本文通过Aβ25-35诱导体外原代培养的SD乳大鼠海马神经元,建立Aβ毒性损伤细胞模型,结合AnnexinV-FITC/PI荧光双染法流式细胞术、MTT比色法、实时荧光定量PCR及Western blot方法检测川芎嗪(tetrameth-ylpyrazine,TMP)对原代培养的海马神经元细胞活性、早期凋亡率和Bax、Bcl-2基因表达的影响。结果显示川芎嗪高、中剂量可明显增强细胞活性,增加神经元细胞的存活率(P<0.01),可显著抑制海马神经元细胞早期凋亡(P<0.01),抑制凋亡蛋白Bax的表达(P<0.01),增强抗凋亡蛋白bcl-2的表达(P<0.01)。川芎嗪可通过调节Bax/Bcl-2平衡抵抗Aβ25-35诱导的海马神经元凋亡,降低Aβ的神经元毒性,对海马神经元损伤有明显的保护作用。 相似文献

A wealth of molecular interaction data is available in the literature, ranging from large-scale datasets to a single interaction confirmed by several different techniques. These data are all too often reported either as free text or in tables of variable format, and are often missing key pieces of information essential for a full understanding of the experiment. Here we propose MIMIx, the minimum information required for reporting a molecular interaction experiment. Adherence to these reporting guidelines will result in publications of increased clarity and usefulness to the scientific community and will support the rapid, systematic capture of molecular interaction data in public databases, thereby improving access to valuable interaction data. 相似文献

Cement factory emissions into air cause serious air pollution and affect the plant and animal life in the environment. Herein, we report the effects of cement industry emissions (O3, SO2 and NO2) in air, as pollutants, at Riyadh City on Datura innoxia Mill. plant. Morphological characters including plant height, leaves area and number, fresh and dry weight of shoot and root systems of D. innoxia showed a significant reduction from their normal control plants as a response to exposure to pollutant emissions. Chlorophyll and carotenoid contents recorded reductions in values compared to control plant, and the lowest values of chlorophyll A, B, total chlorophyll, carotenoids and total pigments were 0.431, 0.169, 0.60, 0.343 and 0.943 mg/g respectively at a distance of 1–5 m from the cement factory in fruiting stage. These changes in values may be attributed to a probable deceleration of the biosynthetic process rather than degradation of pigments. Further D. innoxia showed a significant (P < 0.01) reduction in non-reducing and total sugars, protein and total lipid contents compared with the control plant. The root system recorded the lowest values of reducing sugars (0.350 mg/g f. wt.), non-reducing sugars (0.116 mg/g f. wt.), total sugars (0.466 mg/g f. wt.), protein content (0.931 mg/g f. wt.) and total lipids content (0.669 mg/g f. wt.) in fruiting stage at a distance of 1–5 m from the cement factory. The peroxidase activity of shoot and root systems of the studied plant was also significantly higher than those of control plant. Thus a highest value of (29.616 units/g f. wt.) peroxidase activity was recorded in vegetative stage of shoot system at a distance 1–5 m from the cement factory. Results of the study indicated that cement industry emission strongly influence the physiology and morphology of date palm D. innoxia which contribute date fruits, a staple food in the Arab world. 相似文献

The prion protein is central to the disease pathogenesis of a variety of neurodegenerative diseases such as CJD. The protein is only able to initiate the disease process following post-translational modification. The main characteristic of this change is the ability of this altered isoform to polymerise. We wish to determine if altered cleavage of the protein could generate a protein fragment able to initiate polymerisation. During normal metabolic breakdown the protein is initially cleaved at a single site at around amino acid residue 111/112 in the mouse sequence. A second site before amino acid residue 90 has been postulated as an alternative cleavage point. We have provided evidence that hydrogen peroxide as low as 50 microM in the presence of copper, iron or manganese (but not nickel, magnesium or zinc) can cleave the recombinant protein near this site and requires a GXXH motif in the protein sequence. This reaction results in the production of 6 and 19 kDa fragments of the protein. This cleavage pattern occurs in prion proteins from different species (mouse, chicken and turtle) and is enhanced by modification of the octameric repeat region. The 19 kDa fragment produced by this reaction is protease sensitive. This fragment in a pure form caused the polymerisation of wild-type prion protein by a seeding mechanism. Therefore our results provide a possible mechanism by which altered cleavage of the prion protein could result in the kind of protein polymerisation associated with prion diseases. 相似文献

水淹状况是湿地植被动态的重要影响因素。该研究基于谷歌地球引擎(GEE)平台, 利用2000-03-01至2020-02-29所有覆盖研究区域的MODIS遥感影像数据, 分析20年间水淹频率(IF)、增强型植被指数(EVI)的时空变化以及湿地植被对IF变化的响应, 得出以下结论: (1) 20年来鄱阳湖水文节律发生了明显改变, 高IF (IF > 75%)水域面积呈现下降趋势, 从2000年1 435.3 km2下降至2019年的510.25 km2, 降幅为64.45%; (2)区域平均EVI呈显著上升趋势, 植被扩张主要集中在中部IF下降区域; (3)分析不同总水淹频率区域中平均EVI年际变化, 发现EVI与水淹状况的变化趋势相似, 2009年之后鄱阳湖水域面积萎缩趋势缓解, EVI增长速度出现下降; (4)鄱阳湖湿地植被主要沿水域面积萎缩方向扩张, 基于像元统计20年间IF与EVI的变化趋势, 发现它们在空间分布上高度吻合, 这种空间异质性进一步证实水淹状况起到调节植被动态变化的作用。 相似文献

Background
Identifying the location and the volume of the prostate is important for ultrasound-guided prostate brachytherapy. Prostate volume is also important for prostate cancer diagnosis. Manual outlining of the prostate border is able to determine the prostate volume accurately, however, it is time consuming and tedious. Therefore, a number of investigations have been devoted to designing algorithms that are suitable for segmenting the prostate boundary in ultrasound images. The most popular method is the deformable model (snakes), a method that involves designing an energy function and then optimizing this function. The snakes algorithm usually requires either an initial contour or some points on the prostate boundary to be estimated close enough to the original boundary which is considered a drawback to this powerful method. 相似文献78.

Candidatus Magnetoglobus multicellularis (CMm) is a multicellular organism in which each constituent cell is a magnetotactic bacterium.
It has been observed that disaggregation of this organism provokes the death of the individual cells. The observed flagellar
movement of the CMm indicates that the constituent cells move in a coordinated way, indicating a strong correlation between
them and showing that this aggregate could be considered as an individual. As every constituent cell is a magnetotactic bacterium,
every cell contributes with a magnetic moment vector to the resultant magnetic moment of the CMm organism that can be calculated
through the vectorial sum of all the constituent magnetic moments. Scanning electron microscopy images of CMm organisms have
shown that the constituent cells are distributed on a helix convoluted on a spherical surface. To analyze the magnetic properties
of the distribution of magnetic moments on this curve, we calculated the magnetic energy numerically as well as the vectorial
sum of the magnetic moment distribution as a function of the number of cells, the sphere radius and the number of spiral loops.
This distribution proposes a magnetic organization not seen in any other living organism and shows that minimum energy configurations
of magnetic moments are in spherical meridian chains, perpendicular to the helix turns. We observed that CMm has a high theoretical
degree of magnetic optimization, showing that its geometrical structure is important to the magnetic response. Our results
