Calcium ion stimulated endogenous protein kinase catalyzed phosphorylation of basic proteins in myelin subfractions and myelin-like membrane fraction from rat brain

Polypeptide composition and endogenous phosphorylation were investigated in the subfractions of rat brain myelin isolated by either discontinuous or continuous sucrose density gradient centrifugation of myelin. Similarly, a myelin-like membrane fraction (SN4) was also studied. Observations were made that strongly indicated the presence of a calcium-stimulated protein kinase in a highly purified myelin preparation and which exclusively phosphorylated myelin basic proteins of the membrane preparation. Adenosine cyclic 3',5'-phosphate (cAMP) stimulated kinase on the other hand was found to be considerably enriched in the myelin-like membrane fraction. Although this latter enzyme is also capable of phosphorylating the basic proteins, its effect was at least 5 times weaker compared to the calcium-stimulated myelin protein kinase. Within the gradient subfractions there appeared a close relation between the amount of basic proteins and their calcium-stimulated phosphorylation; a similar relationship, however, was not obtained in the case of cAMP-dependent phosphorylation of myelin basic proteins. The former (i.e., Ca2+-stimulated phosphorylation) was found to require a protein factor that functionally resembled calmodulin. The results thus raises an interesting possibility of the presence of calmodulin-like proteins and a calcium-stimulated protein kinase in adult myelin membrane from mammalian brain, both of which have been hitherto unrecognized constituents of myelin membranes.     

Epidemiological characteristics and the importance of carriers of the surface antigen of the hepatitis B virus (HBsAg)

The occurrence of HBsAg carriership in Leningrad has been found to be 1.4% according to the results of countercurrent immunoelectroosmophoresis (CIEO) and 2.1% according to the results of the passive hemagglutination (PHA) test. In children HBsAg occurs with higher frequency: 1.9% according to the results of CIEO and 3.4% according to the results of the PHA test. The latter test reveals HBsAg carriers more completely, especially in women who have usually less pronounced antigenemia than men. Most of chronic HBsAg carriers are patients with chronic forms of hepatitis B (chronic active hepatitis and chronic persistent hepatitis); frequently they become the source of infection among their relatives under the conditions of family contacts. A complex of antiepidemic measures is necessary in the foci of chronic HBsAg carriership.     

The Role of the Vascular Factor in the Reorganization of Water Metabolism in Denervated Liver after Bacterial Endotoxin Poisoning

Intoxication with bacterial lipopolysaccharide (endotoxin) is accompanied by considerable rearrangements in the systems of blood microcirculation and water metabolism of the liver. These rearrangements are manifested as increased sinusoid area, changed total area of the cytoplasm and nuclei as well as the nucleocytoplasmic ratio in hepatocytes, increased content of total water in the organ, and changed magnetic relaxation properties (spin-lattice and spin-spin relaxation times). Preliminary parasympathetic denervation of the liver (vagotomy) changes the pattern of the organ response to bacterial endotoxin poisoning as indicated by the kinetics of studied morphological and biophysical parameters.     

Technical note: an improved method for differential staining of Bacillus thuringiensis crystals

A new staining method is described using naphthalene black 12B and Gurr's improved R66 Giemsa for staining all known crystal types produced by Bacillus thuringiensis.     

Identification of Phytopathogenic Virus Strains by their Luminescent Properties

A quick test has been developed to identify phytopathogenic virus strains by variations in their photoluminescence parameters. When heated, the virus suspension shows a jumpwise drop or rise in fluorescence intensity caused by conformational changes in the macromolecules of the virion protein capsules. The relative value of the jump in fluorescence intensity and the temperature value at which the jump occurs are essentially strain-specific. The minimum time needed for one test is from 15 to 20 minutes.     

Polysaccharide derivatives as coats for nylon tube urease

Urease was immobilized on O-alkylated nylon tubes coated with polyaminated derivatives of starch or dextran. The specific activity of the enzyme coil and the relative stability of the immobilized enzyme, compared with immobilized urease derived from other nylon tube modifications, were enhanced. Also, the nonspecific binding of urease to O-alkylated nylon tubes was virtually eliminated by the coating process.     

Effects of taxol on endothelial cells of the developing semilunar heart valves in the chick embryo

Recent ultrastructural studies have revealed that there are differences in endothelial cell shape and cytoskeletal architecture between the arterial and ventricular faces of developing semilunar valves. In the present work we have analyzed valvular endothelial cell response to taxol in the chick embryo. Following taxol administration, abnormal mitotic cells appear in the endothelium of both faces of the cusps. Interphase cells show few structural alterations. No differential response of the valvular endothelium to the drug was observed in the arterial or ventricular faces of the cusps.
