface models, especially those incorporating environmental variables, were more consistently successful at explaining g s across species.  相似文献

There are numerous hypotheses to explain the evolution of sexual dimorphism in spiders. One of the most controversial is the differential mortality model (DMM) which proposes that differing rates of (adult) male and female mortality can result in a skewed operational sex ratio and lead to the evolution of small males. This hypothesis has been examined using a comparative approach which assumes that the behaviour of males and females could be used as a surrogate measure of mortality. We tested this assumption using two model species, Hogna helluo and Pardosa milvina (Araneae: Lycosidae) that differ in the degree of sexual dimorphism both in terms of body size and level of activity. Our data demonstrate that differences in male and female behaviour are not predictive of differences in mortality. Rather, as in other organisms, mortality is a complex phenomenon dependent on activity as well as size. These data call into question the methods previously used to test the DMM and suggest that understanding sexual size dimorphism (SSD) in spiders will require evaluation of historical constraints as well as how size currently influences fitness in each sex.  © 2003 The Linnean Society of London .
